Osteoporosis is likely to become the most common disorder of our aging population. An understanding of factors that affect the incidence of osteoporosis is critical if we are to successfully minimize the impact of this disorder on morbidity, mortality, and the cost of health care. This article will provide an overview of the frequency, distribution, and consequences of the various types of osteoporotic fractures in different populations. We will discuss the major risk factors, emphasizing those that are reversible. Fracture risk not only involves abnormalities of bone mass and architecture, bu

The purpose of this study was to explore the impact of having a child with ataxia-telangiectasia (A-T) as well as to assess parental understanding of the genetics of A-T and attitudes toward carrier testing. Sixty-eight parents of individuals with A-T were interviewed. Ninety percent of the parents correctly believed if there is a child with A-T, both are obligate heterozygotes. Only 9% knew each well sib had a two-thirds chance of being a carrier. Eighty-four percent would have their unaffected child tested for carrier status prior to age 18 years. Eighty-two percent believed heterozygosity i

In December, 1993, we initiated a pilot project in which DNA fragile X (fraX) testing was offered during routine prenatal or genetic counseling to all pregnant women seen at the Genetics & IVF Institute, most of whom were referred for the indication of advanced maternal age. A brochure on fragile X syndrome was sent to each patient prior to her appointment and was reviewed by a counselor or physician during the counseling session. As of June 1995, 3,345 patients were offered testing; 474 women with no identified family history of mental retardation or learning disability and 214 women with a p

The treatment of severe slipped capital femoral epiphysis (SCFE) remains problematic. Treatment of most mild and moderate cases has been simplified, not by our increasing knowledge of physiology or molecular genetics, but by the advances in radiographic imaging, metallurgy, and the development of cannulated screw technology. Most centers agree that the placement of a single pin or screw across the center of the epiphysis, stabilizing the unreduced epiphysis and keeping the implant out of the joint space until physeal closure is achieved, is the most efficient and effective method for the major

Because of growing evidence for genetic causes of Alzheimer disease, clinicians are often asked about risks for this disorder among relatives of patients. This paper briefly reviews the evidence for genetic and other causes of Alzheimer disease, including specifically its typical late-onset variant. Several recent developments in the molecular genetics of early-onset Alzheimer disease are described briefly. These findings, however, may not apply to more typical, late-onset Alzheimer disease. Genetic studies of the latter disorder are complicated by the censorship of any inherited predispositio

Social interactions are ubiquitous in nature and have the potential to affect trait evolution, particularly in group-living animals such as cooperative breeders. Interactions among conspecific individuals can affect the amount of additive genetic variation for a trait when the phenotype of an individual is also affected by the genotype of its social partner(s) via indirect genetic effects. Thus, quantifying both direct and indirect genetic effects of social partners is critical for understanding and predicting evolutionary trajectories. While much is known about maternal indirect genetic effec

ETHNOPHARMACOLOGICAL RELEVANCE: Scutellaria baicalensis Georgi (SBG, Huangqin), a prominent traditional Chinese medicine, has a long history of clinical use for clearing heat and detoxifying, a classic TCM therapeutic concept corresponding to its well-documented anti-inflammatory, antimicrobial and hepatoprotective activities in modern pharmacological research. Despite its widespread therapeutic benefits, growing clinical evidence has linked SBG and its preparations with herb-induced liver injury (HILI). However, the population-level incidence of SBG-related drug-induced liver injury (SBG-DILI

The investigation into two bodies unearthed in 1946, suspected to be those of an infamous pair of siblings executed in 1742 for murder, has undergone comprehensive genetic investigations. Four libraries were prepared and sequenced for each of the two remains, yielding a 4X and 14X mean genome wide coverage. The data suggests high bacterial content and a high amount of unspecific sequence, with only 5-10% of reads mapping to the human genome, as well as high duplication levels (44-76%). With the aid of a dedicated group of local historians, ten distant relatives descended from the duo were iden

BACKGROUND AND HYPOTHESIS: Risk factors for psychosis spectrum disorders have been studied for many years, but recent research has begun to focus on resilience factors that protect against illness onset and mitigate illness course. Understanding resilience pathways will help us to apply interventions that can prevent psychosis and/or significantly improve illness trajectory.
STUDY DESIGN: Recent studies emphasize two complementary approaches for identifying resilience pathways: (1) At the population genetics level-by examining how the effect of psychosis risk genes may be counterbalanced by g

Colorectal cancer (CRC) has raised considerable health concerns worldwide, with increasing incidence rates, specifically among younger populations. Despite remarkable progress in diagnosing and treating various diseases, the genetic basis of CRC remains only partially understood. This paper aims to examine novel genetic variants associated with CRC in Iranian patients. We performed whole-exome sequencing (WES) in two Iranian families with a history of early-onset CRC. Candidate variants were validated by Sanger sequencing and assessed for segregation. Pathogenicity was evaluated through compre

Standardized microhaplotype databases for eight diverse crops enable multiallelic analyses, comparative genetics, and breeding decisions. Microhaplotypes are short genomic segments that contain multiple tightly linked variants, providing multi-allelic data that can enhance genetic resolution compared to traditional biallelic single nucleotide polymorphism (SNP) markers. Here, we present the creation and utilization of separate microhaplotype databases for eight crop species representing diverse genome sizes, ploidy levels, and breeding systems. We developed a standardized, species-agnostic pip

Biodiversity is structured on the landscape through a combination of spatial, environmental, and stochastic factors. In freshwater river networks, the interaction of the linear river network and the three-dimensional terrestrial landscape creates multiple pathways for ecological connectivity that affect aquatic community structure through habitat filtering and dispersal dynamics. Here, we use the high throughput capacity of environmental DNA (eDNA) metabarcoding to explore the influence of spatial relationships and environmental heterogeneity across an expansive and diverse landscape. We colle

With the global population aging, intervertebral disc degeneration (IDD), an aging-related disease, has been confirmed to be an important cause of low back pain (LBP). LBP can affect people of all ages and has now become one of the leading noncommunicable diseases, resulting in reduced life expectancy and functional disability in patients. Previous studies have mostly focused on the effects of factors such as mechanical load and genetics on intervertebral disc degeneration. With the deepening of research, the focus has gradually shifted to metabolic disorders, among which lipid metabolism play

BACKGROUND: Restrictive cardiomyopathy (RCM) is a rare, severe cardiac disease with a heterogeneous genetic basis. Both genetic and nongenetic factors contribute to RCM pathogenesis. Identifying the underlying molecular causes is important for diagnosis and family screening. In this study, we investigated the genetic basis of RCM in a 52-year-old woman with a family history of RCM and heart disease.
METHODS: Genetic predisposition was evaluated using whole-exome sequencing (WES). Candidate variants identified in the proband were validated by Sanger sequencing and interpreted using bioinformat

Next-generation sequencing (NGS) has facilitated the widespread use of DNA metabarcoding in fields such as biodiversity, population research, and food adulteration. To improve the identification of animal-derived DNA in samples containing degraded and mixed DNA, a DNA metabarcoding system targeting the mitochondrial Cytb, COI, 16 S rRNA, and 12 S rRNA genes using targeted NGS (tNGS) was developed for vertebrate species. In line with the International Society for Forensic Genetics (ISFG) recommendations for non-human DNA analysis, the forensic performance of the novel assay was systematically e

Killer-cell immunoglobulin-like receptors (KIRs) are critical regulators of the innate immune system and are found on the surfaces of natural killer (NK) cells. The KIR encoding genes, located on chromosome 19q13.4, are genetically diverse and associated with HIV progression. However, there is limited knowledge on the diversity of KIR from Uganda and Botswana HIV-infected paediatric cohorts. We applied next-generation sequencing technologies on 312 participants (Uganda: n = 246, Botswana: n = 66), Pushing Immunogenetics to the next-generation (PING) bioinformatics pipeline, logistic regression

BACKGROUND: Primary carnitine deficiency (PCD) is an autosomal recessive rare disorder of carnitine cycle and carnitine transport caused by pathogenic variants in the SLC22A5 gene. The prevalence of PCD is unclear. This study aimed to estimate the carrier frequency and genetic prevalence of PCD using Genome Aggregation Database (gnomAD) data.
METHODS: The pathogenicity of SLC22A5 variants was interpreted according to the American College of Medical Genetics and Genomics (ACMG) standards and guidelines. The minor allele frequency (MAF) of the variants of the SLC22A5 gene in 807,162 individuals
