A disputed evidence on obesity: comparison of the effects of Rcan2(-/-) and Rps6kb1(-/-) mutations on growth and body weight in C57BL/6J mice.

It is widely accepted that body weight and adipose mass are tightly regulated by homeostatic mechanisms, in which leptin plays a critical role through hypothalamic pathways, and obesity is a result of homeostatic disorder. However, in C57BL/6J mice, we found that Rcan2 increases food intake and plays an important role in the development of age- and diet-induced obesity through a leptin-independent mechanism. RCAN2 was initially identified as a thyroid hormone (T3)-responsive gene in human fibroblasts. Expression of RCAN2 is regulated by T3 through the PI3K-Akt/PKB-mTOR-Rps6kb1 signaling pathwa

Effects of differentially fermentable carbohydrates on the microbial fermentation profile of the gastrointestinal tract of broilers.

The objective of the present study was to evaluate the effects of dietary inulin and sucrose on the fermentation profile of the gastrointestinal microflora in chicken. Day-old broilers (n = 80) were assigned to four dietary treatments, either fed a basal diet or the same diet supplemented with sucrose (4%), inulin (1%) or sucrose and inulin. At day 35, birds were killed and pH, lactate, ammonia, short chain fatty acids (SCFA) and biogenic amines were determined in different parts of the digestive tract. Final body weights and the relative weights of liver, pancreas, crop, gizzard and small int

Evaluating dietary starch concentrations in reduced-forage rations fed to lactating dairy cattle.

Increasing frequency of severe weather events such as droughts and floods may reduce the inventory of high-quality forage available for feeding dairy cattle. Thus, optimizing rumen health and milk production when feeding reduced-forage rations is critical for dairy farmers and nutritionists. Our experiment investigated the effects of replacing NDF with starch in diets with 12.6% forage NDF on rumen metabolism, rumen inflammation, feed intake, and milk production. We hypothesized that increasing dietary starch would increase inflammatory cytokines and alter T lymphocytes in rumen tissue. We als

Transcriptomic Analyses Reveal Intestine and Liver Responses of Large Yellow Croaker (Larimichthys crocea) to Plant Protein Diet.

The utilization of plant protein in aquafeeds to reduce dependence on fishmeal has become an important strategy for improving the sustainability of marine aquaculture. This study investigated the effects of low-fishmeal plant protein diets on growth performance and gut-liver molecular responses in large yellow croaker. A total of 2,048 fish were fed two diets for 206 days: a commercial-like control diet containing 280 g/kg fishmeal (C group) and a low-fishmeal diet containing 100 g/kg fishmeal with increased inclusion of plant protein sources (P group). Growth trials indicated that fish in the

Brain-wide activity mapping reveals the somatosensory cortex as a sex-specific regulator of hedonic feeding.

Palatable food consumption engages reward circuitry and promotes neuroadaptations that contribute to overeating and obesity. While mesolimbic dopamine pathways are central to hedonic feeding models, the contribution of sensory cortical systems remains poorly understood. Here, we performed brain-wide activity mapping using Targeted Recombination in Active Populations (TRAP) and network analysis to define the distributed neural consequences of short-term high-fat diet (HFD) exposure in male and female mice. HFD increased caloric intake in both sexes, with females consuming significantly more tha

Immunorestorative effects of Limosilactobacillus reuteri LR6 in protein-energy malnourished mice under pathogen and allergen challenge.

Protein-energy malnutrition (PEM) is the most severe form of malnutrition, often linked with impaired immunity and increased susceptibility to infections. In this study, mice were initially fed a normal semi-synthetic well-nourished diet for one week and then divided into five groups. The animals in Group C (control) continued on the normal well-nourished diet, while Groups M (malnourished), SM (skim milk), PFM (probiotic fermented milk), and BS (probiotic suspension) received a protein- and energy-restricted diet (2% casein, 315.2 kcal) until PEM was established. Later, the groups SM, PFM and

Rifaximin Protects Against Inflammation and Fibrosis in MASH: Any Role for Ethanol-Producing Bacteria?

Metabolic Dysfunction-Associated Steato-Hepatitis (MASH) is a multiple-hit disease. Endotoxins, ethanol, and other metabolites of certain gut microbiota can reach the liver and accelerate inflammation and disease progression. Targeting ethanol-producing colonic bacteria with rifaximin could affect the progress of MASH. In the present study, thirty mice were assigned to three groups (n = 10 mice per group). Mice received either a normal diet, a Western diet, or a Western diet with oral rifaximin. After 12 weeks, liver function, serum levels of TNF-α, interleukin (IL)-1β, IL-6, and lipopolysacch

Microbiota-derived 4-HPAA alleviates Crohn's disease by stabilizing SIRT1 and reprogramming macrophage immunometabolism.

Crohn's disease (CD) is characterized by mucosal immune dysregulation, gut microbiota disturbance, and epithelial barrier dysfunction. This study investigated whether an Aronia berry polyphenol-rich diet attenuates CD-related intestinal inflammation through microbiota-derived metabolic signals. IL-10-/- and TNBS-induced colitis mouse models were used to evaluate the effects of dietary intervention, bacterial strain supplementation, and candidate metabolite administration. Integrated metagenomic, metabolomic, and transcriptomic analyses were combined with in vivo and in vitro mechanistic experi

Long-Term Fructose Intake Induces Moderate Liver Inflammation but Does not Overlap with the Detrimental Effects of the Ketogenic Diet on Hepatic Steatosis in Rats.

An excess of fat and carbohydrate exerts a negative role on liver metabolism, favouring the development of metabolic dysfunction-associated steatotic liver disease (MASLD). However, it is not clear which component of the diet is more detrimental to liver homeostasis. We assessed the long-term effect of high fructose and the ketogenic diet on hepatic steatosis. For this purpose, 18 adult male Wistar rats were randomized into three groups: Control Diet (CD), consisting of commercial chow; Fructose Diet (FD), which included commercial chow and water with 30% fructose; and Ketogenic Diet (KD), com

Arachidonic acid is involved in high-salt diet-induced coronary remodeling through stimulation of the IRE1α/XBP1s/RUNX2/OPN signaling cascade.

BACKGROUND: The impact of a high-salt (HS) diet on metabolic disturbances in individuals with coronary heart disease remains unclear. The arachidonic acid (AA) metabolic pathway is closely linked to the development of cardiometabolic diseases and atherosclerotic cardiovascular diseases. Furthermore, endoplasmic reticulum stress (ERS) has emerged as a major contributor to cardiometabolic diseases. AA-related inflammation and ERS are hypothesized to play a role in HS diet-induced coronary remodeling. METHODS: Rats were subjected to an HS diet for 4 weeks, and the serum concentration of AA was m

Combined dietary omega-3 and omega-6 fatty acids protect against hyperglycemia-associated retinopathy in neonatal mice.

Retinopathy of prematurity (ROP) with early vessel loss (Phase I) followed by uncontrolled vessel growth (Phase II) causes visual impairment in premature infants. Although supplementation with omega-3 (n-3) docosahexaenoic acid (DHA) alone shows mixed results in preventing ROP, supplementation with both n-3 DHA and n-6 arachidonic acid (ARA) in early postnatal life reduces severe ROP by 50 % (Mega Donna Mega study). In the Mega Donna Mega study, 146 (72.6 %) of 201 included infants had at least one hyperglycemic episode during the first 14 days of life, which is a strong ROP risk factor. We th

The therapeutic potential of a polyunsaturated fatty acid-enriched high-fat diet in Leigh syndrome: Insights from a preclinical model.

INTRODUCTION: Leigh syndrome is often caused by Ndufs4 mutations. The Ndufs4 knockout (KO) mouse model recapitulates key disease features, including systemic inflammation, neurodegeneration, and motor deficits. While dietary interventions such as the ketogenic diet show promise in mitigating mitochondrial dysfunction, conflicting results highlight uncertainties regarding its efficacy. Here, we evaluate the therapeutic potential of a polyunsaturated fatty acid (PUFA)-enriched high-fat diet (HFD) in Ndufs4 KO mice. METHODS: Dietary intervention began at postnatal day 23, with mice receiving eit

Ameliorating effect of probiotic on nonalcoholic fatty liver disease and lipolytic gene expression in rabbits.

Nonalcoholic fatty liver disease (NAFLD) is a condition that affects about 24% of people worldwide. Increased liver fat, inflammation, and, in the most severe cases, cell death are all characteristics of NAFLD. However, NAFLD pathogenesis and therapy are still not clear enough. Thus, this study aimed to determine the effect of a high-cholesterol diet (HCD) inducing NAFLD on lipolytic gene expression, liver function, lipid profile, and antioxidant enzymes in rabbits and the modulatory effects of probiotic Lactobacillus acidophilus (L. acidophilus) on it.
