Cotransmission from sympathetic vasoconstrictor neurons to small cutaneous arteries in vivo.

This study has characterized constrictions of small cutaneous arteries in the guinea pig ear in response to electrical stimulation of the cervical sympathetic nerve (SNS) in vivo. Video microscopy and on-line image analysis were used to examine diameter changes of ear arteries (80-140 micrometers resting diameter) in anesthetized guinea pigs. Trains of 50-300 impulses, but not single pulses or short trains, produced frequency-dependent (2-20 Hz) constrictions. The purinoceptor antagonist suramin (30 microM) greatly reduced constrictions produced by exogenous ATP but did not affect constriction

A new sensitive cardiac Troponin T rapid test (TROPT) for the detection of experimental acute myocardial damage in rats.

Cardiac Troponin T (cTnT) is a cardiac structural protein which is released in the circulation during myocardial cell damage. In this study we addressed the question of whether beta-sympathomimetic induced myocardial cell damage in rats can be detected in blood by the TROPT sensitive rapid test strip which is primarily manufactured for the detection of acute myocardial infarction in humans. Sixteen male rats and 16 female animals were treated once with orciprenalinesulphate s.c. to induce tachycardia. The control group which consisted of 16 rats of both sexes received vehicle (physiological sa

[Pain caused by ephaptic transmission occurring in the recovery phase of diabetic vascular mononeuropathy was effectively suppressed with L-threo-3,4-dihydroxyphenyl-serine].

A 59-year-old man with an 8-years history of diabetes mellitus had an acute onset of sharp pain in the anterior part of right lower leg. One month later, the pain changed to deep dull nature in the deeper site of the peroneal region. The pain increased, when he stood up, walked, and stayed in the cold room and decreased by rest. It was more painful in the evening than in the morning. The first sharp pain was thought to be caused by ischemia due to diabetic vascular neuropathy. The next dull pain was considered to be originated from ephaptic transmission between sympathetic efferent fibers to g

Sequential relationship between actions of CRF and PGE2 in the brain on splenic sympathetic nerve activity in rats.

An infusion of corticotropin-releasing factor (CRF, 0.1-5.0 micrograms/10 microliters/rat) into the third cerebral ventricle (i.c.v.) for 10 min increased the splenic sympathetic nerve activity in a dose-dependent manner in urethane + alpha-chloralose anesthetized rats. No changes in the arterial blood pressure and body temperature were observed. The CRF (1.0 micrograms/10 microliters)-induced enhancement of the splenic sympathetic nerve activity was completely blocked by an i.c.v. pretreatment with a CRF antagonist, alpha-helical CRF9-41 (alpha-hCRF, 10 micrograms/10 microliters). The increas

Does orthostatic testing have any role in the evaluation of the young subject with mild hypertension?: an insight from the HARVEST study.

The aim of the study was to assess the clinical significance of the blood pressure (BP) reaction to standing in 1029 stage I hypertensives. Office BP was measured six times in the supine position and six times after 2 min of standing. All subjects underwent 24-h ambulatory BP monitoring, and measurements of 24-h urinary epinephrine and norepinephrine excretion. Echocardiography was performed in 636 patients. With use of mixture analysis we could single out a population with abnormal diastolic BP response to standing (hyperreactors, n = 95). These subjects had a diastolic BP increase from lying

Reduced sympathetic sprouting occurs in dorsal root ganglia after axotomy in mice lacking low-affinity neurotrophin receptor.

Following sciatic nerve injury, sympathetic axons sprout into the L5 dorsal root ganglion (DRG). The low-affinity neurotrophin receptor (p75) is strongly expressed by glial cells in DRG following axotomy. We tested the hypothesis that p75 is involved in this sympathetic sprouting by comparing sprouting following sciatic nerve cut in wild-type (CD1) and p75 knockout mice. Twenty-one days after injury sympathetic sprouting had occurred in both mouse strains, though the extent of sprouting (measured as length of sprouts per unit area of DRG) was less in p75 knockout mice. We conclude that p75 exp

[Effects of octreotide on experimental orthostatic neurogenic hypotension].

The synthetic somatostatin analogue, octreotide, has recently been proposed for the treatment of both postprandial and orthostatic hypotension (OH) in humans with autonomic failure related to multiple system atrophy (MSA) or diabetes mellitus. However, pharmacodynamic data are not still available in experimental models of orthostatic hypotension. We investigated in a model of neurogenic orthostatic hypotension, obtained by chronic sinoaortic denervation (SAD) in chloralose-anaesthetized dogs, the effects of octreotide (0.1 mg/kg, subcutaneous route) during a double-blind cross-over study vs pl

[Periodic decerebrate spasm with ocular dipping, Cheyne-Stokes respiration and hypersympathetic activity].

Decerebrate spasm is a generalized muscular spasm produced by some stimuli on decerebrate posture. Such spasm are called "tonic fit" or "decerebrate extensor spasm". We reported a 50-year-old man with periodic decerebrate spasm after cerebral hemorrhage. On admission, the patient was comatose. The pupils were round but anisocoric and did not react to light. Corneal reflexes were absent. The face, arms, and legs did not move voluntarily. Two weeks after admission, he was found in decerebrate rigidity. Periodic decerebrate spasms were also observed and were accompanied by ocular dipping. Cheyne-

[Change in cross-bridge activation rate constant (Kac) after six-minute walk in patients with heart failure].

Cardiac adrenergic activity is increased in patients with congestive heart failure (CHF) and is excessively increased during mild exercise. Changes in cardiac adrenergic activity were examined in normal individuals and patients with CHF before and after mild exercise using the cross-bridge activation rate constant (Ka), which may represent the adrenergic activity related to the working left ventricular myocardium. Ten normal volunteers and 31 patients with stable CHF underwent echocardiography. The left ventricular ejection fraction (LVEF) was measured in all the CHF patients using left ventri

The effects of low-frequency ultrasound on the inner ear: an electrophysiological study using the guinea pig cochlea.

By examining 218 albino guinea pigs, electrophysiological methods were used to investigate the effects of low frequency ultrasound at moderate sound pressure levels after long-term exposure to the inner ear. From 10 kHz to 28 kHz, low frequency ultrasound below 100 dB SPL induced significant changes in cochlear microphonics, elevated thresholds and decreased maximum output voltage of action potentials and decreased absolute values of negative potentials of the endocochlear potentials.

Effects of nerve stimulation on blood flow in the urinary bladder, urethra and pelvic floor in the dog.

Pelvic organs have triple innervation from the pelvic, sympathetic and pudendal nerves. Peripheral effects can be studied by neurostimulation of the nerves, whereas the topography of the spinal cord neurons can be determined by horseradish peroxidase tracing. We have evaluated the neurophysiologic effects of the nerves at their target organs by intraoperative electric stimulation and compared the effects to the anatomical innervation areas. These zonal areas were determined by blood flow alterations, measured with nine different microspheres. Pelvic nerve stimulation showed bladder contraction

Substrate-bound GP130/F11 will promote neurite outgrowth: evidence for a cell surface receptor.

GP130/F11 is a neuronal cell adhesion molecule belonging to the Ig superfamily. Preliminary evidence suggests that it may be important for the fasciculation of axons and cell-cell interactions during synaptogenesis. Previous work has shown that when F3 (the mouse homologue) is transfected into Chinese hamster ovary (CHO) cells it will act as a substrate for neuronal growth and adhesion. In this study we have shown that isolated GP130/F11 will also act as a substrate for the growth of chick dorsal root ganglia cell neurons. The concentration of GP130/F11 on the surface of sympathetic neurons, g

Temporary threshold shift due to noise exposure in guinea pigs under emotional stress.

One of the factors which can affect the amount of temporary threshold shift (TTS) due to acoustic overstimulation is known to be the general metabolic state of the exposed subject. The present study was conducted to elucidate how preexisting intense emotional stress in awake guinea pigs could influence the TTS induced by exposure of 4 kHz at 120 dB SPL for 20 min. Considering the hypersympathetic activity in both 'stress' and 'noise' it was assumed that the two factors would act synergetically on the cochlear function. However, an unexpected result was obtained. The mean threshold for the stre

Treatment of inflammatory, neuropathic and sympathetically maintained pain in a patient with Sjögren's syndrome.

We present a case of Sjögren's syndrome with a painful peripheral neuropathy where pain control required both anti-inflammatory and sympatholytic treatments. This case suggests that the pain in some inflammatory disorders with nerve injury may have a component mediated by an alpha adrenergic receptor.

Effects of body heating and mental arithmetic on facial sweating and blood flow in unilateral migraine headache.

Facial sweating and pulse amplitude during body heating were investigated during the headache-free period in 38 subjects who suffered from unilateral migraine headache that usually recurred on the same side. In 22 of these subjects, pulse amplitude recorded from the forehead circulation was examined during a computerized mental arithmetic test. Thermally-induced increases in forehead pulse amplitude were smaller on the side habitually affected by headache than on the other side of the forehead. Although sweating from the upper part of the forehead did not differ consistently between the two si

Forskolin mediates the survival of nerve growth factor-dependent sympathetic neurons of chick embryo by a cyclic AMP-independent mechanism.

Forskolin has become an invaluable tool for exploring the involvement of cyclic AMP in a variety of cellular functions. The diterpine directly activates the catalytic subunit of adenylate cyclase, causing a marked increase in cyclic AMP content. Because of this well-characterized action, practically all the observed effects of forskolin are commonly attributed to cyclic AMP-dependent processes. We show here that forskolin exerts a neurotrophic action that is almost identical to that of nerve growth factor (NGF) and phorbol 12,13-dibutyrate (PDB) but independent of cyclic AMP. Sympathetic neuro

Norepinephrine blocks a calcium current of adult rat sympathetic neurons via an alpha 2-adrenoceptor.

The effects of alpha-adrenoceptor agonist and antagonist drugs on the Ca2+ current of acutely isolated adult rat superior cervical ganglion (SCG) neurons were investigated to characterize the adrenoceptor which mediates a catecholamine-induced decrease of the Ca2+ current. Ca2+ currents were recorded using the whole-cell variant of the patch-clamp technique from neurons isolated enzymatically from adult rat SCG. Norepinephrine (1 microM) produced a rapid, reversible, and concentration-dependent decrease in Ca2+ current amplitude and slowed the rising phase of the Ca2+ current. These effects co

Sympathetic vasoconstriction during exercise in ambulatory patients with left ventricular failure.

In patients with heart failure, exercise is thought to increase sympathetic vasoconstrictor tone. To investigate the extent of this sympathetic activation, we studied the effect of maximal exercise on nonexercising vascular beds in 35 patients with left ventricular failure (ejection fraction, 21 +/- 8%; peak exercise oxygen uptake (VO2), 12.3 +/- 3.5 ml/min/kg). In 28 patients, cardiac output and leg blood flow were measured during maximal upright bicycle exercise. Total flow to nonexercising tissue was then calculated as cardiac output--(2 x leg flow). In seven patients and six normal subject
