Impact of Various Inactivation Approaches on Surrogate Proteinaceous Particles for Sample Return Missions.

Return of Mars samples is a high priority in the planetary science community and has remained an enduring goal of planetary exploration programs. Development of sterilization techniques to prevent potential contamination of Earth's biosphere with unknown life-forms that could exist on planetary bodies requires the use of the most robust biological indicators. We argue that self-seeding proteinaceous particles (prions) represent the most robust biological agents found on Earth. To evaluate the impact of various sterilization techniques on prion activity, we used derivatives of yeast prion prote

Transcriptome Analysis of Saccharomyces cerevisiae and Hanseniaspora uvarum During Direct Co-Fermentation.

The ability of Hanseniaspora uvarum to improve wine quality is limited by Saccharomyces cerevisiae due to growth inhibition by S. cerevisiae. In this study, dual RNA-Seq was used to study the interaction mechanism of S. cerevisiae and H. uvarum at T1 (24 h) and T2 (72 h) during direct co-fermentation (DCF). The results indicated that H. uvarum exhibited a significant growth inhibition during DCF. The sugar consumption and ethanol production of S. cerevisiae were stronger than those of H. uvarum during single fermentation (SF). The presence of H. uvarum significantly reduced the sugar consumpti

DcH3.3 and DcNAC1 Regulate the Expression of UGT73A93 Involved in the Changes in Flower Colour and Fungal Resistance in Carnation.

Carnation (Dianthus caryophyllus) contains abundant flavonoid glycosides (FGs), which are important natural functional and colour components. However, there are few reports on the modification of UDP-glycosyltransferases (UGTs) in relation to flavonoids in carnation. In this study, we cloned and characterised a flavonoid 3'-O-glucosyltransferase (UGT73A93) in carnation in vitro. Overexpression of UGT73A93 in carnation and tobacco increased flavonoid glycoside accumulation, particularly kaempferol glycosides, while decreasing anthocyanin content and lightening flower colour. UGT73A93 also enhan

Anti-Mycotoxin Agents in Pig Production Through the Lens of Circular Economy and Life Cycle Assessment: A Comprehensive Review.

Mycotoxin contamination of cereal-based pig feeds is a persistent threat to swine health and global pork safety. Pigs are uniquely susceptible to mycotoxicosis, and the principal toxins-aflatoxins (AFs), deoxynivalenol (DON), zearalenone (ZEN), fumonisins (FUMs), and ochratoxin A (OTA)-cause impaired growth, reproductive failure, hepato- and nephrotoxicity, immunosuppression, and carry-over residues in edible tissue. Anti-mycotoxin feed additives, including mineral adsorbents, biotransforming agents, and multicomponent mycotoxin-detoxifying agents (MMDAs) combining clays, phytogenic antioxidan

Overexpression of the MAPK transcription factor gene MSS11 enhances ethanol biosynthesis in Saccharomyces cerevisiae via coordinated metabolic and stress-response reprogramming.

Bioethanol is a pivotal sustainable alternative to fossil fuels. Saccharomyces cerevisiae is the primary microorganism for its industrial production. Enhancing the fermentative performance of S. cerevisiae is crucial for sustainable bioethanol production. To systematically enhance the fermentative capacity of yeast, this study employed targeted genetic engineering to overexpress MSS11-a principal transcription factor within the mitogen-activated protein kinase (MAPK) signaling cascade. The resultant recombinant strain, Eng-1, demonstrated a marked 23.9% enhancement in final ethanol titer (18.9

Biallelic protein truncating EXOSC6 variants cause a neurodevelopmental disorder with cerebellar atrophy, ataxia, and global developmental delay.

The multisubunit RNA exosome complex provides an essential, highly conserved multifunctional 3' to 5' exoribonuclease activity in the eukaryotic nucleus and cytoplasm. Inherited bi-allelic single amino acid variants in the core of the RNA exosome complex have been implicated in causing Mendelian syndromes that affect brain development, collectively termed exosomopathies. The core RNA exosome consists of nine subunits, and pathogenic variants in eight of them (EXOSC1- 5 and EXOSC7-9) have been described in exosomopathies. Here, we describe a patient with cerebellar atrophy, ataxia, and global d

The malaria parasite Plasmodium falciparum Vps15 is a Phosphatidylinositol 3-kinase interactor important for host-cell cytosol delivery to the food vacuole and apicoplast and mitochondrion dynamics.

The malaria parasite Plasmodium falciparum is an obligate intracellular organism that spends an important part of its lifecycle inside human erythrocytes. The endocytosis of host-cell cytosol and its delivery to a lysosome-like organelle called the food vacuole are critical for the parasite's survival and proliferation. Recent work has started to identify some of the molecular players involved in this pathway, but much remains to be discovered. Evidence suggests that phosphatidylinositol-3-phosphate (PI3P) plays a central role in this process. In unicellular eukaryotes, such as yeast, PI3P is

Metabolic engineering of Saccharomyces cerevisiae for de novo biosynthesis of isoorientin.

The flavonoid glycoside isoorientin has demonstrated antitumor, cardioprotective, and gut-protective activities, as well as potential applications as a natural biopesticide and antimicrobial agent. However, large-scale production and industrial development of isoorientin are limited by its low abundance in plants. In this study, we used metabolic engineering to construct an isoorientin-producing budding yeast (Saccharomyces cerevisiae) cell factory. To enhance production of the intermediate naringenin, we overexpressed key genes in the shikimic acid and naringenin pathways, reinforced the cyto

Strain-level typing of Wickerhamomyces anomalus using Fourier transform infrared spectroscopy and whole-genome sequencing.

In industrial settings, identifying the source of microbial contamination is crucial for effective microbiological risk assessment. While various strain identification technologies exist, many struggle with practicality, accuracy, and reproducibility. Fourier Transform Infrared Spectroscopy (FT-IR) has emerged as a rapid method, demonstrating a strong correlation with whole-genome sequencing (WGS) for certain bacteria. However, its accuracy for identifying yeast strains has been limited. This study focuses on improving the accuracy of FT-IR for yeast strain identification by optimizing pretrea

Integrated transcriptomic and metabolomic analyses highlight the 2-phenylethanol impact on fermentation performance of Saccharomyces cerevisiae 31 from fermented sour fish.

2-Phenylethanol (2-PE), as a quorum-sensing signaling molecule, is widely present (produced by yeast) and can enhance yeast-driven fermentation flavors in foods such as wine. Previous studies have shown that 2-PE is also present/produced by the dominant strain S. cerevisiae 31 in fermented sour fish and affects its growth, metabolism and flavor. While extensive research has characterized the role of 2-phenylethanol (2-PE) as a quorum-sensing molecule in relatively simple fermentation systems like wine, its molecular mechanism in complex, high-protein matrices such as fermented sour fish remain

Bio-orthogonal Labeling of Chitin in Native Pathogenic Candida Species via the Chitin Scavenge Pathway.

The fungal cell wall is essential for the integrity of the cell, providing strength and shape, as well as protection against environmental stimuli. For pathogenic fungi, the cell wall is also the initial point of contact with the host. Specific cell wall features such as hypha tails and smaller glycan components modulate a wide range of fungal interactions with the immune defenses. Here, a bio-orthogonal labeling method utilizing N-acetyl-glucosamine (NAG) probes is developed to fluorescently label native, pathogenic yeast via the chitin scavenging pathway. A panel of NAG probes was assembled,

Characterization of Candida albicans myosin-1 motor function and its regulation by phosphorylation at TEDS site.

Class I myosin is a single-headed myosin, distributed from fungi to mammals. The pathogenic yeast Candida albicans expresses a solo class I myosin gene (CaMyo1). The yeast-to-hyphae transition of Candida albicans depends on CaMyo1 and phosphorylation at its TEDS site (S366). In this study, we characterized the motor activity of CaMyo1 using purified CaMyo1IQ2 (a truncated CaMyo1 containing the motor domain and two IQ motifs) recombinantly expressed in insect Sf9 cells. We found that S366 phosphorylation activated the actin-activated ATPase activity of CaMyo1IQ2 from ∼4 s-1 in the dephosphoryla
