BACKGROUND: α-Smooth muscle actin (α-SMA)-positive myofibroblasts play a pivotal role in progression and metastasis of solid carcinomas. Epithelial-mesenchymal transition (EMT) of cancer cells and lymphogenesis of tumor microenvironment are the important events in tumor metastasis. This study aimed to investigate the relationship between the expression of myofibroblasts marker, α-SMA, and clinicopathological features, EMT, lymphogenesis, and prognostic status in oral tongue squamous cell carcinoma (OTSCC).
METHODS: Immunohistochemisty was used to detect α-SMA expression in 50 OTSCCs. EMT and …

Inflammation is part of the body's response to internal and external environmental stimuli that normally eliminate the aggressor agent and r … tive action against colon adenomas, breast, prostate, and lung cancers. The inflammation present on tumor microenvironment is characterized by leukocyte infiltration, ranging in size, distribution and composition, as: tumor-associated macrophages (TAM), mast cells, dendritic cells, natural killer (NK) cells, neutrophils, eosinophils and lymphocytes. These cells produce …

Microsatellite-stable (MSS) colorectal cancer (CRC) accounts for approximately 85% of all CRC cases and exhibits limited responsiveness to immune checkpoint inhibitors (ICIs), representing a major challenge in current cancer immunotherapy. At the tumor-cell intrinsic level, low tumor mutational burden (TMB), impaired neoantigen generation, aberrant activation of oncogenic pathways such as WNT/β-catenin signaling, and defects in interferon-γ signaling contribute to impaired immune recognition and immune escape. Within the TME, the accumulation of immunosuppressive populations, including regulat

Meningiomas are the most common intracranial tumors. DNA methylation analysis in benign and aggressive meningiomas showed decreased MIR21 methylation and overexpression of hsa-miR-21-5p in atypical and anaplastic tumors. Transcriptomic analysis of distinct WHO grade meningiomas showed multiple predicted hsa-miR-21-5p target genes as differentially expressed. They were mainly related to processes of intercellular and intracellular signaling. Intercellular communication in meningioma was investigated using the deposited scRNA-seq dataset and deconvolution of our RNA-seq data. We found WHO grade-

Immune checkpoint inhibitors (ICIs) have significantly changed the treatment landscape of several gastrointestinal (GI) malignancies, particularly microsatellite instability-high (MSI-H) and mismatch repair-deficient (dMMR) tumors. However, most GI cancers remain resistant to current PD-1/PD-L1-based immunotherapy because of complex and highly suppressive tumor microenvironments characterized by immune stromal exclusion, myeloid-driven immune suppression, defective antigen presentation, and adaptive immune resistance mechanisms. Emerging evidence suggests that alternative inhibitory pathways, …

UNLABELLED: BAP1 inactivation, observed across multiple human cancers, is linked to immune checkpoint blockade (ICB) resistance and adverse clinical outcomes. The mechanisms underlying BAP1-associated ICB sensitivity could provide potential targets to enhance ICB efficacy. In this study, we showed that BAP1 inactivation fosters an immunosuppressive tumor microenvironment, marked by increased infiltration of M2-like macrophages and neutrophils. Single-cell transcriptomic analysis revealed an expansion of SPP1+ neutrophils in ICB-treated, BAP1-inactivated tumors. These SPP1+ neutrophils displaye…

Poor response to Bacillus Calmette-Guérin (BCG) immunotherapy remains a major barrier in the management of patients with non-muscle invasive bladder cancer (NMIBC). Multiple factors are associated with poor outcomes, including biological aging and female sex. More recently, it has emerged that a B-cell-infiltrated pretreatment immune microenvironment of NMIBC tumors can influence the response to intravesically administered BCG. The mechanisms underlying the roles of B cells in NMIBC are poorly understood. Here, we show that B-cell-dominant tertiary lymphoid structures (TLSs), a hallmark featur

The delivery of therapeutics into sites of action by using cargo-delivery platforms potentially minimizes their premature degradation and fast clearance from the bloodstream. Additionally, drug-loaded stimuli-responsive supramolecular assemblies can be produced to respond to the inherent features of tumor microenvironments, such as extracellular acidosis. We report in this framework the use of pH-responsive polymersomes (PSs) manufactured using poly([N-(2-hydroxypropyl)] methacrylamide)35-b-poly[2-(diisopropylamino)ethyl methacrylate]75 as the building unit (PHPMA35-b-PDPA75). BACKGROUND: We aimed to evaluate the clinical significance of microvessel density (MVD), lymphatic vessel density (LVD), and cancer-associated fibroblasts (CAFs) in relation to tumor location in advanced colorectal cancer (CRC).
METHODS: Using immunohistochemistry, we examined 181 advanced CRC patients for CD31 and D2-40 to measure MVD and LVD, respectively, α-smooth muscle actin (SMA) and desmin to identify CAFs, and PTEN to examine genetic changes of CAFs. To evaluate the regional heterogeneity of these properties, we examined tissue from four sites (the center and periphery of the primary

Ovarian cancer, in particular epithelial ovarian cancer (EOC), is commonly diagnosed when the tumor has metastasized into the abdominal cavity with an accumulation of ascites fluid. Combining histopathology and genetic variations, EOC can be sub-grouped into Type-I and Type-II tumors, of which the latter are more aggressive and metastatic. Metastasis and chemoresistance are the key events associated with the tumor microenvironment that lead to a poor patient outcome. Kallikrein-related peptidases (KLKs) are aberrantly expressed in EOC, in particular, in the more metastatic Type-II tumors. KLKs…

Breast cancer is a highly heterogeneous disease characterized by distinct molecular subtypes, dynamic tumor-microenvironment interactions, and variable therapeutic responses. Despite the availability of multiple preclinical platforms, a major challenge remains the lack of a coherent multiscale framework capable of integrating biological complexity across experimental systems. This limitation reduces the predictive power of individual models and highlights the need for complementary strategies that reproduce disease progression across multiple biological scales. In this context, advanced imagin

The review summarizes recent advances, challenges, and future perspectives in targeted drug delivery systems (DDSs) for pancreatic cancer (P … uli (light, heat, magnetic fields). Importantly, DDSs are designed to remodel the immunosuppressive tumor microenvironment (TME) by reprogramming tumor-associated macrophages, inhibiting myeloid-derived suppressor cells, activating innate immune sensing pathways, and overcoming stromal immune exclusion via stroma-immune co-modulation or transcytosis-enabled penetration. …

Chimeric antigen receptor (CAR) T-cell therapy has delivered unprecedented clinical benefit in hematological malignancies, yet its successful translation to solid tumors remains a major unmet clinical challenge. Glioblastoma (GBM), the most aggressive primary brain tumor, exemplifies the barriers confronting cellular immunotherapy, including profound intratumoral heterogeneity, antigenic escape, a highly immunosuppressive tumor microenvironment, and anatomical constraints imposed by the blood-brain barrier. Together, these features limit CAR-T cell trafficking, persistence and sustained antitu…

Palmitoyltransferase ZDHHC5 (DHHC5) is a palmitoylation regulator that adds palmitate to various protein substrates. Its role in pancreatic cancer (PC) remains unclear, thus posing significant challenges to the promotion of the "cold tumor" immune microenvironment, the development of new drugs, and targeted therapy. In this study, through comprehensive bioinformatics analysis, we determined the association between DHHC5 and various immune and metabolic components which we refer to as post-translational modification checkpoint of palmitoylation (Palm-PTM-checkpoint). We found that in pancreatic

BACKGROUND: Resistance to immune checkpoint inhibitors (ICIs) in prostate cancer (PCa) is driven by a "cold" tumor immune microenvironment. Astragaloside IV (AS-IV), a major bioactive saponin isolated from Astragalus membranaceus, exhibits immunomodulatory potential, yet its direct molecular targets remain elusive. Concurrently, PPP1R14B is overexpressed in PCa, but its role in mediating immune evasion is unclear.
PURPOSE: To investigate whether AS-IV targets PPP1R14B to remodel the tumor immune microenvironment (TIME) and sensitize PCa to anti-PD-1 therapy.
METHODS: The AS-IV/PPP1R14B inter
