Lurbinectedin for extensive stage - small cell lung cancer (ES-SCLC): real world response patterns and survival outcomes.

BACKGROUND: Small cell lung cancer (SCLC) remains a therapeutic challenge with limited treatment options and poor survival outcomes. Lurbinectedin has been approved for the post-platinum setting, but there is a lack of real-world evidence and information related to its optimal sequencing and effectiveness across different lines of therapy. This study aimed to analyze line-dependent outcomes and response patterns of lurbinectedin in SCLC patients. METHODS: In this retrospective multi-center study analysis, we enrolled SCLC patients treated with lurbinectedin between January 2020 and December 2

Factors associated with viral load suppression among children and adolescents on dolutegravir-based antiretroviral regimen in Tanzania: a longitudinal analysis.

INTRODUCTION: Antiretroviral therapy (ART) reduces morbidity and mortality due to human immunodeficiency virus (HIV) infection. In 2019, Tanzania adopted Dolutegravir (DTG) as a first-, second-line, and third-line treatment for children and adolescents living with HIV (CALHIV). Viral load suppression (VLS) is desirable in the prevention of HIV transmission thus achieving the third '95' target. DTG treatment has highly potent antiviral activity, a high genetic barrier to resistance, and a high safety profile. We aimed to determine VLS and associated factors among CALHIV on DTG-based ART in Tanz

Facilitating access to direct-acting antivirals in a community-based point-of-diagnosis model for hepatitis C treatment: The role of the pharmacy team in the no one waits (NOW) study.

BACKGROUND: Despite simplified hepatitis C virus (HCV) treatment algorithms, insurance-related barriers prevent same-day HCV treatment upon diagnosis in the US. We assessed how direct partnerships with a pharmacy team facilitated HCV treatment initiation among socially marginalized populations in a community setting. METHODS: The No One Waits (NOW) Study, a single-arm trial conducted between July 1, 2020, and October 31, 2021, in San Francisco, CA, targeted individuals experiencing homelessness, injecting drugs, and eligible for simplified HCV treatment. Upon positive HCV RNA results, partici

Novel analytical strategy integrating enzymatic hydrolysis with UPLC-MS/MS for precise quantification of free and bound heterocyclic aromatic amines in crispy fried pork.

The discovery of bound heterocyclic amines (HAAs) has led to the realization that the HAAs content in meat may have been underestimated before, so the determination of bound HAAs content in meat products has become important. In this method, methanol-hydrochloric acid solution was selected as the extraction solution of free HAAs in crispy fried pork, and then the bound HAAs in meat was extracted by innovative enzymatic hydrolysis method. The optimal extraction process parameters of bound HAAs were determined by single-factor experiment combined with response surface method. Under the optimal e

Denutrition status prevails over a standard AML risk assessment in older adults.

Older adults with acute myeloid leukemia (AML) have a poor prognosis because frailty and the characteristics of the disease limit the use of intensive chemotherapy (ICT). Treatment with 5-azacitidine (5-AZA) or low-dose cytarabine (Cytarabine) (LDAC) - with or without venetoclax - is currently recommended in this setting. However, we lack real-life data on response rates and treatment outcomes. We conducted a retrospective, multicenter registry study of 279 older adults with AML (median [interquartile range (IQR)] age: 76 [70-81]) having undergone first-line treatment with LDAC (n = 87) or 5-A

Erythroid-predominant myelodysplastic neoplasms exhibit a distinct genomic landscape with poor outcomes after venetoclax-based therapy.

Erythroid differentiation confers BCL-XL dependence and venetoclax resistance in acute myeloid leukemia (AML). However, whether myelodysplastic neoplasms (MDS) with erythroid predominance (EP), defined by ≥50% erythroid bone marrow elements, have distinct biology and drug sensitivities remains unknown. To study this, we evaluated an MDS patient cohort (n = 371) and showed that EP MDS (n = 67, 18%) are characterized by higher TP53 (multihit TP53: 36% vs 17%, p = 0.004), BCOR and WT1 and lower ASXL1 and SRSF2 mutation frequencies compared to non-EP (NEP) MDS (n = 304, 82%). Anti-tumor efficacy of RAF/MEK inhibitor VS6766 in KRAS-mutated colorectal cancer cells.

PURPOSE: Colorectal cancer (CRC) ranks third among most prevalent cancers worldwide. KRAS is the most frequently (30-40%) mutated oncogene in CRC, which has been defined as an "undruggable" therapeutic target over the past four decades. METHODS: In this study, we applied four HT-29 cell lines, namely HT-29-wild-type (HT-29-WT), point-mutated HT-29-KRASG12V, HT-29-KRASG12C and HT-29-KRASG12D, in order to detect the efficiency of RAF-MEK inhibitor VS6766, the BRAFV600E inhibitor PLX4720 was selected as the control. The analyses of in vitro cytotoxicity, cell cycle and apoptosis of HT-29 cell li

SALL2-Mediated Suppression of WNT Signaling Through Transcriptional Control of AXIN2 in Colorectal Cancer Cells.

Colorectal cancer (CRC) remains the second leading cause of cancer-related mortality worldwide, with aberrant activation of the Wnt/β-catenin signaling pathway constituting a key driver of tumorigenesis. SALL2, a zinc finger transcription factor deregulated in various cancers, has been implicated in Wnt signaling regulation through its Xenopus ortholog; however, its role in human CRC remains unclear. In this study, we investigated the expression and function of SALL2 in CRC. Immunohistochemical analysis revealed that SALL2 is present in the epithelium and stroma of normal colon tissue but is s

Successful Dose Reduction of Janus-kinase Inhibitors in Well-controlled Atopic Dermatitis: A Prospective, Multicentre, Real-world Study.

Janus kinase 1 (JAK1) inhibitors are effective for atopic dermatitis, but carry risks of adverse events. Dose reduction may mitigate these risks, although real-world evidence of dose reduction strategies is limited. This prospective, real-world study investigated JAK1 inhibitor dose reduction among 60 adults with controlled atopic dermatitis treated with abrocitinib or upadacitinib for ≥ 3 months. Doses were halved or administered every other day, as capsules cannot be split. Patients could resume their original dose if needed. The primary outcome was the proportion of patients maintaining the

Precision prediction of venetoclax-azacitidine treatment efficacy in acute myeloid leukemia via integrative drug screening and machine learning.

Venetoclax-azacitidine (VEN/AZA) has transformed acute myeloid leukemia (AML) therapy, yet reliable predictors of response remain lacking. We employ a multidisciplinary strategy combining ex vivo drug sensitivity testing, transcriptomic profiling, functional assays, and clinical data to identify determinants of VEN/AZA response. Core genes consistently associated with responsiveness are validated through CRISPR-Cas9 screening, with silencing of BCL2L1 and PINK1 preferentially enhancing drug sensitivity. Building on these insights, we develop and validate an eight-gene random forest model (RF8)

NAMPT inhibition uncovers therapeutic vulnerabilities to venetoclax and chemotherapy in acute myelogenous leukemia.

Acute myeloid leukemia (AML) cells depend on nicotinamide adenine dinucleotide (NAD+) biosynthesis via nicotinamide phosphoribosyltransferase (NAMPT) for survival. Single-cell RNA sequencing revealed robust NAMPT expression across diverse AML subtypes. Proteomic profiling showed that NAMPT inhibition with KPT-9274 induced adaptive upregulation of BCL2, an anti-apoptotic protein, highlighting a survival mechanism. BH3 profiling confirmed that AML cells hierarchically depend on BCL2, followed by MCL1 and BCLxL, for survival. Combining KPT9274 with the BCL2 inhibitor venetoclax synergistically en

Engineering a Radiohybrid PSMA Ligand with an Albumin-Binding Moiety and Pharmacokinetic Modulation via an Albumin-Binding Competitor for Radiotheranostics.

The prostate-specific membrane antigen (PSMA) is a well-established target for radiotheranostics in prostate cancer. We previously demonstrated that 4-(p-astatophenyl)butyric acid (APBA), an albumin-binding moiety (ABM) labeled with astatine-211 (211At), enables the modulation of pharmacokinetics and enhancement of therapeutic efficacy when combined with the post-administration of an albumin-binding competitor. However, this strategy has not been explored in PSMA-targeting ligands. We designed and synthesized [211At]6, a novel PSMA ligand structurally analogous to PSMA-617 with APBA. The compo
