Parkinson's disease (PD) is classified as a synucleinopathy due to the accumulation of protein inclusions rich in the alpha-synuclein (aSyn) protein. Identifying effective pharmacological therapies is important to slow the progression and minimize the symptoms of these diseases. Cannabis sativa has a diverse chemical profile depending on its genotype, including several classes of substances, such as cannabinoids, flavonoids, terpenes, and alkaloids. In this study, we evaluated the effects of four C. sativa extracts with different phytocannabinoid chemical profiles in two cellular models that r

Candida albicans, a major human fungal pathogen undergoes filamentation from yeast to hyphal state under filamentation-inducing conditions. Gcn5 and Esa1 are key histone H3 and H4 acetyltransferases, respectively, encoded by the budding yeast and other eukaryotes. While Gcn5, a subunit of the SAGA complex, and Esa1, a subunit of the NuA4 complex, are critical for C. albicans virulence and hyphal induction, how the relative HAT activities impinge on hyphal gene expression during filamentation is less understood. We found that hyphal gene promoters are hyperacetylated at H3K9 and H4 upon filamen

The incidence of infections caused by non-albicans Candida (NAC) species, including Candida glabrata and Candida tropicalis, has recently in … 25 in C. auris and C. glabrata indicated that Erg25 is crucial for the survival of these pathogenic yeasts within the host. Furthermore, we have successfully deleted the ERG25 alleles in C. tropicalis and Candida parapsilosis, demonstrating the effectiveness of using both the CRISPR-Cas9 and Cre-loxP systems in these species for the first time.

Mass spectrometry imaging (MSI) has become a cornerstone of spatial biology research. However, various factors that are intrinsic to the technology limit the quantitative capacity of MSI-based spatial metabolomics and thus reliable interpretation. Here we developed an improved quantitative MSI workflow, based on isotopically 13C-labelled yeast extract as internal standards, to overcome these pitfalls. Using brain and kidney tissue, we demonstrate that this approach allows for quantification of more than 200 metabolic features. Applying our workflow to a stroke model allowed us to not only map

Lampreys use variable lymphocyte receptors (VLR) comprising leucine-rich-repeat (LRR) segments for antigen recognition, distinct from immunoglobulin-based receptors of jawed vertebrates. Lamprey VLRs are as diverse and antigen-specific as immunoglobulin-based antibodies, with unique advantages including high avidity, pH stability, and recognition of novel antigen epitopes. Here we describe the generation of VLR monoclonal antibody against histidine rich protein-2 (HRP-2) of Plasmodium falciparum, a causative agent of malaria. HRP-2, expressed by all parasite stages and secreted into plasma, se

Polymer-encapsulated dye nanoparticle sensors are a valuable approach to achieving in situ analyte measurements with luminescence; however, typical emulsion-based nanosensors are poorly suited for large-scale biological samples due to limitations of synthesis scalability and stability. Branched polyethylenimine (PEI) is a versatile polymer scaffold ideal for constructing nanoparticles with various covalently conjugated moieties due to their high density of reactive primary amines, high water solubility, and biological stability. In this work, we used branched polyethylenimine as a scaffold-bas

Although agriculture and forestry provide enormous lignocellulose resources, it remains to explore a novel technology for efficient biomass conversion into renewable bioethanol followed with complete recycling for valuable bioproduction with zero- waste release. As classic chemical (acid and alkali) pretreatments could cause chemical waste liberation, this study performed optimal pretreatments with hot-FeCl3 for sequentially enhancing biomass enzymatic saccharification in three bioenergy crops, which led to achieving hexoses yields raised by 4-7 times compared to the controls. By employing eng

Hard kombucha is an alcoholic beverage produced by the fermentation of a 40% (v/v) kombucha solution with selected yeast strains and sugar. Among 35 yeast strains examined, Lachancea thermotolerans, known for its high ethanol production, was selected to prepare hard kombucha using tap and deep ocean water. After 21 days of fermentation, the pH of the filtered tap water hard kombucha (TWHK) and deep ocean water hard kombucha (DOWHK) decreased to 2.6-2.69. In contrast, their titratable acidity (TA) increased to 93.68 g acetic acid/L and 107.73 g acetic acid/L, respectively. The ethanol content r

Saccharomyces cerevisiae's ability to withstand formic acid, the most impactful weak acid on yeast growth and fermentation commonly encountered in lignocellulosic bioethanol, is decisive towards industrial applications. Nevertheless, the mechanisms of formic acid resistance inS. cerevisiaeremain poorly understood. This study explored the transcriptional and metabolic responses of three strains - two resistant (YI30 and CESPLG05), and the sensitive DSM 70449 - to 4.0 g/L formic acid for defining the first model of formic acid resistance in S. cerevisiae. Resistance was linked to the upregulatio

Nuclease-helicase DNA2 is a multifunctional genome caretaker that is essential for cell proliferation in a range of organisms, from yeast to human1-4. Bi-allelic DNA2 mutations that reduce DNA2 concentrations cause a spectrum of primordial dwarfism disorders, including Seckel and Rothmund-Thomson-related syndromes5-7. By contrast, cancer cells frequently express high concentrations of DNA2 (refs. 8-11). The mechanism that precludes cell proliferation in the absence of DNA2 and the molecular aetiology of DNA2-linked diseases remain elusive. Here we used yeast and human cells to demonstrate that

Candida albicans (C. albicans) is the most common pathogen responsible for a wide spectrum of human infections ranging from superficial mucocutaneous mycoses to systemic life-threatening diseases. Its main virulence factors are the morphological transition between yeast and hyphal forms and the ability to produce biofilm. Novel antifungal strategies are required given the severity of systemic candidiasis, especially in immunocompromised patients, and the lack of effective anti-biofilm treatments. We previously demonstrated that all-trans retinoic acid (ATRA), an active metabolite of vitamin A,

Quitral (Tristerix corymbosus), a Chilean and Argentine parasitic mistletoe, is traditionally used by Mapuche natives to treat stomach ulcers, nervous disorders, and cholesterol reduction, although scientific support is scarce. Methanolic and chloroform extracts from its leaves and stems were prepared. Chemical analysis included antioxidant capacity assays (ORAC-FL and DPPH) and chromatographic determinations. The antimicrobial activity was tested against nine bacteria and two yeast strains. Additionally, cytotoxicity (hemolysis) and toxicity (against Caenorhabditis elegans) assays were perfor

Trim47, a TRIM C-VII subgroup protein characterized by a conserved SPRY domain, has been primarily studied for its ubiquitin-dependent roles in mammals. This study reports a paradigm-shifting finding in teleost immunology: grass carp Trim47 (gcTrim47) employs its SPRY domain to execute a novel, ubiquitin-independent antiviral pathway, selectively degrading GCRV-I nonstructural proteins NS38/NS80 via autophagy-mediated clearance. Unlike mammalian TRIMs, gcTrim47 antiviral activity is strictly dependent on its SPRY domain-devoid of RING/B-box domains critical for E3 ligase function-revealing an

This study characterizes the molecular epidemiology of Sporothrix species in Shanxi Province, a low-prevalence region in China, providing real data on genetic profiles and antifungal susceptibility patterns. Clinical records of sporotrichosis cases from a tertiary hospital in Shanxi (2019-2024) were retrospectively analyzed. Isolates were identified through multilocus sequencing targeting the internal transcribed spacer, calmodulin, β-tubulin, and mating-type locus genes, with phylogenetic analysis. Antifungal susceptibility testing against seven agents was performed for both yeast and mycelia

Drosophila oogenesis has long been an important model for understanding myriad cellular processes controlling development, RNA biology and patterning. Flies are easily fed drugs to disrupt various molecular pathways. However, this is often done under poor nutrient conditions that adversely affect oogenesis, thus making analysis challenging. Cycloheximide is a widely used compound that binds to and stalls the ribosome, therefore reducing protein synthesis. As egg production is a highly nutrient-dependent process, we developed a method to feed female Drosophila a rich diet of yeast paste supplem

A rise in temperature triggers a structural change in the human Type I 40 kDa heat shock protein (Hsp40/DnaJ), known as DNAJA1. This change leads to a less compact structure, characterized by an increased presence of solvent-exposed hydrophobic patches and β-sheet-rich regions. This transformation is validated by circular dichroism, thioflavin T binding, and Bis-ANS assays. The formation of this β-sheet-rich conformation, which is amplified in the absence of zinc, leads to protein aggregation. This aggregation is induced not only by high temperatures but also by low ionic strength and high pro

Strategies against the opportunistic fungal pathogen Candida albicans based on probiotic microorganisms represent a promising alternative to traditional antifungals. Here, we investigated the effects of Lactobacillaceae isolates from fermented foods or the human vagina, alone or in combination with the probiotic yeast Saccharomyces cerevisiae CNCM I-3856, against C. albicans in vitro. Nine out of nineteen tested strains of Lactobacillaceae inhibited growth of C. albicans with inhibition zones of 1-3 mm in spot assays. Five out of nineteen lactobacilli tested as such or in combination with S. c
