DNA damage at an early developmental stage affects neurodevelopment in sterlet (Acipenser ruthenus).

BACKGROUND: Exposure to DNA damaging agents during fish embryogenesis can induce malformations and developmental disorders. However, there is scarcity of knowledge on dynamics of embryonic DNA repair and its effect on organogenesis. To address this knowledge gap, we exposed sterlet (Acipenser ruthenus) embryos to DNA-alkylating agent, methyl methanesulfonate (MMS), during blastulation (from 2 to 24 h post-fertilization (hpf)) and observed the effects at hatching (8 days post-fertilization (dpf)) and post-hatching stage (10 dpf). RESULTS: In contrast to control, DNA alkylation damage in treate

Cytokines and related signaling pathways in traumatic brain injury.

Traumatic brain injury (TBI) represents a critical public health challenge with profound consequences for patients' neurological function and quality of life. The delayed secondary injuries following TBI can lead to devastating long-term sequelae encompassing cognitive deficits, emotional disturbances, post-traumatic epilepsy, and neurodegeneration manifested as Alzheimer's disease or Chronic Traumatic Encephalopathy (CTE). Emerging evidence highlights neuroinflammation as a pivotal mechanism driving secondary injury progression, establishing it as a prime therapeutic target in TBI management.

UBE2M Identified by CRISPR Screening as a Key Regulator of Cisplatin-Induced Acute Kidney Injury via the p53 Pathway.

INTRODUCTION: Acute kidney injury caused by cisplatin (Cis-AKI) is a major limitation in its clinical use, primarily due to the lack of effective therapeutic targets to mitigate nephrotoxicity. Although several molecular pathways are involved in Cis-AKI, identifying reliable and actionable therapeutic targets has been challenging. Through a CRISPR-based genome-wide screening approach, UBE2M was identified as a novel gene involved in cellular survival during cisplatin-induced stress. However, its expression, biological function, and underlying mechanism in Cis-AKI have not been thoroughly inves

Targeting the ubiquitin-proteasome system: An emerging paradigm for overcoming resistance to targeted therapy in non-small cell lung cancer.

The introduction of tyrosine kinase inhibitors (TKIs) targeting driver oncogenes such as EGFR, ALK, and KRAS has substantially reshaped the treatment landscape of non-small cell lung cancer (NSCLC) and improved patient outcomes. However, inevitable acquired resistance remains the core obstacle to therapeutic success. Unlike previous studies that have largely focused on secondary mutations or bypass activation, the role of proteostasis imbalance in the evolution of resistance is increasingly recognized. The ubiquitin-proteasome system (UPS), as the central machinery for intracellular protein de

Integrative analysis identifies Hspa5 as a key regulator of the ERS/UPR-immune axis in spinal cord injury.

BACKGROUND: Endoplasmic reticulum stress (ERS) and the unfolded protein response (UPR) are critical in secondary spinal cord injury (SCI), but their systematic characterization and link to immune infiltration remain unclear. This study aimed to identify key ERS/UPR-related genes and their association with the immune microenvironment to find robust biomarkers for SCI. METHODS: We analyzed bulk RNA-seq data (GSE151371) to identify differentially expressed genes (DEGs), followed by pathway enrichment analyses. Machine learning (SVM-RFE) and protein-protein interaction networks were used to scree

Nitric oxide and hydrogen sulfide signaling pathways are involved in benign prostatic hyperplasia: pathophysiological development and novel therapeutic strategies.

Benign prostatic hyperplasia is a common pathological condition in aging men. Although its pathophysiology is not yet fully understood, dysfunctions in the nitric oxide (NO) and hydrogen sulfide (H2S) signaling pathways may be critically involved. In the present review, we address the physiological roles of these gasotransmitters in the prostate and discuss how their modulation may be a valuable therapeutic strategy in benign prostatic hyperplasia. Reduced NO bioavailability, decreased expression of soluble guanylate cyclase, and increased oxidative stress are associated with prostatic smooth

CXCL3 deficiency ameliorates COPD by suppressing cigarette smoke-induced NETs formation via MAPK signaling.

Chronic obstructive pulmonary disease (COPD) is a common respiratory disorder characterized by persistent airflow limitation, and its pathogenesis is closely associated with cigarette smoke (CS) exposure. Neutrophil extracellular trap (NET) formation contributes critically to airway inflammation and tissue damage in COPD. However, the upstream regulators of NETosis remain poorly defined. In this study, a CS-induced mouse model of COPD was established, and transcriptomic sequencing of lung tissues was performed to identify differentially expressed genes. CXCL3 was significantly upregulated in C

Proteomic Analysis of Extracellular Vesicles Identifies CDCP1 as Critical Metastasis-Related Glycoprotein in Lung Cancer.

Lung cancer is the most prevalent malignancy worldwide, with the majority of fatalities attributed to metastasis. Recent studies have demonstrated the pivotal role of extracellular vesicles (EVs) and glycoproteins in tumor progression. In this study, we compared the glycoproteome of EVs from 95C (low metastatic) and 95D (high metastatic) lung cancer cells to discover key targets in metastasis. Through coupling lectin affinity chromatography with quantitative proteomics, 1562 glycoproteins were identified. Compared to 95C EVs, 23 glycoproteins were significantly upregulated more than 20-fold in

T cell-intrinsic PRR signaling in immunity and pathology.

The immune system orchestrates a delicate balance between robust defense against pathogens and restraint to prevent tissue damage, with T cells serving as central mediators of adaptive immunity. The canonical pathway for T-cell activation hinges on the precise recognition of peptide antigens presented by major histocompatibility complex (MHC) molecules via the T-cell receptor (TCR), which is complemented by essential co-stimulatory signals. However, this model alone cannot fully explain the nuanced contextualization of immune responses, particularly how T cells integrate signals related to the

METTL3 promotes podocyte pyroptosis in diabetic nephropathy through N6-methyladenosine modification of TRIM29 mRNA.

Multiple studies have revealed the critical roles of epigenetic modifications in the development of diabetic nephropathy (DN). Methyltransferase-like 3 (METTL3)-mediated N6-methyladenosine (m6A) RNA modification in podocytes represents a new disease mechanism in DN. The tripartite motif-containing (TRIM) family member TRIM29 was reported to promote podocyte pyroptosis by activating the nuclear factor-κB/NLR family pyrin domain containing 3 (NLRP3) inflammasome pathway. However, whether METTL3-mediated m6A modification of TRIM29 mRNA is involved in podocyte injury remain unknown. Here, we found

Decoding Cellular Communication Networks and Signaling Pathways in Bone, Skeletal Muscle, and Bone-Muscle Crosstalk Through Spatial Transcriptomics.

Bone and skeletal muscle are essential components of the musculoskeletal system, enabling movement, load-bearing, and systemic regulation. These tissues communicate through dynamic bone-muscle crosstalk mediated by cytokines, growth factors, and extracellular matrix (ECM) proteins. The spatial organization of these mediators is critical to maintaining tissue integrity, and disruptions contribute to diseases such as osteoporosis, sarcopenia, and metabolic syndrome. Despite the importance of spatial context, studies using spatial transcriptomics (ST) to investigate bone-muscle interactions remai

Disruption of purine de novo synthesis pathway impairs membrane homeostasis, intracellular survival, and virulence of Brucella melitensis.

INTRODUCTION: Brucellosis is a significant zoonotic infectious disease caused by Brucella. As a facultative intracellular pathogen, the ability of Brucella to acquire essential nutrients within host cells is critical for its intracellular survival and pathogenicity. Previous studies have indicated that disruption of the de novo purine biosynthesis pathway significantly attenuates the virulence of Brucella, although the underlying mechanisms remain incompletely understood. METHODS: Using Brucella melitensis M5 as the parental strain, we constructed a deletion mutant and a complemented strain o

Toxicological profile of chemical interference of thyroid hormone metabolism via deiodinase enzymes in a rodent model.

Thyroid hormones (THs) are highly iodinated molecules that regulate many aspects of mammalian physiology and are essential for normal growth and development. Deiodinases (Dio) are a family of metabolizing enzymes controlling systemic and local availability of the major TH secreted from the thyroid gland. Despite their critical role in TH regulation, the toxicological characterization of the effects of chemical interference of Dios in mammalian models has remained relatively underexplored. Here, we investigated the effects of exposure to iopanoic acid (IOP), a potent in vitro inhibitor of Dio e

Galectin-1 regulates scar hyperplasia by modulating NASP variable splicing to generate ROS.

Galectin-1, a constituent of the mammalian β-galactoside-binding lectin family, plays a pivotal regulatory role in fibrotic cascades. Dysregulated fibrogenic cellular activity has been implicated as a critical driver of hypertrophic scar (HS) pathogenesis. Nevertheless, the precise mechanistic contributions and molecular pathways through which Galectin-1 modulates HS development remain incompletely characterized. qRT-PCR and western blot techniques were employed to explore the expression of Galectin-1 in hypertrophic scar tissues and cells. The Galectin-1 knockdown cell line was established by
