ATM Variants in Breast Cancer: Implications for Breast Radiation Therapy Treatment Recommendations.

PURPOSE: Advances in germline genetic testing have led to a surge in identification of ataxia-telangiectasia mutated (ATM) variant carriers among breast cancer patients, raising numerous questions regarding use of breast radiation therapy (RT) in this population. METHODS: A literature search using PubMed identified articles assessing association(s) between the germline ATM variant status and the risk of toxicity after breast RT. An expert panel of breast radiation oncologists, genetic counselors, and basic scientists convened to review the association between ATM variants and radiation-induce

De-duplicating patient records from three independent data sources reveals the incidence of rare neuromuscular disorders in Germany.

BACKGROUND: Estimation of incidence in rare diseases is often challenging due to unspecific and incomplete coding and recording systems. Patient- and health care provider-driven data collections are held with different organizations behind firewalls to protect the privacy of patients. They tend to be fragmented, incomplete and their aggregation leads to further inaccuracies, as the duplicated records cannot easily be identified. We here report about a novel approach to evaluate the incidences of Duchenne muscular dystrophy (DMD) and spinal muscular atrophy (SMA) in Germany. METHODS: We perfor

Detection of copy number variants reveals association of cilia genes with neural tube defects.

BACKGROUND: Neural tube defects (NTDs) are one of the most common birth defects caused by a combination of genetic and environmental factors. Currently, little is known about the genetic basis of NTDs although up to 70% of human NTDs were reported to be attributed to genetic factors. Here we performed genome-wide copy number variants (CNVs) detection in a cohort of Chinese NTD patients in order to exam the potential role of CNVs in the pathogenesis of NTDs. METHODS: The genomic DNA from eighty-five NTD cases and seventy-five matched normal controls were subjected for whole genome CNVs analysi

Involvement of closely related strains of a new clonal group of Listeria monocytogenes in the 1998-99 and 2002 multistate outbreaks of foodborne listeriosis in the United States.

In 1998-99, a multistate outbreak of listeriosis in the United States was associated with contaminated hot dogs and was caused by a strain of Listeria monocytogenes serotype 4b that had been only rarely encountered before in the national PulseNet database. Upon further characterization, the strains from this outbreak were designated as Epidemic Clone II (ECII). ECII isolates exhibited diversification in a genomic region ("region 18") that was otherwise conserved among L. monocytogenes of serotype 4b. Additional unique genetic markers were identified through genome sequencing of one of the isol

Association of IL-13 Gene Polymorphism (rs20541) With Chronic Inflammatory Diseases: A Systematic Review and Meta-Analysis.

Over the years, accumulating evidence has been associating interleukin-13 gene (IL-13) variants with a wide array of chronic inflammatory diseases. Also, recent findings have associated the potential role of a single nucleotide polymorphism (SNP) of IL-13, rs20541, to promote either anti- or pro-inflammatory responses in chronic inflammatory diseases. Although rs20541 has been widely associated with various immune-related and inflammatory conditions, its precise functional relevance in the pathogenesis of human diseases has yet to be fully clarified. Nonetheless, its consistent associations an

Mapping evidence on the risk factors associated with pediatric cancers in sub-Saharan Africa: a scoping review.

BACKGROUND: The rarity and heterogeneity of pediatric cancers make it difficult to assess risk factors associated with the development of cancer in this group. This also determines the quantity and quality of evidence for etiological factors linked to pediatric cancers. Evidence on the risk factors associated with pediatric cancers is scarce; however, it has been accumulating slowly over the years. As the disease burden shifts from communicable to non-communicable diseases, most of these low- to middle-income countries (LMICs) find themselves overburdened with changing health care priorities a

Impact of interaction between individual genomes and preeclampsia on the severity of autism spectrum disorder symptoms.

OBJECTIVES: Autism spectrum disorder (ASD) is a complex neurodevelopmental disorder. Prior research suggests that genetic susceptibility and environmental exposures, such as maternal preeclampsia (PE) during pregnancy, play key roles in ASD pathogenesis. However, the specific effects of the interaction between genetic and environmental factors on ASD phenotype severity remain unclear. This study aims to investigate how interactions between de novo variants (DNVs) and common variants in individual genomes and PE exposure affect ASD symptom severity by constructing a gene-environment model. Interaction Between Age and the NOTCH3 p.R544C Variant in Determining Ischemic Stroke Risk: Insights from the Taiwan Biobank Study of 8,588 Patients.

Genetic susceptibility to ischemic stroke may vary across the life course, yet the extent to which aging modifies the effect of specific high-risk variants remains incompletely understood. The NOTCH3 rs201118034 (p.R544C) variant, a CADASIL-related mutation with relatively high prevalence in East Asian populations, has been associated with cerebrovascular disease. We investigated whether age modifies the association between the NOTCH3 p.R544C variant and ischemic stroke risk at the population level. We integrated data from the Taiwan Biobank (TWB, 2008-2020) with the National Health Insurance

Genetic characterization and whole-genome sequencing-based genetic analysis of influenza B in Shandong Province during 2015-2024.

Influenza B virus (IBV) remains a significant concern due to its role in seasonal epidemics, frequently leading to localized outbreaks characterized by high morbidity and mortality rates. This study analyzed 109 IBV strains isolated in Shandong Province using phylogenetic and amino acid variation analyses. These sequences were aligned with vaccine strains and representative isolates obtained from the GISAID influenza database. Phylogenetic trees were constructed using the maximum likelihood (ML) method in MEGA software, while amino acid substitutions were visualized using MegAlign software, an

Clinically Translatable Mutation-Based Biomarkers in Ascending Aortic Aneurysm: A Bibliometric Study.

Ascending aortic aneurysm is often clinically silent until rupture or acute dissection, and reliance on diameter-based thresholds leaves substantial risk heterogeneity that may be reduced by genetic variant-informed biomarkers. In plain language, clinically translatable variant biomarkers are genetic findings that can change diagnosis, family screening, surveillance intensity, or prophylactic surgical timing. We performed a cross-database bibliometric study to delineate the intellectual structure and thematic evolution of clinically translatable variant-based biomarker research in ascending ao

Biological Context-Informed and Population-Stratified Strategies Improve Genetic Diagnosis of CCDC22-Related Disorder.

BACKGROUND: The CCDC22 gene is a key regulator of endosomal trafficking and NF-κB signaling, and its dysfunction is implicated in a spectrum of X-linked neurodevelopmental disorders, including Ritscher-Schinzel syndrome 2, intellectual disability, and epilepsy. Despite the identification of CCDC22 variants having increased, a comprehensive biological characterization, including its spatiotemporal expression and functional molecular networks, remains to be systematically delineated. Furthermore, the rarity of CCDC22 variants created a significant challenge in distinguishing their pathogenicity.

Benchmarking and Optimization of Methods for the Detection of Identity-By-Descent in High-Recombining Plasmodium falciparum Genomes.

Genomic surveillance is crucial for identifying at-risk populations for targeted malaria control and elimination. Identity-by-descent (IBD) is increasingly being used in Plasmodium population genomics to estimate genetic relatedness, effective population size (N e ), population structure, and signals of positive selection. Despite its potential, a thorough evaluation of IBD segment detection tools for species with high recombination rates, such as P. falciparum, remains absent. Here, we perform comprehensive benchmarking of IBD callers - probabilistic (hmmIBD, isoRelate), identity-by-state-bas

Predictive and prognostic value of genetic aberrations of EGFR-TKI treatment: a real-world analysis of EGFR-mutant non-small cell lung cancer.

BACKGROUND: Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs) are the standard first-line therapy for patients with EGFR-mutant (EGFRm) non-small cell lung cancer (NSCLC). Although many patients benefit from targeted therapies, a significant proportion of patients experience suboptimal outcomes. The mechanisms underpinning this poor efficacy and the prognostic factors influencing outcomes remain insufficiently explored. This study aims to identify the genetic and clinical biomarkers that influence treatment efficacy in patients with advanced EGFRm NSCLC. METHODS: Overa

Exploring potential therapeutic targets for cardiomyopathy: A proteome-wide Mendelian randomization analysis.

Cardiomyopathy often results in heart failure and mortality, significantly impairing patients' quality of life. Advancements in genomics and proteomics now enable the identification of proteins associated with cardiomyopathy, offering valuable insights for its diagnosis and treatment. However, numerous potential pathogenic proteins remain unidentified, underscoring the need for further exploration of novel drug targets for cardiomyopathy. This study aims to employ Mendelian randomization (MR) to explore genetic associations between plasma proteins and cardiomyopathies, with the objective of id

Circulating Cytokines Mediate the Causal Relationship between Plasma Lipids and Migraine and its Subtypes: A Mediated Mendelian Randomization Study.

INTRODUCTION: Migraine, a primary headache disorder, is the third disabling disease of neurological disorders worldwide. The pathological mechanism underlying migraine remains poorly understood. Lipid metabolism may be related to migraine pathophysiology. We aimed to investigate the causal relationship between plasma lipids and migraine or its subtypes, including migraine with aura (MA) and migraine without aura (MO), and explore whether the circulating cytokines serve as mediators in the pathway from plasma lipids to migraine. METHODS: A two-step Mendelian randomization (MR) approach was use
