Brain atrophy is well documented in various kinds of dementia, particularly in Alzheimer's disease. Here, we evaluated gray matter volume of 87 cortical and subcortical areas in 460 individuals characterized according to the Clinical Dementia Rating (CDR) as cognitively unimpaired (n = 352), undetermined (n = 72), or mild dementia (n = 36). We found a highly significant correspondence between increased dementia severity and reduced brain volume, particularly for the amygdala and temporal cortical areas, including the hippocampus, middle temporal gyrus, and inferior temporal gyrus. The negative…

Alzheimer's disease is a progressive neurodegenerative disease characterized by memory decline and the accumulation of abnormal protein aggregates in the brain. While the precise cause of Alzheimer's disease remains under investigation, recent research suggests that dysregulation of brain-specific microRNAs (miRs) plays a significant role in Alzheimer's disease pathogenesis. Brain-specific miRs are predominantly expressed within the central nervous system and are crucial for neuronal development, and function, potentially in brain disorders. This review identifies some key brain-specific miRs …

Functional ultrasound imaging (fUSI) has emerged as a promising non-invasive neuroimaging modality that leverages neurovascular coupling to capture hemodynamic changes associated with neuronal activity. This study investigates the feasibility of fUSI for brain state classification in both healthy and pathological conditions using computational simulations based on the Wilson-Cowan neural mass model. Simulated electrophysiological signals were transformed into fUSI-like traces via convolution with an experimentally derived hemodynamic response function (HRF). Pathological conditions representin…

PURPOSE: The common marmoset is a small nonhuman primate that has emerged as a valuable animal model in neuroscience research. Accurate analysis of brain tissue is crucial to understand marmoset neurophysiology and to model neurodegenerative diseases. Many studies to date have complemented magnetic resonance imaging (MRI) with histochemical staining rather than immunofluorescent labeling, which can generate more informative and higher resolution images. There is a need for high-throughput immunolabeling and imaging methodologies to generate resources for the burgeoning marmoset field, particul…

The γ-rhythm plays a key role in coordinating the activity of the major brain systems and facilitating higher-level neurological processes. Several pathological conditions are associated with impaired generation or regulation of γ-oscillations. Modulating the γ-rhythm using periodic signals is considered a potential way to halt and/or treat such neurodegenerative processes. Despite the extensive knowledge gained in this field over the last 70 years, a unified theory linking the effectiveness of γ-stimulation to the characteristics of the stimulus and the stimulated remains elusive. In this rev…

The increasing prevalence of multi-site diffusion-weighted magnetic resonance imaging (dMRI) studies potentially offers enhanced statistical power to investigate brain structure. However, these studies face challenges due to variations in scanner hardware and acquisition protocols. While several methods for dMRI data harmonization exist, few specifically address structural brain connectivity. We introduce a new distribution-matching approach to harmonizing structural brain connectivity across different sites and scanners. We evaluate our method using structural brain connectivity data from thr…

Brain metastases (BM) represent an unmet medical need in human medicine, and they are poorly documented in dogs. The aim of this multi-centre retrospective study was to report the clinical characteristics, primary solid cancer histology, advanced imaging findings, treatment modalities and potential prognostic factors in dogs with presumed BM that occurred either at the time of initial diagnosis or during follow-up. BM diagnosis was established through either imaging studies or histologic examination of specimens obtained during necropsy. A total of 58 client-owned dogs with histologically prov…

Despite advancements in understanding macroscopic and functional brain changes in developing nonhuman primates, a comprehensive in vivo understanding of underlying quantitative microstructural and compositional tissue changes remains elusive. To address this knowledge gap, we applied a quantitative multimodal magnetic resonance imaging (MRI) approach, integrating T1 relaxation time, R2* relaxation rate, and voxel-based morphometry, alongside hematology and serum chemistry analyses, in 94 rhesus monkeys of varying ages (62 males, 32 females; mean age = 5.66±2.08 yr; age range = 1.96-10.3 yr). O…

Alzheimer's disease (AD) is a progressive neurodegenerative disorder with limited treatment options, underscoring the need for novel therapeutic targets. The gut-brain axis has emerged as a critical bidirectional communication system, with growing evidence establishing gut dysbiosis as a causal factor in AD pathogenesis. This dysbiosis, characterized by a reduction in beneficial microbes and an increase in pro-inflammatory taxa, compromises intestinal and blood-brain barrier integrity, promoting systemic inflammation and the translocation of neurotoxic agents like lipopolysaccharide (LPS). Con…

BACKGROUND: Intraventricular blockage of cerebrospinal drainage by brain tumors has been proposed as a more powerful mechanism of producing high intracranial pressure (ICP) than tumor size. We tested that hypothesis by investigating the MRI signs closely associated with high ICP in brain tumors.
METHODS: We conducted a single-institution retrospective chart review of patients with brain tumors between 1990 and 2023. We selected 100 cases from a larger eligible cohort without regard to clinical or imaging features. Based on ophthalmologic examination, 57 patients had papilledema and 43 patient…

The gut–brain axis, a bidirectional communication pathway, permits the central nervous system (CNS) to exert influence over gastrointestinal function in response to stress, while the gut microbiota regulates the CNS via immune, neuroendocrine, and vagal pathways. Current research highlights the importance of the gut microbiota in stress-related disorders and the need for further research into the mechanisms of gut–brain communication, with potential therapeutic implications for a wide range of health conditions. This is a challenge taken on in this Scientific Reports Collection on the Gut-Brai…

BACKGROUND: Anatomical studies have indicated that the brain and the heart are connected through multiple pathways. However, the functional interplay between them is unclear, especially for different task states. This study explored the brain-heart interplay under reflexive and voluntary saccade tasks.
METHODS: The Synthetic Data Generation model was used to quantify the interplay between the brain and heart.
RESULTS: Bidirectional interplay patterns were found between the brain and heart under different frequency bands for the two types of saccade task. There were significant variations in …

Spindles are hallmark oscillations during non-rapid-eye-movement (NREM) sleep. Together with slow oscillations (SOs), they are thought to play a mechanistic role in the consolidation of learned information. The quantity and spatial distribution of spindles have been linked to brain activity during learning before sleep and to memory performance after sleep. If spindles are drawn to cortical areas excited through pre-sleep learning tasks, this begs the question of whether the spatial distribution of spindles is flexible, and whether their regional expression can also be manipulated with experim…

Brain aging is accompanied by profound changes in neuroimmune interactions that shape the balance between resilience and vulnerability. Under healthy conditions, glial cells, neurons, vascular elements, and peripheral immune inputs cooperate to sustain homeostasis. With advancing age, however, immune remodeling and systemic inflammaging drive shifts in microglial surveillance, astrocytic reactivity, and neuronal susceptibility, creating conditions that compromise synaptic function and cognitive performance. These processes unfold along a continuum, from subtle impairments in normal aging to ma…

INTRODUCTION: Alzheimer's disease (AD) is characterised by inter-individual heterogeneity in brain degeneration, limiting diagnostic and prognostic precision. We present a novel framework integrating Morphometric Inverse Divergence (MIND) networks with hierarchical Bayesian large-scale population modelling to identify individual-level neuroanatomical deviations.
METHODS: MIND networks quantify similarity between brain regions using multivariate MRI features. A normative model of regional MIND values trained on UK Biobank (N=35,133) was applied to the National Alzheimer's Coordinating Center c…

The lifetime prevalence of mood disorders such as major depressive disorder (MDD) is thought to approach up to 50% of the world's population. Traditionally, research into the mechanisms of these disorders has focused on neurotransmission, but emerging evidence highlights neuroimmune interactions-the molecular signaling between immune and brain cells-as key regulators of brain plasticity, affective behavior, and potential vulnerability to mood disorders. Chronic stress models have unearthed how immune cell responses modify neural circuit activity, synaptic connectivity, and behaviors relevant f…

UNLABELLED: Deep brain stimulation (DBS) of the ventral capsule/ventral striatum (VCVS) can treat obsessive-compulsive disorder (OCD) and other psychiatric conditions. Yet, optimizing its clinical efficacy is a major challenge, often hindered by incomplete knowledge of how stimulation parameters and targets affect neural activity and behavior. VCVS DBS is thought to work in part by improving cognitive control, an important decision-making component that is impaired in OCD and other illnesses. The magnitude of this cognitive control enhancement was shown to be lateralized, with right-unilateral…

Currently available therapeutic modalities for alcohol use disorder (AUD) produce limited effect sizes or long-term compliance. Recent methods that were developed to modulate brain activity represent potential novel treatment options. Various methods of brain stimulation, when applied repeatedly, can induce long-term neurobiological, behavioral, and cognitive modifications. Recent studies in alcoholic subjects indicate the potential of brain stimulation methods to reduce alcohol craving, consumption, and relapse. Specifically, deep brain stimulation (DBS) of the nucleus accumbens or non-surgic…

OBJECTIVE: The Blood-Brain Barrier (BBB) is the gatekeeper of the central nervous system, effectively shielding the brain from blood-borne threats but simultaneously representing a significant challenge for treating neurological diseases. Altering the permeability of the BBB enables increasing the local drug concentration enhancing therapeutic effects. This study aims to explore the BBB permeability alteration through localized mild temperature increases, addressing the challenge of accurately determining and monitoring thermal dosages during hyperthermia treatments.
METHODS: To investigate t…
