The role of corticotrophin-releasing factor in stress-induced relapse to alcohol-seeking behavior in rats.

RATIONALE: Intermittent footshock stress reliably reinstates extinguished alcohol-taking behavior in drug-free rats, but the neurochemical events involved in this effect are not known. OBJECTIVE: We studied here whether two main modulators of stress responses, corticotropin-releasing factor (CRF) and corticosterone, are involved in reinstatement of alcohol seeking induced by the intermittent footshock stressor. METHDOS: Rats were given alcohol in a two-bottle choice procedure (water versus alcohol) for 30 days and were then trained for 60 min per day to press a lever for alcohol (12% w/v) for

Selective blockade of the mineralocorticoid receptor impairs hypothalamic-pituitary-adrenal axis expression of habituation.

The present study investigated the role of mineralocorticoid receptors (MR) and glucocorticoid receptors (GR) in the expression of habituation of the hypothalamic-pituitary-adrenal (HPA) axis response to stress. Male rats were restrained for 1 h per day for six consecutive days. On day 6, 1 h prior to restraint stress, both restraint-naive and repeatedly restrained rats were injected s.c. with either vehicle (propylene glycol) or one of three corticosteroid receptor antagonist treatments: selective MR antagonist (RU28318 or spironolactone), selective GR antagonist (RU40555), or both MR and GR

A unique hormonal and behavioral hyporesponsivity to both forced novelty and d-amphetamine in periadolescent mice.

The identification of critical ontogenetic periods of increased vulnerability to the effects of drugs of abuse could have a great psychobiological and clinical-therapeutical importance. Potential age-related differences in the response of the hypothalamic-pituitary-adrenal (HPA) axis to both stress and psychostimulants has been tested here in an animal model of adolescence. Periadolescent (PND 33-43) and Adult (PND>60) mice of both sexes were injected with d-amphetamine (AMPH, 0, 2, or 10 mg/kg i.p.) and immediately faced with a mild psychological stress experience, i.e. placement in a novel e

Long-term intracerebroventricular corticotropin-releasing hormone administration induces distinct changes in rat splenocyte activation and cytokine expression.

The effects of long-term corticotropin-releasing hormone (CRH) infusion in the lateral ventricle of the rat on hypothalamic-pituitary-adrenocortical (HPA) axis parameters and on the immune system function were studied. Compared with infusion of vehicle, the CRH treatment produced a sustained overactivity of the HPA axis, as evidenced by elevated plasma ACTH and corticosterone levels, increased anterior pituitary POMC messenger RNA (mRNA) expression, and adrenal enlargement. Long-term CRH treatment also inhibited body weight gain and reduced thymus and spleen weight. In the CRH-treated animals,

Corticotropin-releasing hormone and its receptor distribution in fetal membranes and placenta of the rhesus monkey in late gestation and labor.

Maternal plasma corticotropin-Releasing Hormone (CRH) rises from midgestation to term and increases further during labor in pregnant women. The primate placenta contains both the CRH peptide and its gene and is the likely source of circulating CRH. In the present study, we examined CRH messenger RNA (mRNA) and peptide expression in fetal membranes and the placenta of 14 pregnant rhesus monkeys (140-161 days gestational age) using Northern blot analysis, in situ hybridization, and immunocytochemistry to define the cellular distribution of CRH during the last third of pregnancy and in relation t

Extrapituitary gonadotropin-releasing hormone (GnRH) binding sites in goldfish.

In teleosts, as in other vertebrates, the secretion of pituitary gonadotropin (GTH) is mediated by the hypothalamic decapeptide, gonadotropin-releasing hormone (GnRH). Recent findings in teleosts indicate that GnRH receptors are not restricted to the pituitary gonadotropes and are also associated with somatotropes as well as being present in a number of other tissues. In the present study, we provide novel information on GnRH binding in a number of extrapituitary tissues in goldfish. However, we do not intend to provide full characterization of GnRH binding sites in various extrapituitary tiss

Function of the growth hormone-insulin-like growth factor I axis in the profoundly growth-retarded diabetic child: evidence for defective target organ responsiveness in the Mauriac syndrome.

Mauriac syndrome (MS) consists of a triad of poorly controlled diabetes, profound growth retardation, and hepatomegaly. The mechanisms involved in the growth retardation of those patients are not well understood. In an attempt to determine whether the growth retardation was secondary to somatroph secretory failure, abnormal pulsatile secretion, deletion of the growth hormone (GH) receptor, inadequate insulin-like growth factor I (IGF-I) generation, or abnormal IGF-I binding proteins (IGFBPs) two patients with MS were studied and their results compared with those of age-matched diabetic boys of

Blood pressure and renal function in a novel vasopressin-deficient, genetically hypertensive rat strain.

1. Hereditary hypothalamic diabetes insipidus was introduced into the New Zealand genetically hypertensive (NZGH) rat and its normotensive substrain (NZN) by cross-breeding males with female Brattleboro diabetes insipidus (DI) rats. 2. Selective breeding of the resultant DI/hypertensive (DI/H) rats on the basis of maximum systolic blood pressure and vasopressin deficiency produced animals in the F6 generation with blood pressures at 10 weeks of age higher than in DI/normotensive rats (DI/N), but much lower than in age-matched NZGH animals. Age-matched NZN and DI/N rats had comparable blood pre

AA in Obesity and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) Nonalcoholic Fatty Liver Disease (NAFLD).

Obesity and MASLD (NAFLD) are chronic, low-grade systemic inflammatory diseases resulting from complex interactions among diet, adipose tissue, gut microbiota, liver, muscle, pancreas, and the brain. Central to this integrative framework are AA and other polyunsaturated fatty acids (PUFAs), which regulate inflammation, insulin sensitivity, neuroendocrine signaling, and energy homeostasis.Excess consumption of energy-dense foods rich in saturated and trans fats leads to adipocyte hypertrophy, altered membrane fluidity, increased antigenicity, and enhanced recruitment of macrophages and T cells

Serotonergic Toxicity and Hyponatremia Associated with Antidepressant Therapy: The Pivotal Role of SIADH.

Serotonin syndrome (SS) and the syndrome of inappropriate antidiuretic hormone secretion (SIADH) are well-recognized adverse drug reactions (ADRs) associated with serotonergic agents, particularly antidepressants. Although traditionally regarded as distinct clinical entities, accumulating evidence suggests that SIADH may represent a key pathophysiological link between serotonergic toxicity and acute hyponatremia, thereby explaining their frequent clinical overlap and, in some cases, severe or fatal outcomes. Excessive serotonergic activity can stimulate hypothalamic pathways regulating arginin

The NeuroImmunoEndocrine Circuit of Umami Peptides: A Systems Biology Approach.

Background/Objectives: Umami peptides enhance flavor and contribute to appetite regulation (satiety) and metabolic health. By signaling to the orbitofrontal cortex, umami has been shown to improve cognitive function in Alzheimer's disease dementia. This taste boosts the immune system and induces saliva secretion. However, the molecular mechanisms linking umami peptides to systemic physiology remain poorly understood. This study provides the first integrated analysis of neurological, immunological, and endocrinological pathways activated by umami peptides. Methods: Novel umami peptides were ide

Oxytocin as a neurotherapeutic target for nicotine withdrawal and craving modulation.

Nicotine addiction is a chronic relapsing disorder driven by neuroadaptive changes in reward, stress, and emotional regulation systems. Withdrawal symptoms, including anxiety, irritability, cravings, and anhedonia, significantly hinder smoking cessation efforts. Despite current treatment options such as nicotine replacement therapy, relapse rates remain high. Traditionally, nicotine's reinforcing properties have been attributed to its action on nicotinic acetylcholine receptors (nAChRs) within the mesolimbic dopamine system, particularly the ventral tegmental area (VTA) and nucleus accumbens (

Modern Determinants of Earlier Menarche and Mental Health: A Translational Review for Clinicians.

Background: Over the past decades, the global onset of menarche has progressively advanced, driven by complex interactions between metabolic, psychosocial, and environmental factors. Beyond its reproductive significance, menarche represents a critical developmental milestone that coincides with increased vulnerability to mental health difficulties. Contemporary determinants such as childhood obesity, early-life adversity, and exposure to endocrine-disrupting chemicals (EDCs) may accelerate pubertal timing while simultaneously shaping adverse psychological trajectories during adolescence. Thyroid dysfunction and Research Domain Criteria framework-based depression phenotypes: Insights from a historical longitudinal cohort.

BACKGROUND: Depression is a clinically heterogeneous disorder, making it difficult to identify its biological underpinnings, including potential links to dysregulation of the hypothalamic-pituitary-thyroid axis, which plays a central role in the pathophysiology of depression. The Research Domain Criteria (RDoC) framework provides a multi-dimensional approach to parsing depressive phenotypes. This study investigates the association between thyroid-stimulating hormone (TSH) abnormalities and depressive symptoms mapped to RDoC domains. METHODS: In a retrospective cohort from 2000 to 2021, adults

Beyond Self-Reports: Integrating Cortisol Measurement in Psychotherapy Process Research among Adolescents with Borderline Personality Pathology.

UNLABELLED: <p>Introduction: Psychotherapy is the primary treatment for adolescent borderline personality pathology (BPP), yet its mechanisms remain unclear. Given potential self-report biases due to alexithymia and impaired interoception, this study examined cortisol responses as a physiological stress marker alongside session ratings from adolescent BPP patients and their therapists to assess its potential as a complementary measure in psychotherapy process research. METHODS: N = 56 adolescents (94.6% female) with BPP (≥3 DSM-IV BPD criteria) receiving Adolescent Identity Treatment or Diale

Melatonin Alleviates Circadian Rhythm Disruption-Induced Enhanced Luteinizing Hormone Pulse Frequency and Ovarian Dysfunction.

Circadian rhythm disruption (CRD), stemming from sleep disorders and/or shift work, is a risk factor for reproductive dysfunction. CRD has been reported to disturb nocturnal melatonin signaling, which plays a crucial role in female reproduction as a circadian regulator and an antioxidant. The hypothalamic-pituitary-ovarian (HPO) axis regulates female reproduction, with luteinizing hormone (LH) pulse pattern playing a pivotal role in folliculogenesis and steroidogenesis. However, the effect of CRD on the HPO axis and the involvement of melatonin remains unclear. Female CBA/CaJ mice underwent CR

Limitations of hormonal and clinical markers in predicting GnRH agonist trigger success in polycystic ovary syndrome: A critical reappraisal.

BACKGROUND: Polycystic ovary syndrome (PCOS) is associated with altered hypothalamic-pituitary-ovarian function, which may affect the success of GnRH agonist triggers used during IVF. Identifying reliable predictors of oocyte yield in these patients remains a clinical challenge. PURPOSE: To evaluate possible predictors of suboptimal oocyte retrieval per aspirated follicle when ovulation is triggered with GnRH agonist in PCOS patients. METHODS: Between 30/04/2021 and 30/12/2022, a prospective cohort study was conducted among women with PCOS (n = 104) in which a GnRH agonist trigger was employ

Optimal functioning of hypothalamic-pituitary-adrenal axis after cessation of prednisolone therapy: a cross-sectional study in children with nephrotic syndrome.

BACKGROUND: Hypothalamic-pituitary-adrenal (HPA) axis recovery after cessation of steroid therapy in children with nephrotic syndrome (NS) has hardly been studied in the literature. METHODS: This 22-month cross-sectional study recruited children (2-14 years) with NS, having received a minimum 3 months of prednisolone, now in remission, and off steroids for 1, 3, or 6 months. Serum cortisol-basal and stimulated (with long-acting intramuscular adrenocorticotropic hormone), and factors affecting them, were assessed. Low basal and stimulated cortisol were taken as < 138 nmol/L and < 500 nmol/L, r

Anxiolytic and Antidepressant Effects of Organic Polysulfide, Dimethyl Trisulfide Are Partly Mediated by the Transient Receptor Potential Ankyrin 1 Ion Channel in Mice.

Background/Objectives: Dimethyl trisulfide (DMTS) is a naturally occurring polysulfide with known antioxidant and neuroprotective properties. DMTS is a lipophilic transient receptor potential ankyrin 1 (TRPA1) ligand that reaches the central nervous system (CNS). Its role in the CNS, particularly regarding depression-like behaviour, has yet to be explored. This study investigates the influence of DMTS on stress responses and whether this effect is mediated through the TRPA1 ion channel, known for its role in stress adaptation. Using a mouse model involving three-week exposure, we examined the
