The SINE-encoded B2 retrotransposon is an RNA Polymerase III (POL-III)-derived transcript whose expression is substantially upregulated during various cellular stress responses. Beyond retrotransposition, the B2 non-coding RNA can directly bind and repress the activity of RNA Polymerase II (POL-II), leading to a significant downregulation of transcripts during stress. Notably, our recent findings have shown that B2 is a self-cleaving ribozyme whose activity can be induced by interactions with chromatin-modifying factors through non-canonical epigenetic mechanisms that co-regulate its function

Functional MRI (fMRI) and single-cell transcriptomics are pivotal in Alzheimer's disease (AD) research, each providing unique insights into neural function and molecular mechanisms. However, integrating these complementary modalities remains largely unexplored. Here, we introduce scBIT, a novel method for enhancing AD prediction by combining fMRI with single-nucleus RNA (snRNA). scBIT leverages snRNA as an auxiliary modality, significantly improving fMRI-based prediction models and providing comprehensive interpretability. It employs a sampling strategy to segment snRNA data into cell-type-spe

Amyotrophic lateral sclerosis (ALS) is a devastating neurodegenerative disease characterized by the selective loss of motor neurons. Among its genetic subtypes, mutations in the fused in sarcoma (FUS) gene represent an aggressive form, often associated with early onset and rapid progression. FUS is a ubiquitously expressed DNA/RNA-binding nuclear protein involved in maintaining DNA damage repair and RNA metabolism. It also plays a crucial role in the formation of ribonucleoprotein (RNP) granules such as cytoplasmic stress granules and nuclear paraspeckles under stress. In ALS, pathogenic FUS m

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder distinguished by progressive motor neuron degeneration, with diverse clinical manifestations and complex genetic and environmental triggers. The variability in disease progression underscores the necessity for tailored diagnostic and therapeutic approaches. MicroRNAs (miRNAs), small non-coding RNAs that regulate gene expression, have emerged as promising biomarkers and therapeutic targets in ALS. Dysregulation of specific miRNAs has been linked to mechanisms of ALS, including neuromuscular dysfunction, neuroinflammation,

Pediatric acute myeloid leukemia (AML) is a heterogeneous hematologic malignancy that accounts for about 15%-20% of childhood leukemias. Despite therapeutic advances, relapses remain common, and survival for high-risk patients is below 60%. Unlike adult AML, pediatric AML displays distinct genetic mutations, including FLT3-ITD, NPM1, KMT2A rearrangements, and core-binding factors (CBF) fusions, as well as extensive epigenetic dysregulation. Aberrant DNA methylation, histone modifications, and altered non-coding RNA expressions disrupt hematopoietic differentiation and activate oncogenic transc

mRNA plays a pivotal role in cellular processes of genetic information transfer, and post-transcriptional modifications of its nucleotides enable regulation of these processes with each particular mRNA. m5C methylation is a specific RNA modification that is rather common in tRNA, rRNA, lncRNA and other types of non-coding RNAs, whereas in mRNA it is found not very often. However, the functioning of m5C-methylated mRNAs differs from the non-methylated ones quite dramatically. Of the eight m5C RNA methyltransferases in humans, only two, NSUN2 and NSUN6, were found to be capable of modifying the

To identify efficient cellulose-degrading microbes suitable for the animal intestinal environment and to address the low utilization of crude fiber in feed, eight cellulolytic strains were isolated from the ileum of Yangzhou geese. Among them, strain A2 showed the highest cellulolytic activity (D/d = 1.48) via the CMC (carboxymethyl cellulose) agar transparent zone method. Based on whole-genome-based identification, strain A2 was identified as Bacillus subtilis. Whole-genome sequencing revealed a circular chromosome of 4.02 Mb with a GC content of 43.72%, containing 4083 protein-coding sequenc

microRNAs (miRNAs) are master regulators of gene expression, guiding ARGONAUTE proteins to bind to and repress target RNAs. Interestingly, a special class of target RNAs, termed target mimics (TMs), can in turn trigger miRNA degradation in plants-a process genetically dependent on the F-box protein HAWAIIAN SKIRT (HWS). However, the pairing rules governing effective TM sites remain unclear. Here, we systematically investigate the pairing architectures that enable plant TM-directed miRNA degradation. Plant diseases impose major constraints on global crop productivity and pose a major threat to food security. Here, we review transcription factors (TFs) as central orchestrators of plant defense, consolidating recent advances in how these regulators connect pathogen perception to immune signaling, transcriptional reprogramming, and durable defense responses. Initially, we combined a literature-based synthesis with a natural language processing (NLP) analysis of 1647 PubMed abstracts published between 2021 and 2026 to map dominant and underexplored TF families associated with plant immunity. Alzheimer's disease (AD) is a complex neurological ailment that is associated with memory loss, confusion, and mood disturbances. Genetic, molecular, and cellular factors, including oxidative stress, inflammation, neurotransmitter alterations, and amyloid β (Aβ) plaques and neurofibrillary tangles (NFTs), are associated with the disease. These can be associated with protein and DNA damage, mitochondrial dysfunction, energy shortages, inflammation, and hippocampal neuron death. Circular non-coding RNAs (circRNAs) are covalently closed and essential to many physiological and pathological process

MOTIVATION: Phylogenetics faces a growing challenge from increasingly large and complicated data sets enabled by ever-improving sequencing technologies. The issue is particularly acute for somatic evolution studies, such as cancer cell lineages, where single-cell data sets may include tens of thousands of mutations in hundreds of thousands of genetically distinct cells. Simultaneously, the biological complexity of somatic evolution has led to complex phylogeny methods that struggle to scale to even modest data sizes.
RESULTS: We explore the theoretical and empirical basis for one strategy for

Male infertility affects approximately 7 to 8% of men worldwide and is influenced by both genetic and epigenetic factors. Epigenetic mechanisms - including DNA methylation, histone modifications, and non-coding RNAs - are essential for spermatogenesis, sperm function, and early embryonic development. Disruptions in these processes can impair fertility and embryo viability. Assisted reproductive technologies (ART), particularly intracytoplasmic sperm injection (ICSI), have enabled conception in previously untreatable cases; however, by bypassing natural selection barriers, they may facilitate t

PURPOSE OF REVIEW: Gene expression is regulated at multiple levels beyond genetic and epigenetic modifications of DNA. Even when DNA is accurately transcribed into RNA, transcript abundance often does not correlate with protein levels, underscoring the importance of posttranscriptional regulation. Chemical modifications of RNA occur in both coding and noncoding RNAs and add an additional layer of gene control. More than 170 distinct RNA modifications have been identified across different RNAs in all domains of life. Although the abundant modifications have been investigated, their roles in car

Atypical sensory processing is a common feature of autism, yet the neural computations that give rise to these differences, particularly in relation to biological sex and etiological origin, remain unclear. Here we examine predictive auditory processing at the single-neuron level in the inferior colliculus of two adult rat models of autism: a genetic model with a heterozygous Grin2b deletion (Grin2b + /-) and an environmental model based on prenatal valproic acid exposure. We recorded neuronal responses to an auditory oddball paradigm and a cascade control sequence across lemniscal and non-lem

Exchange of genetic information by natural transformation shapes bacterial evolution. In Helicobacter pylori it is thought to drive its unusually high recombination rate, which has a crucial role in the evolution of virulence and the propagation of antibiotics resistance genes. While in most cases uptake of the incoming DNA into the periplasm is mediated by type IV pili, in H. pylori this initial step of natural transformation requires ComB, a unique competence-specific type IV secretion system (T4SS). The mechanisms by which ComB mediates DNA uptake are still poorly understood, since T4SS are

OBJECTIVES: Successful transition from paediatric to adult healthcare is integral in the management of chronic conditions. Osteogenesis imperfecta (OI) is a rare genetic connective tissue disorder affecting the skeleton and several other organ systems, with lifelong implications for clinical management. Consensus guidelines for the transition of patients with OI do not currently exist. We aimed to evaluate our institution's transition program for patients with OI and identify strengths and weaknesses of the current process.
METHODS: Participants were recruited from April to August 2023 for th

Multiple myeloma (MM) is a plasma cell malignancy that originates in the bone marrow (BM) and is characterized by the clonal expansion of Bcell-derived plasma cells producing abnormal monoclonal immunoglobulins. Despite significant therapeutic advances that have improved patient outcomes, drug resistance remains a major obstacle to effective disease management. Genetic and epigenetic heterogeneity are key features of MM that drive disease onset, progression, and therapeutic resistance. Both early and advanced stages of MM exhibit global and locus specific DNA methylation abnormalities that sil
