The interleukin-5 messenger RNA expression in a patient with idiopathic hypereosinophilic syndrome.

Interleukin-5 has a specific role in various eosinophilic activities. It is the predominant cytokine produces by activated T-lymphocytes isolated from patients with idiopathic hypereosinophilic syndrome. We studied a young patient suffering from idiopathic hypereosinophilic syndrome who presented with Horner's syndrome, peripheral neuropathy and skin ulcers. The IL-5 gene expression by CD4+ T-lymphocytes and the peripheral eosinophil count were raised. The skin ulcers continued to deteriorate despite a swift reduction of the IL-5 gene expression and peripheral eosinophil count following system

Studies on schistosomiasis in western Kenya: I. Evidence for immune-facilitated excretion of schistosome eggs from patients with Schistosoma mansoni and human immunodeficiency virus coinfections.

Persons employed as vehicle washers in the town of Kisumu, Kenya are exposed for several hours each day to water in Lake Victoria that contains Schistosoma mansoni-infected Biomphalaria pherifferi snails. This results in a focus of high endemicity for schistosomiasis and these persons have very high concentrations of eggs in their feces (mean +/- SD = 1,469 +/- 1,581 eggs per gram [EPG] of feces). Fecal egg counts, but not circulating cathodic antigen (CCA) levels, in these schistosomiasis patients differed strikingly based on the patient's seropositivity for human immunodeficiency virus (HIV)

MT-2 cell tropism as prognostic marker for disease progression in human immunodeficiency virus type 1 infection.

The ability of human immunodeficiency virus type 1 (HIV-1) isolates to replicate in MT-2 cells was investigated as a prognostic marker for disease progression and CD4+ lymphocyte depletion in 53 HIV-1-infected, asymptomatic individuals. MT-2-negative viruses were isolated from 49% of the patients both early and late during the follow-up period; 38% converted from being MT-2 negative to MT-2 positive, while 11% were MT-2 positive throughout the study. One individual showed a fluctuating virus phenotype. The loss of CD4+ lymphocytes was significantly more rapid in MT-2-positive patients. We foun

Evidence for a role of virulent human immunodeficiency virus (HIV) variants in the pathogenesis of acquired immunodeficiency syndrome: studies on sequential HIV isolates.

Sequential human immunodeficiency virus (HIV) isolates, recovered from a panel of longitudinally collected peripheral blood mononuclear cells obtained from 20 initially asymptomatic HIV-seropositive homosexual men, were studied for differences in replication rate, syncytium-inducing capacity, and host range. Eleven individuals remained asymptomatic; nine progressed to acquired immunodeficiency syndrome (AIDS) or AIDS-related complex (ARC) at the time point at which the last HIV isolate was obtained. In 16 individuals, only non-syncytium-inducing (NSI) isolates, with a host range restricted to

LAMP2 regulates autophagy in the thymic epithelium and thymic stroma-dependent CD4 T cell development.

Within the thymus, thymic epithelial cells (TECs) provide dedicated thymic stroma microenvironments for T cell development. Because TEC functionality is sensitive to aging and cytoablative therapies, unraveling the molecular elements that coordinate their thymopoietic role has fundamental and clinical implications. Particularly, the selection of CD4 T cells depends on interactions between TCRs expressed on T cell precursors and self-peptides:MHC II complexes presented by cortical TECs (cTECs). Although the macroautophagy/autophagy-lysosomal protein degradation pathway is implicated in CD4 T ce

Nickel Challenge In Vitro Affects CD38 and HLA-DR Expression in T Cell Subpopulations from the Blood of Patients with Nickel Allergy.

Nickel allergy is a major health problem and shows clinical manifestation of contact eczema. The response of specific lymphocyte subpopulations in sensitized patients after new challenge to nickel has until now not been studied in detail. To evaluate if nickel-based elicitation reaction could be objectively identified by multi-parametric flow cytometry, immunophenotyping of specific T cells was applied. White blood cells from 7 patients (4 positive in patch test, 3 negative) were challenged by nickel and in vitro short-term culture. Standardized antibody-dye combinations, specific for T helper

Effects of pyridaben on the spleen and T lymphocytes and intervention strategy in grass carp (Ctenopharyngodon idella).

The widespread use of pyridaben (PDB) poses serious threat to aquatic organism health. 6-Gingerol, a bioactive component of ginger, has been demonstrated to possess both antioxidant and immunomodulatory properties. However, the effects of spleen injury of PDB in aquatic species and the potential mitigating effects of 6-Gingerol remain insufficiently characterized. The present study aimed to characterize the toxic effects of PDB on the spleen of grass carp and to evaluate whether 6-Gingerol could effectively counteract PDB-induced toxicity. The results showed that PDB exposure could induce sple

Hyper-immunoglobulin M syndrome caused by a mutation in the promotor for CD40L.

Hyper-immunoglobulin M (IgM) syndrome (HIGM) is a rare primary immunodeficiency characterized by elevated or normal IgM and absent or markedly decreased amounts of IgG, IgA and IgE. The X-linked form (HIGM1) is the most common type and is caused by mutations in the gene for CD40L, a T-cell surface molecule required for T-cell driven immunoglobulin class switching by B cells. In the present study we have identified a patient with X-linked hyper-IgM who failed to express CD40L on the cell surface of CD4(+) T lymphocytes. Sequence analysis of CD40L genomic DNA showed no mutations. CD40L mRNA was

Whole-protein screening and multi-modal profiling of antigen-specific CD4+ T cells at single-cell resolution.

Systematic whole-protein screening and comprehensive profiling of antigen-specific CD4+ T cells are crucial for advancing vaccine design and cancer immunotherapies, yet remain technically challenging. Here, we present a high-throughput platform that utilizes large-scale class II single-chain trimer libraries to detect antigen-specific CD4+ T cells, while simultaneously profiling their antigen specificity, TCRα/β sequences, MHC restriction, whole transcriptomes, and patient/timepoint origins at single-cell resolution. Upon rigorous platform validation, we screened the full SARS-CoV-2 spike rece

Identification of Novel Yellow Fever Class II Epitopes in YF-17D Vaccinees.

Yellow fever virus (YFV) is a mosquito-borne member of the genus flavivirus, including other important human-pathogenic viruses, such as dengue, Japanese encephalitis, and Zika. Herein, we report identifying 129 YFV Class II epitopes in donors vaccinated with the live attenuated YFV vaccine (YFV-17D). A total of 1156 peptides predicted to bind 17 different common HLA-DRB1 allelic variants were tested using IFNγ ELISPOT assays in vitro re-stimulated peripheral blood mononuclear cells from twenty-six vaccinees. Overall, we detected responses against 215 YFV epitopes. We found that the capsid and

Molecular Regulation of HIV-1 Expression and Persistence Across Diverse Cellular Reservoirs.

Despite the remarkable success of antiretroviral therapy (ART) in suppressing human immunodeficiency virus type 1 (HIV-1) replication, viral persistence remains a major barrier to cure. This persistence is sustained by heterogeneous cellular reservoirs in which viral expression is tightly regulated by host-dependent molecular mechanisms. Beyond the canonical cluster of differentiation 4 (CD4+) T-cell reservoirs, HIV-1 establishes long-lived infection in myeloid cells, glial populations within the central nervous system (CNS), and additional non-canonical cellular niches, each characterized by

Immunological evaluation of vaccines to prevent Chlamydia trachomatis infection using the human in vitro MIMIC system.

The Modular Immune In vitro Construct (MIMIC) system is an established laboratory-based methodology that reproduces in vitro the human in vivo T-cell immune response. In this study, we utilized the MIMIC system to establish a dendritic cell and CD4+ T-cell co-culture model that recapitulates T helper cell responses to Chlamydia trachomatis infection in humans. We demonstrate that C. trachomatis infection induces C. trachomatis-specific IFN-γ-producing CD4+ T-cell responses, with IFN-γ as the dominant cytokine, accompanied by significant production of TNF-α, IL-13, IL-8, and IL-6. Live C. trach

BACH2 effector-to-memory switch promotes HIV persistence and CAR-T efficacy.

PURPOSE OF REVIEW: HIV integration into BACH2 is significantly enriched in people living with HIV (PLWH), but not in vitro . Specifically, HIV integration sites are enriched in the same orientation and upstream of BACH2 translation. HIV drives high levels of BACH2 expression through HIV-to- BACH2 splicing. BACH2, a transcription repressor that competes with AP-1 binding, is an effector-to-memory switch that restrains effector function, drives T cell stemness, and promotes long-lived memory. RECENT FINDINGS: Retroviral infection mouse model containing retroviral splice sites recapitulated the

BASTA, a simple whole-blood assay for measuring β cell antigen-specific CD4+ T cell responses in type 1 diabetes.

Type 1 diabetes (T1D) is an autoimmune disease where T cells mediate the destruction of the insulin-producing β cells found within the islets of Langerhans in the pancreas. Autoantibodies to β cell antigens are the only tests available to detect β cell autoimmunity. T cell responses to β cell antigens, which are known to cause T1D, can only be measured in research settings because of the complexity of assays and the large blood volumes required. Here, we describe the β cell antigen-specific T cell assay (BASTA). BASTA is a simple whole-blood assay that can detect human CD4+ T cell responses to

IFN-inducible human phospholipid scramblase 1 (PLSCR1) protein restricts HIV-1 infection by inhibiting membrane fusion.

Human phospholipid scramblase 1 (PLSCR1) is an interferon-stimulated gene (ISG) that inhibits viral infections through various mechanisms. Here, we identify PLSCR1 as a host restriction factor that inhibits HIV-1 entry by impairing membrane fusion mediated by the envelope glycoprotein (Env). Using multiple cell types including the human SupT1 T cell line and purified CD4+ T cells, we demonstrate that PLSCR1 inhibits the replication of HIV-1 with diverse tropisms and subtypes, as well as HIV-2 and SIV. Mechanistically, we find that PLSCR1 blocks viral entry and cell-to-cell transmission, in par

Asymptomatic vivax malaria is associated with an IFN-γ-program on adaptive immunity.

The adaptive immunity against Plasmodium vivax is thought to be essential to limit parasite growth during asymptomatic malaria, preventing the occurrence of symptoms. However, the mechanisms governing clinical immunity during asymptomatic infections are not understood. Here, we investigated the adaptive cellular compartment in asymptomatic P. vivax-infected individuals (ASY) compared to symptomatic patients (SY) and healthy donors (CTL). Our integrative analysis revealed a TH1-biased immune signature with expanded populations of TH1 CD4+ T cells associated with the asymptomatic infection. In a

CD4+CD28+ T lymphocyte is associated with the 28-day mortality of patients with sepsis: a retrospective study.

BACKGROUND: Sepsis is a major health concern with high mortality, which is associated with immunosuppression. CD28, a co-stimulatory molecule on T lymphocytes, promotes T cell proliferation, survival, and cytokine production. CD4+CD28+ T cells play an important role in immune activation and regulation. This study aimed to determine whether CD4+CD28+ T lymphocytes were associated with 28-day mortality in patients with sepsis. METHODS: A retrospective analysis was performed in 80 adult patients with sepsis admitted to the department of intensive care unit. Peripheral blood CD4+CD28+ T cells wer
