Effects of sequential aeromedical evacuations following traumatic brain injury in swine.

INTRODUCTION: Traumatic brain injuries (TBI) represent a significant percentage of critical injuries in military conflicts. Following injury, wounded warfighters are often subjected to multiple aeromedical evacuations (AE) and associated hypobaria, yet the impact in TBI patients remains to be characterized. This study evaluated the impact of two consecutive simulated AEs in a fluid-percussion TBI model in swine to characterize these effects. METHODS: Following instrumentation, anesthetized Yorkshire swine underwent a frontal TBI via fluid-percussion. A hypobaric chamber was then used to simul

Risk Factors for Infection and Revision Surgery following Patellar Tendon and Quadriceps Tendon Repairs: An Analysis of 3,442 Patients.

Disruption of the extensor mechanism is debilitating with surgical repair being the accepted treatment. The incidence of infection and reoperation after extensor mechanism repair are not well reported in the literature. Thus, the objective of the current study was to (1) determine the incidence of surgical site infection and reoperation within 1 year of primary extensor mechanism repair and (2) identify independent risk factors for infection and reoperation following patellar and quadriceps tendon repair. A retrospective review of the 100% Medicare Standard Analytic files from 2005 to 2014 was

Effect of mineralocorticoid receptor blockade on the renal renin-angiotensin system in Dahl salt-sensitive hypertensive rats.

BACKGROUND: The (pro)renin receptor exists in the kidney, blood vessels and the heart. (Pro)renin binds to the receptor and induces tissue injuries directly, completely independent of angiotensin II (Ang II). The renal renin-angiotensin-aldosterone system is activated in salt-sensitive hypertensive rats with in-vitro studies showing aldosterone increases angiotensin-converting enzyme (ACE) activity, renin production and angiotensin II type 1 receptor (AT1R) activity. However, the effect of blockade of mineralocorticoid receptor on the renal (pro)renin receptor, angiotensinogen, ACE and AT1R in

CKD screening: urinary albumin or uDKK3 or both?

Approximately 750 million people worldwide suffer from chronic kidney diseases (CKD)-a leading cause of cardiovascular morbidity and premature mortality-but early identification of progressive kidney function decline in patients at risk is still a challenge. CKD diagnosis relies on estimated glomerular filtration rate (eGFR) and albuminuria screening, primarily reflecting glomerular injury. Nevertheless, in a substantial proportion of affected subjects progressive CKD initially remains undetected, since kidney function deteriorates through ongoing tubulointerstitial injury without significant

MiR-199a-5p aggravates hypoxia/reoxygenation-induced cardiomyocyte ferroptosis by blocking HSPB1-Keap1/Nrf2/ARE signaling.

Ferroptosis plays a crucial role in hypoxia/reoxygenation (H/R)-induced cardiomyocyte injury and acute myocardial infarction (AMI), yet the involvement of microRNA-199a-5p (miR-199a-5p) in this process remains insufficiently understood. In this study, serum miR-199a-5p levels were markedly elevated in AMI patients and positively correlated with myocardial injury markers cardiac troponin I and creatine kinase-MB, while H/R stimulation similarly upregulated miR-199a-5p expression in AC16 cardiomyocytes. Functional experiments demonstrated that miR-199a-5p overexpression exacerbated oxidative str

Inhaled Halogen-Induced Oxidative Renal Damage and Dysfunction: A Lung Heart Kidney Axis.

Acute exposure to halogen gases causes extensive injury to the lungs and heart that may be fatal. To evaluate secondary renal complications subsequent to pulmonary and cardiac dysfunction, rats were exposed to bromine and chlorine, and their renal function and injury biomarkers were assessed post exposure. Bromine or chlorine caused a significant increase in arterial blood creatinine and urea nitrogen (BUN) suggesting acute renal stress. Rats exposed to either of these halogens also exhibited increased total protein, albumin, and retinol binding protein 4 (RBP4) in the urine indicating signifi

Soluble Epoxide Hydrolase Inhibitors Improve Cornea Alkali Wound Healing.

Cornea alkali injuries are difficult to treat and potentially blinding, and there are still no specific treatments. The cytochrome P450 arachidonic acid metabolism pathway generates epoxy fatty acids (EpFAs) which have analgesic, anti-inflammatory, and anti-fibrotic activities. EpFAs have anti-fibrotic effects in several tissues including kidney, heart, liver, and lungs, while the cornea has never been examined. Soluble epoxide hydrolase (sEH) metabolizes EpFAs to biologically less active diols, and its inhibitors (sEHi) can alleviate or prevent fibrosis. The current studies were designed to e

CXCL5-CXCR2 axis mediates myocardial ischemia/reperfusion injury by inducing infiltration of myeloid-derived macrophages and neutrophils.

BACKGROUND: Myeloid cell-associated immune responses play a critical role in the pathogenesis of myocardial ischemia/reperfusion (MI/R) injury. Chemokine receptor CXCR2 is expressed mainly in myeloid cells, including neutrophils and macrophages, and is crucial for regulating immune homeostasis. However, the roles of CXCL5 and its receptor CXCR2 in MI/R injury remain unknown. METHODS: To determine the functional significance of CXCL5-CXCR2 axis in MI/R injury, we employed genetic deletion of CXCL5, myeloid-specific deletion of CXCR2, and pharmacological blockade of CXCR2. Cardiac function was

LGR6 protects against myocardial ischemia-reperfusion injury via suppressing necroptosis.

Regulated necrosis (necroptosis) and apoptosis are important biological features of ischemia-reperfusion (I/R) injury. However, the molecular mechanisms underlying myocardial necroptosis remain elusive. Leucine rich repeat containing G protein-coupled receptor 6 (LGR6) has been reported to play important roles in various cardiovascular disease. In this study, we aimed to determine whether LGR6 suppresses I/R-induced myocardial necroptosis and the underlying molecular mechanisms. We generated LGR6 knockout mice and used ligation of left anterior descending coronary artery to produce an in vivo

MKK3 depletion attenuates intestinal injury after traumatic hemorrhagic shock by restoring mitochondrial function.

BACKGROUND: Traumatic hemorrhagic shock (THS) is a complex pathophysiological process resulting in multiple organ failure. Intestinal barrier dysfunction is one of the mechanisms implicated in multiple organ failure. The present study aimed to explore the regulatory role of mitogen-activated protein kinase kinase 3 (MKK3) in THS-induced intestinal injury and to elucidate its potential mechanism. METHODS: Rats were subjected to trauma and hemorrhage to establish a THS animal model. MKK3-targeted lentiviral vectors were injected via the tail vein 72 h before modeling. Twelve hours post-modeling

Paradoxical breathing during sleep is associated with increased sleep apnea and reduced ventilatory capacities in high-level spinal cord injury.

Sleep-disordered breathing is highly prevalent in individuals with high-level spinal cord injury. In addition, chest mechanics are known to be altered, leading to paradoxical breathing. Here we investigated the interaction between paradoxical breathing and sleep quality in these patients, and its association with measurements of respiratory function, hypercapnic ventilatory response and peak exercise ventilation. Home-based polysomnography was performed in 13 patients with spinal cord injury (C4 to T4) untreated for sleep-disordered breathing. We defined paradoxical breathing as counterphase b

METTL3, m6A modification, and EGR1: interplay affecting myocardial I/R injury outcomes.

The occurrence of severe myocardial ischemia/reperfusion (I/R) injury is associated with the clinical application of reestablishment technique for heart disease, and understanding its underlying mechanisms is currently an urgent issue. Prior investigations have demonstrated the potential enhancement of MIRI through EGR1 suppression, although the precise underlying regulatory pathways require further elucidation. The core focus of this investigation is to examine the molecular pathways through EGR1 regulates mitophagy-mediated myocardial cell pyroptosis and its impact on MIRI. Cardiomyocyte hyp

Bioactive Compounds from Germinated Brown Rice Protect Cardiomyocytes Against Simulated Ischemic/Reperfusion Injury by Ameliorating Mitochondrial Dysfunction.

PURPOSE: Ischemic/reperfusion (I/R) injury is the principal mechanism during Ischemic Heart Disease (IHD). The key modulator of I/R injury is dysregulation of mitochondria function. Germinated Brown Rice (GBR) has been recommended as a bio-functional food and has clarified the potential properties in several effects. However, the effect of GBR mediated cardioprotective properties, focusing on mitochondrial function's role, remains unexplored. Thus, this study aims to investigate the cardioprotective effects of GBR pretreatment against simulated I/R injury. METHODS: H9c2 cardiomyocytes were in

Oxoeicosanoid receptor inhibition alleviates acute myocardial infarction through activation of BCAT1.

5-Oxo-6,8,11,14-eicosatetraenoic acid (5-oxo-ETE) is an arachidonic acid metabolite produced along with leukotrienes via the 5-lipoxygenase pathway. Metabolomics studies have shown that 5-oxo-ETE level is elevated in the serum in acute myocardial infarction (AMI). The actions of 5-oxo-ETE are mediated by the highly selective oxoeicosanoid receptor (OXE-R). Moreover, increased OXE-R content was verified in AMI patients and mice. However, the precise role of OXE-R in AMI is unclear. In the present study, we demonstrate that 5-oxo-ETE triggered myocardial injury in mice. Pathway enrichment analys

Hydrogen gas inhalation alleviates myocardial ischemia-reperfusion injury by the inhibition of oxidative stress and NLRP3-mediated pyroptosis in rats.

AIMS: Reperfusion therapy is the most common and effective treatment against ischemic heart disease (IHD), but the process inflicts massive ischemia/reperfusion (I/R) injury for which no treatment exists. Notably, reperfusion after ischemia causes ischemia/reperfusion injury (IR injury) and the "no-reflow" phenomenon seriously affecting the therapeutic effects in clinical practice. The principle purpose of this study is to validate the effect of hydrogen gas on IHD and further explore the mechanism of hydrogen gas in alleviating myocardial I/R injury and no-reflow phenomenon.
