Doxorubicin (DOX), a frontline chemotherapeutic agent, is severely limited by reactive oxygen species (ROS) overload-mediated cardiotoxicity involving multiple programmed cell death pathways, with pyroptosis validated as one major mechanism, whereas conventional cardioprotective interventions may compromise antitumor efficacy. Here, we report a selectively redox-responsive approach based on self-crosslinked lipoic acid nanoparticles (LANPs) that enables bidirectional pyroptosis regulation in cardiac vs. tumor tissues, achieving concurrent cardioprotection and antitumor sensitization during DOX

Knee osteoarthritis (KOA) is a highly prevalent degenerative joint disease characterized by osteophyte formation at joint margins, subchondral bone sclerosis, and progressive degeneration of the articular cartilage. In advanced stages, it can result in severe functional impairment of the knee joint, imposing a substantial burden on patients' quality of life and on healthcare systems. Conventional intra-articular treatments, such as hyaluronic acid and corticosteroid injections, provide temporary pain relief but fail to achieve true tissue repair or regeneration. In recent years, several novel

<p>Background: In cancers, key prognostic determinants such as clinical stage and mutational load are well recognized. However, mucinous adenocarcinomas (MACs) display distinct biological behavior as a large proportion of their tumor volume consists of extracellular mucin, predominantly the MUC2 protein, rather than neoplastic cells. MUC2 provides a protective barrier for cancer cells against external factors, including chemotherapeutic agents and cytotoxic lymphocytes, while its anti-inflammatory and tumor-suppressive properties paradoxically support tumor persistence and immune evasion. The repair of alveolar cleft defects remains a formidable challenge in craniofacial surgery, with implications for dental arch continuity, tooth eruption, speech, and facial aesthetics. Traditional bone grafting methods (especially iliac crest autografts) have remained the gold standard, yet donor-site morbidity, graft resorption, and limitations in large defects drive the search for more advanced strategies. In recent years, developments in biomaterials, stem cell-based tissue engineering, and computer-aided surgery have opened new conceptual and practical pathways for alveolar cleft repair.

Traditional histopathological methods, such as hematoxylin and eosin staining and chromogenic immunohistochemistry, are still primarily used in clinical practice, however, they are limited in their ability to simultaneously detect multiple biomarkers and analyze spatial relationships between cell populations. These limitations are overcome by multiplex immunohistochemistry (mIHC) methods that allow detailed spatial analysis of formalin-fixed paraffin-embedded tissues with detection of multiple epitopes in a single sample. Detailed characterization of immune cell populations within tumor microe

Long noncoding RNAs (lncRNAs) have been shown to be closely related to cancer progression and therapy. However, the clinical significance of lncRNAs and the mechanisms by which they function in glioma are largely unknown. In this study, using online data sets combined with collected clinical glioma tissues, we determined that the lncRNA KB-1460A1.5 is downregulated and positively correlated with prognosis in glioma. Functional experiments showed that overexpression of KB-1460A1.5 inhibits glioma cell proliferation, migration and invasion in vitro and in vivo, while downregulation of KB-1460A1.

Defining the latent human immunodeficiency virus type 1 (HIV-1) burden in the human brain during progressive infection is limited by sample access. Human hematopoietic stem cells (hu-HSCs)-reconstituted humanized mice provide an opportunity for this study. The model mimics, in measure, HIV-1 pathophysiology, transmission, treatment, and elimination in an infected human host. However, to date, brain HIV-1 latency in hu-HSC mice during suppressive antiretroviral therapy (ART) was not studied. To address this need, hu-HSC mice were administered long acting (LA) ART 14 days after HIV-1 infection w

BACKGROUND: Infection-induced preterm birth is a major cause of neonatal mortality and morbidity and leads to preterm premature rupture of placental chorioamniotic membranes. The loss of amniotic epithelial cells and tensile strength preceding membrane rupture is poorly understood. We hypothesized that intrauterine bacterial infection induces changes in microRNA (miRNA) expression, leading to amniotic epithelial cell loss and membrane weakening.
METHODS: Ten pregnant pigtail macaques received choriodecidual inoculation of either group B Streptococcus (GBS) or saline (n = 5/group). Placental c

Dysfunctional endothelial cells contribute to the pathophysiology of many diseases, including vascular disease, stroke, hypertension, atherosclerosis, organ failure, diabetes, retinopathy, and cancer. Toward the goal of creating a new RNA-based therapy to correct aberrant endothelial cell gene expression in humans, efficient gene silencing in the endothelium of nonhuman primates was achieved by delivering small interfering RNA (siRNA) with 7C1, a low-molecular weight, ionizable polymer that forms nanoparticles. After a single intravenous administration of 1 mg of siRNA per kilogram of animal,

Bioactive agents, including proteins and peptides, can be loaded into hydrogels to improve bone regenerative capacity with their controlled release. However, the current loading method has focused on physical mixing, which has limited release control. Therefore, alternative conjugation of bioactive agents with hydrogels is highly recommended. Direct chemical conjugation of synthetic peptides containing a functional moiety with a hydrogel would be ideal. Here, we synthesized a bioactive calcium accumulating peptide (CAP) containing a collagen binding motif, which can induce osteogenic different

GOLPH2 and GOLPH3 are Golgi-related proteins associated with aggressiveness and progression of a number of cancers. Their prognostic significance in melanoma has not yet been analyzed. We performed immunohistochemical analysis for GOLPH2 and GOLPH3 in 20 normal skin, 30 benign nevi and 100 primary melanoma tissue samples and evaluated their expression in three compartments: cancer cells, tumor-associated macrophages (TAMs) and cancer-associated fibroblasts (CAFs). High levels of both proteins in melanoma cells were associated with characteristics of aggressive disease, and shorter disease-free

Specific drugs, most of them heterocyclic compounds, are leading in the therapy of parasitic infections despite their relative toxicity and potent mutagenicity. In some parasitic diseases chemotherapy is of low efficacy (hydatid and multilocular echinococcosis, African trypanosomiasis) or practically absent (American trypanosomiasis) mostly due to suppression or distortion of an immune response. Methods of immunocorrection using recombinant cytokines (interleukins, interferons) or their inducers are to be borrowed from the practice of treatment of oncological, lymphoproliferative diseases and

BACKGROUND: Metabolic diseases, including type 2 diabetes (T2D), hypertension, hyperlipidemia and metabolic dysfunction-associated fatty liver disease (MAFLD), are globally prevalent. However, traditional measures, such as body mass index (BMI), fail to capture body composition heterogeneity to accurately predict metabolic diseases. Magnetic resonance imaging (MRI) provides a precise quantification of body composition; however, the causal links among these compartments, immune phenotypes, and metabolic traits remain unclear. This Mendelian randomisation (MR) study aimed to investigate the comp

Mesenchymal stem cells (MSCs) are known to be able to modulate immune responses, possess tissue-protective properties, and exhibit healing capacities with therapeutic potential for various diseases. The ability of MSCs to secrete various cytokines and growth factors provides new insights into autoimmune-diseases such as rheumatoid arthritis (RA). RA is a systemic autoimmune disease that affects the lining of synovial joints, causing stiffness, pain, inflammation, and joint erosion. In recent years, MSCs-based therapies have been widely proposed as promising therapies in the treatment of RA. Ho

Solid organ transplantation is a life-saving procedure, yet its long-term success is limited by rejection and other pathological processes. Traditional histopathology and bulk molecular analysis lack the necessary spatial resolution to resolve the complex cellular interactions within the allograft microenvironment. The emergence of spatial-omics technologies offers new capabilities for molecular analysis within intact tissue architecture. This review provides an overview of major spatial transcriptomic and proteomic platforms, including sequencing-based technologies, imaging-based technologies

The environmental light/dark cycle has left its mark on the body's physiological functions to condition not only our inner biology, but also the interaction with external cues. In this scenario, the circadian regulation of the immune response has emerged as a critical factor in defining the host-pathogen interaction and the identification of the underlying circuitry represents a prerequisite for the development of circadian-based therapeutic strategies. The possibility to track down the circadian regulation of the immune response to a metabolic pathway would represent a unique opportunity in t

Current therapeutic drugs for ulcerative colitis (UC) remained inadequate due to drug dependence and unacceptable adverse events. Reactive oxygen species (ROS) played a critical role in the occurrence and development of UC, which most likely benefited from treatment in scavenging ROS. In this study, we developed a pH-sensitive molybdenum-based polyoxometalate (POM) nanocluster, which might contribute to site specific colonic delivery and enhance systemic efficacy of UC treatment. Our results demonstrated that POM displayed robust ROS scavenging ability in vitro. POM could significantly allevia

As a first-line drug for breast cancer chemotherapy, the effectiveness of doxorubicin (DOX) is challenged by high doses and high toxicity. Studies showed the combination of Tanshinone IIA (TSIIA) and DOX could enhance the efficacy of DOX for cancer and reduce the toxic effects to normal tissues. Unfortunately, free drugs are easily metabolized in the systemic circulation, which are less prone to aggregation at the tumor site to exert anticancer efficacy. In present study, we prepared a carboxymethyl chitosan-based hypoxia-responsive nanoparticles loaded with DOX and TSIIA for the treatment of

Intervertebral disc degeneration is the main cause of low back pain. In the past 20 years, the injection of mesenchymal stromal cells (MSCs) into the nucleus pulposus of the degenerative disc has become the main approach for the treatment of low back pain. Despite the progress made in this field, there are still many barriers to overcome. First, intervertebral disc is a highly complex load-bearing composite tissue composed of annulus fibrosus, nucleus pulposus and cartilaginous endplates. Any structural damage will change its overall biomechanical function, thereby causing progressive degenera
