Wharton's Jelly-Derived Mesenchymal Stem Cells with High Aurora Kinase A Expression Show Improved Proliferation, Migration, and Therapeutic Potential.

Mesenchymal stem cells (MSCs) are effective therapeutic agents that contribute to tissue repair and regeneration by secreting various factors. However, donor-dependent variations in MSC proliferation and therapeutic potentials result in variable production yields and clinical outcomes, thereby impeding MSC-based therapies. Hence, selection of MSCs with high proliferation and therapeutic potentials would be important for effective clinical application of MSCs. This study is aimed at identifying the upregulated genes in human Wharton's jelly-derived MSCs (WJ-MSCs) with high proliferation potenti

A rare case of malignant biliary stenosis due to retroperitoneal metastasis from breast invasive ductal carcinoma.

A 50-year-old woman was referred to our hospital for elevated hepatobiliary enzymes. She had a medical history of mastectomy for left breast invasive ductal carcinoma about 10 years ago, and no apparent recurrence had been observed. Contrast-enhanced computed tomography (CT) revealed soft-tissue shadows surrounding the portal vein, celiac artery, and other vessels. The lesions involved the hilar bile duct, and the upstream bile ducts were dilated. Endoscopic retrograde cholangiography showed an obstruction in the hilar bile duct, and biopsies were taken at the site of biliary stenosis. H&E sta

The role of apoptosis and autophagy in the insulin-enhancing activity of oxovanadium(IV) bipyridine complex in streptozotocin-induced diabetic mice.

Metal-based therapies (e.g. Vanadium) possess an attractive proposition in medicinal treatment of diabetes mellitus. Defective insulin secretion can result from impaired β-cell function which is mediated by many process including apoptosis and autophagy. In this study. diabetes was induced by administration of streptozotocin then treatment was performed by vanadyl sulfate and [VO(bpy)2 Cl] Cl.H2O complex. Blood glucose level, AST, ALT, BUN, CR, TCHO, TG and total protein were determined in serum. MDA, NO, erythrocyte GSH and SOD were estimated. LC3 and Caspase 3 levels in pancreatic cells were

The Enigma of Heat Shock Proteins in Immune Tolerance.

The fundamental problem of autoimmune diseases is the failure of the immune system to downregulate its own potentially dangerous cells, which leads to destruction of tissue expressing the relevant autoantigens. Current immunosuppressive therapies offer relief but fail to restore the basic condition of self-tolerance. They do not induce long-term physiological regulation resulting in medication-free disease remissions. Heat shock proteins (HSPs) have shown to possess the capacity of inducing lasting protective immune responses in models of experimental autoimmune diseases. Especially mycobacter

Toll-Like Receptor 4 Agonistic Antibody Promotes Host Defense against Chronic Pseudomonas aeruginosa Lung Infection in Mice.

Chronic lower respiratory tract infection with Pseudomonas aeruginosa is difficult to treat due to enhanced antibiotic resistance and decreased efficacy of drug delivery to destroyed lung tissue. To determine the potential for restorative immunomodulation therapies, we evaluated the effect of Toll-like receptor 4 (TLR4) stimulation on the host immune response to Pseudomonas infection in mice. We implanted sterile plastic tubes precoated with P. aeruginosa in the bronchi of mice, administered the TLR4/MD2 agonistic monoclonal antibody UT12 intraperitoneally every week, and subsequently analyzed

Prognostic role of nuclear factor/IB and bone remodeling proteins in metastatic giant cell tumor of bone: A retrospective study.

Giant cell tumor of bone (GCTb) represents 5% of bone tumors, and although considered benign, 5% metastasize to the lung. The expression of proteins directly or indirectly associated with osteolysis and tumor growth was studied on 163 samples of GCTb. Of these, 33 patients developed lung metastasis during follow-up. The impact of tumor-host interaction on clinical aspects was evaluated with the aim of finding specific markers for new biological therapies, thus improving clinical management of GCTb. Protein expression was evaluated by immunohistochemical analysis on Tissue Microarray. The major

Idiopathic bilateral vestibulopathy: an autoimmune disease?

Bilateral vestibulopathy (BV) is the loss of function of both peripheral labyrinths or of the eighth nerves. Its etiology remains obscure in approximately 20% to 50% of cases (so-called idiopathic bilateral vestibulopathy, IBV). Alternatively, the cause could be viral or vascular; to date, causative gene mutations have not been identified. Other potential disease mechanisms include autoimmune disorders. Antibodies have been detected against inner ear tissue (primarily against vestibular membranous labyrinth). The data suggest that the bulk of anti-labyrinthine autoantibodies may be an epipheno

Cardiovascular diseases and the nitric oxide pathway.

Fifteen years after its discovery, NO has fully reached an established position in physiology, medicine and therapeutics. It is difficult to find a biological function or a pathological condition where NO does not play a relevant role. Discoveries in the NO field have historically evolved from cardiovascular research, although its influences have already covered nearly all the medical specialties. This review analyzes, step by step, the pathway through which NO is synthesized in the cells of the cardiovascular system and the main physiological and pathological routes it undergoes once it is re

Tissue proteome analysis for profiling proteins associated with lymph node metastasis in gallbladder cancer.

Lymph node (LN) metastasis is the earliest sign of metastatic spread and an established predictor of poor outcome in gallbladder cancer (GBC). Patients with LN positive GBC have a significantly worse survival (median survival- 7 months) than patients with LN negative disease (median survival- ~ 23 months) in spite of standard treatment which includes extended surgery followed by chemotherapy, radiotherapy and targeted therapy. This study aims at understanding the underlying molecular processes associated with LN metastasis in GBC. Here, we used iTRAQ-based quantitative proteomic analysis using

Microarray analysis of distinct gene transcription profiles in non-eosinophilic chronic sinusitis with nasal polyps.

BACKGROUND: Recent literature has indicated the feasibility of microarray analysis in the characterization of chronic sinusitis. We hypothesized that previously unexplored inflammatory mechanisms would be involved in the pathophysiology of noneosinophilic chronic rhinosinusitis with nasal polyps (NE-CRSwNP) and that this technology could be used to identify the gene expression of these novel and previously known mediators. METHODS: Patients with CRSwNP failing medical therapy were prospectively enrolled and NP tissue was removed at time of surgery. NE-CRSwNP was diagnosed based on clinical pa

Natural killer T cells in intracerebral hemorrhage: phase-dependent immune responses and therapeutic implications.

BACKGROUND: Intracerebral hemorrhage (ICH) is the most lethal subtype of stroke, yet effective disease-modifying therapies remain limited. Beyond the primary mechanical insult, dysregulated neuroinflammation is thought to be a major contributor to hematoma expansion, perihematomal edema, and secondary neuronal injury. Natural killer T (NKT) cells, a specialized population of lipid-reactive lymphocytes linking innate and adaptive immunity, have emerged as potentially relevant immunoregulatory contributors to the post-hemorrhagic response. MAIN BODY: This review summarizes current experimental

High-Throughput Screening of a Lipid Nanoparticle (LNP) Library Identifies Lead Formulations for CRISPR-Cas9 Ribonucleoprotein Delivery and Gene Editing.

Delivery of CRISPR-Cas9 ribonucleoprotein (RNP) using synthetic materials can bypass the host translational machinery and limit the duration of genomic exposure, minimizing the risk of off-target mutations. Despite their advantages, lipid nanoparticle (LNP) platforms for CRISPR-Cas9 RNP delivery that achieve highly efficient editing at low doses remain limited. In the present study, we developed a library of ionizable lipids (ILs) and corresponding LNPs to enhance the delivery and editing efficiencies of CRISPR-Cas9 RNP complexes. Following comprehensive in vitro screening and formulation opti

Mapping the Ischemic Continuum: Dynamic Multi-Omic Biomarker and AI for Personalized Stroke Care.

Although there have been advancements in stroke treatment (reperfusion) therapy, and it has been shown that many individuals continue to suffer from partial recoveries and continuing decline in their neurological status as a result of suffering a stroke, a primary barrier to providing precise care to patients with stroke continues to be the inability to capture changes in molecular and cellular programs over time and in biological compartments. This review synthesizes evidence that represents the entire continuum of ischemia, beginning with acute metabolic failure and excitotoxicity, and endin

From blueprint to build: Metal ions in peripheral nerve development and engineering regeneration.

Peripheral nerve injury (PNI) poses a substantial global health burden, affecting over 20 million individuals annually with frequent suboptimal recovery and persistent disability. The inherent limitations of nerve autografts, such as donor site morbidity and limited supply, underscore the clinical importance of nerve guidance conduits (NGCs) as a promising alternative. Nonetheless, the efficacy of current NGCs remains limited by their inability to recapitulate the spatiotemporally precise molecular cues of the native regenerative microenvironment. Our network meta-analysis highlights a signifi

In Silico Reconstruction of Primary and Metastatic Tumor Architecture Using Geographic Information System-Augmented Spatial Transcriptomics.

UNLABELLED: The tumor microenvironment comprises different cell populations that interact, contributing to tumor heterogeneity and therapy response. Spatial transcriptomics offers valuable insights into the transcriptional complexity and heterogeneity of the tumor microenvironment. In this study, we established a geographic information system (GIS)-augmented in silico reconstruction of tumor architecture (GIS-ROTA), a biologically informed analytic framework that integrates pathway or cell type-based enrichment analysis with spatial autocorrelation measurement to uncover functional spatial dom

The dialogue between breast cancer and microorganisms.

Breast cancer is a complex pathological process involving multiple factors and stages, characterized by pronounced molecular and phenotypic heterogeneity. Its global incidence and mortality rates have shown a continuous upward trend. With the advancement of microbiome research, microbial communities have been recognized as key determinants influencing host health and disease states. Increasing evidence suggests a close association between breast tissue-resident and systemic microbiota and the initiation and progression of breast cancer. Specifically, microorganisms may be associated with abnor
