Sex steroid receptors in male human bladder: expression and biological function.

INTRODUCTION: In male, lower urinary tract symptoms (LUTS) have been associated, beside benign prostatic hyperplasia, to some unexpected comorbidities (hypogonadism, obesity, metabolic syndrome), which are essentially characterized by an unbalance between circulating androgens/estrogens. Within the bladder, LUTS are linked to RhoA/Rho-kinase (ROCK) pathway overactivity. AIM: To investigate the effects of changing sex steroids on bladder smooth muscle. METHODS: ER α, ER β, GPR30/GPER1 and aromatase mRNA expression was analyzed in male genitourinary tract tissues, and cells isolated from bladd

Pancreatic β-cell Na+ channels control global Ca2+ signaling and oxidative metabolism by inducing Na+ and Ca2+ responses that are propagated into mitochondria.

Communication between the plasma membrane and mitochondria is essential for initiating the Ca(2+) and metabolic signals required for secretion in β cells. Although voltage-dependent Na(+) channels are abundantly expressed in β cells and activated by glucose, their role in communicating with mitochondria is unresolved. Here, we combined fluorescent Na(+), Ca(2+), and ATP imaging, electrophysiological analysis with tetrodotoxin (TTX)-dependent block of the Na(+) channel, and molecular manipulation of mitochondrial Ca(2+) transporters to study the communication between Na(+) channels and mitochon

Nifedipine-modulated gut microbiota attenuates hypertensive vascular injury through Bifidobacterium-associated inosine-Adora2b signaling.

Although antihypertensive drugs mitigate hypertensive vascular injury, their vascular benefits are not fully explained by blood pressure reduction alone. Many cardiovascular drugs reshape the gut microbiota, which in turn influences their vascular pharmacology. We therefore investigated whether gut microbiota remodeling contributes to nifedipine-mediated vascular protection in high-salt hypertensive rats and a pilot patient cohort. Combining 16S rRNA sequencing, untargeted metabolomics, fecal microbiota transplantation, microbiota depletion, supplementation, and selective receptor antagonism,

Role and mechanism of ATF6-mediated endoplasmic reticulum stress in ITLN-1 against cerebral ischemia-reperfusion injury.

AIMS: Intelectin-1 (ITLN-1), a novel adipocytokine, exerts neuroprotective effects against cerebral ischemia-reperfusion injury (CIRI), yet its molecular mechanisms remain unclear. This study focused on activating transcription factor 6 (ATF6), a key endoplasmic reticulum stress sensor, to explore its role in CIRI-induced neuronal injury and determine whether ITLN-1 mediates neuroprotection through ATF6 regulation. MATERIALS AND METHODS: We established an N2a cell hypoxia/reoxygenation (H/R) model and a mouse middle cerebral artery occlusion (MCAO) model. Using protein molecular docking, situ

Species-specific functions of CSRP3 in satellite cell-driven muscle regeneration and relevance to porcine stem cell therapy.

BACKGROUND: Skeletal muscle function deteriorates with injury, disease, or aging. While stem cell therapies offer therapeutic potential, concerns remain regarding the safety and efficacy of transplanted cells. Satellite cells (SCs), the primary endogenous stem cells responsible for muscle repair, demonstrate safe transplantation but limited clinical effectiveness. A deeper understanding of the mechanisms regulating SC activation and differentiation is critical for improving muscle regeneration strategies. METHODS: We examined the role of CSRP3 in SC-mediated muscle repair using injury models

Ginsenoside Rd: molecular mechanisms, pharmacological activities, and therapeutic potential.

Ginsenoside Rd (Rd), a major protopanaxadiol saponin from Panax species, is a pleiotropic candidate with potential relevance for treating ischemic, inflammatory, metabolic, and degenerative diseases. Over the past two decades, evidence from cellular systems, animal models, and early clinical studies indicates that Rd provides multi-level protection by modulating oxidative stress, inflammatory signaling, calcium dysregulation, mitochondrial dysfunction, and regulated cell death. Mechanistic studies implicate interconnected pathways, including PI3K/Akt, ERK, NF‑κB, Nrf2‑mediated antioxidant prog

PF4/CXCR3 signaling contributes to the regulation of endothelial cells on smooth muscle cells in CKD vascular calcification.

Vascular calcification is a common complication of chronic kidney disease (CKD) that increases the risks of cardiovascular morbidity and mortality. However, current therapeutic strategies are unable to effectively cure vascular calcification. To elucidate the effect of endothelial cells (ECs) on vascular smooth muscle cells (VSMCs) in CKD vascular calcification, this study observed an increased platelet factor 4 (PF4) level in ECs by a rat CKD vascular calcification model established by 5/6 nephrectomy with a high‑phosphorus diet. The role of PF4 was further investigated using a Vitamin D3-ind

Nuclear Stress Bodies and Mitochondrial Signal Transduction Regulate Thermotolerance in Cattle: Insights Into Molecular Mechanisms.

Heat stress represents a major threat to cattle productivity, welfare, immune competence, and sustainability under climate change. Although genomic studies have identified loci associated with thermoregulatory traits, sequence variation alone does not explain the rapid, reversible cellular adaptations required during acute thermal challenge. Increasing evidence suggests that thermotolerance depends on coordinated regulation between nuclear transcriptional architecture and mitochondrial metabolic stability. This review synthesizes mechanistic insights into two interconnected regulatory axes: he

Tceal7 is a BRG1-regulated target of calcineurin signaling that promotes myoblast differentiation.

The transcriptional control of skeletal muscle differentiation requires the coordinated activity of lineage-defining transcription factors, signal-responsive regulators, chromatin modifiers, and ATP-dependent chromatin remodeling enzymes. Here, we identify TCEAL7, a member of the X-linked, poorly characterized TCEAL family of proteins, as a direct downstream target of BRG1-containing mammalian SWI/SNF (mSWI/SNF) complexes and calcineurin signaling during myoblast differentiation. Analyses of previously published datasets showed that pharmacological inhibition of mSWI/SNF bromodomains or knockd

Targeting eukaryotic elongation factor 2 (eEF2)/eEF2 kinase in neurological and neuropsychiatric Disorders: Mechanisms, therapeutic Implications, and translational challenges.

Eukaryotic elongation factor 2 kinase (eEF2K) phosphorylates eukaryotic elongation factor 2 (eEF2) and slows translation elongation. In the nervous system, this pathway links neuronal activity, calcium signaling, energy status, and stress responses to selective protein synthesis programs that shape synaptic plasticity, circuit excitability, and cell survival. Dysregulated eEF2K/eEF2 signaling has been implicated in epilepsy, Alzheimer's disease, Parkinson's disease, major depressive disorder, Down syndrome, and other brain conditions. However, the literature remains fragmented, largely preclin…

Disruption of dopamine transmission by cholesterol depletion is associated with alterations in protein lipid raft partitioning and actin dynamics.

Disrupted brain cholesterol homeostasis is implicated in neurological disorders involving aberrant dopamine (DA) signaling; however, the direct effects of cholesterol on DA transmission in native tissue have not yet been demonstrated. Using ex vivo fast-scan cyclic voltammetry in nucleus accumbens slices from male rats, we found that membrane cholesterol depletion with methyl-β-cyclodextrin (MβCD, 3-10 mM) significantly reduced evoked DA release and decreased the apparent maximal rate of DA reuptake via the dopamine transporter (DAT). Because cholesterol is critical for the formation of lipid
