Estrogen Modulates the Sensitivity of Lung Vagal C Fibers in Female Rats Exposed to Intermittent Hypoxia.

Obstructive sleep apnea is mainly characterized by intermittent hypoxia (IH), which is associated with hyperreactive airway diseases and lung inflammation. Sensitization of lung vagal C fibers (LVCFs) induced by inflammatory mediators may play a central role in the pathogenesis of airway hypersensitivity. In females, estrogen interferes with inflammatory signaling pathways that may modulate airway hyperreactivity. In this study, we investigated the effects of IH on the reflex and afferent responses of LVCFs to chemical stimulants and lung inflammation in adult female rats, as well as the role

Immune responsive resolvin D1 programs myocardial infarction-induced cardiorenal syndrome in heart failure.

Resolvins are innate, immune responsive, bioactive mediators generated after myocardial infarction (MI) to resolve inflammation. The MI-induced bidirectional interaction between progressive left ventricle (LV) remodeling and kidney dysfunction is known to advance cardiorenal syndrome (CRS). Whether resolvins limit MI-induced cardiorenal inflammation is unclear. Thus, to define the role of exogenous resolvin D (RvD)-1 in post-MI CRS, we subjected 8- to 12-wk-old male C57BL/6 mice to coronary artery ligation. RvD1 was injected 3 h after MI. MI mice with no treatment served as MI controls (d 1 an

Inflammation and chronic colonization of Haemophilus influenzae in sputum in COPD patients related to the degree of emphysema and bronchiectasis in high-resolution computed tomography.

The presence of bacteria in the lower airways in COPD results in inflammation, further airway structural damage, and might lead to repeated exacerbations. We have previously shown that chronic colonization of Haemophilus influenzae during stable disease is related to increased inflammation, and we now aimed to relate previous findings of bacterial colonization and inflammation to the degree of radiological findings of bronchiectasis and emphysema. Thirty-nine patients with COPD were included in their stable state, and a high-resolution computed tomography of the lung was performed. They were f

Apolipoprotein E deficiency and high-fat diet cooperate to trigger lipidosis and inflammation in the lung via the toll-like receptor 4 pathway.

Apolipoprotein E deficiency (ApoE(-/-)) combined with a high-fat Western-type diet (WD) is known to activate the toll-like receptor (TLR4) p … by increased alveolar septal thickness and mean linear intercept as well as elevated production of inflammation mediators (at 12 weeks), and by granuloma formation (at 24 weeks). The expression levels of TLR4, myeloid differentiation primary response 88 (MyD88) and nuclear factor kappa B were markedly upregulated in ApoE(-/-) WD mice at week 12. However, these effects …

A mouse model for pathogen-induced chronic inflammation at local and systemic sites.

Chronic inflammation is a major driver of pathological tissue damage and a unifying characteristic of many chronic diseases in humans including neoplastic, autoimmune, and chronic inflammatory diseases. Emerging evidence implicates pathogen-induced chronic inflammation in the development and progression of chronic diseases with a wide variety of clinical manifestations. Due to the complex and multifactorial etiology of chronic disease, designing experiments for proof of causality and the establishment of mechanistic links is nearly impossible in humans. An advantage of using animal models is t

The "genomic storm" induced by bacterial endotoxin is calmed by a nuclear transport modifier that attenuates localized and systemic inflammation.

Lipopolysaccharide (LPS) is a potent microbial virulence factor that can trigger production of proinflammatory mediators involved in the pathogenesis of localized and systemic inflammation. Importantly, the role of nuclear transport of stress responsive transcription factors in this LPS-generated "genomic storm" remains largely undefined. We developed a new nuclear transport modifier (NTM) peptide, cell-penetrating cSN50.1, which targets nuclear transport shuttles importin α5 and importin β1, to analyze its effect in LPS-induced localized (acute lung injury) and systemic (lethal endotoxic shoc

Spontaneous eosinophilic nasal inflammation in a genetically-mutant mouse: comparative study with an allergic inflammation model.

BACKGROUND: Eosinophilic inflammation is a hallmark of chronic rhinosinusitis with nasal polyps. To model this disease process experimentally, nasal sensitization of mice with ovalbumin or aspergillus has been described. Here, we describe a genetically mutant mouse that develops robust spontaneous nasal eosinophilic inflammation. These mice lack the enzyme SHP-1 that down-regulates the IL-4Rα/stat6 signaling pathway. We compared nasal inflammation and inflammatory mediators in SHP-1 deficient mice (mev) and an ovalbumin-induced nasal allergy model. METHODS: A novel technique of trans-pharynge

P2X Antagonists Inhibit HIV-1 Productive Infection and Inflammatory Cytokines Interleukin-10 (IL-10) and IL-1β in a Human Tonsil Explant Model.

HIV-1 causes a persistent infection of the immune system that is associated with chronic comorbidities. The mechanisms that underlie this inflammation are poorly understood. Emerging literature has implicated proinflammatory purinergic receptors and downstream signaling mediators in HIV-1 infection. This study probed whether inhibitors of purinergic receptors would reduce HIV-1 infection and HIV-1-stimulated inflammation. An ex vivo human tonsil histoculture infection model was developed to support HIV-1 productive infection and stimulated the inflammatory cytokine interleukin-1 beta (IL-1β) a

CD11c+ CD8 T cells cause IFN-γ-dependent autoimmune neuroinflammation that is restrained by PD-1 signaling.

In multiple sclerosis (MS) lesions, CD8 T cells outnumber CD4 T cells, suggesting that they contribute to MS pathology. However, little is known about the role of CD8 T cells in MS, partly due to the prevalent use of experimental autoimmune encephalomyelitis (EAE) models mediated by CD4 T cells, which have limited involvement of CD8 T cells. Importantly, MS and EAE differ in both their distribution of CNS lesions and neurologic deficits, indicating differences in CNS inflammation. MS lesions are more commonly found in the brain, whereas EAE lesions are more frequent in the spinal cord. Additio

Toll-like receptor 4-mediated regulation of spontaneous Helicobacter-dependent colitis in IL-10-deficient mice.

BACKGROUND & AIMS: The commensal microbiota is believed to have an important role in regulating immune responsiveness and preventing intestinal inflammation. Intestinal microbes produce signals that regulate inflammation via Toll-like receptor (TLR) signaling, but the mechanisms of this process are poorly understood. We investigated the role of the anti-inflammatory cytokine interleukin (IL)-10 in this signaling pathway using a mouse model of colitis. METHODS: Clinical, histopathologic, and functional parameters of intestinal inflammation were evaluated in TLR4(-/-), IL-10(-/-), and TLR4(-/-)

Two differing presentations of periocular dermatitis as a side effect of dupilumab for atopic dermatitis.

Dupilumab is a monoclonal antibody used to treat atopic dermatitis. Worsening of atopic dermatitis and conjunctivitis following dupilumab use are reported adverse effects; however, there is little reported on the nature and mechanism of these complications. Here, we describe two patients with chronic atopic dermatitis who developed new or severely worsened periocular dermatitis, believed to be a side effect of dupilumab injections, and resolution after its discontinuation. We explore the possibility of dupilumab-induced suppression of Th2 mediated inflammation and upregulation of Th1 and IFNγ

Improvement in HIV-related endothelial dysfunction using the anti-inflammatory agent salsalate: a pilot study.

We hypothesized that heightened systemic inflammation contributes to the increased rate of cardiovascular events in HIV-infected patients not receiving combination antiretroviral therapy. We performed a pilot trial to assess the effects of the nuclear factor-kappaB inhibitor salsalate on flow-mediated dilation of the brachial artery, a measure of endothelial function. Flow-mediated dilation significantly improved after 8 weeks of salsalate. However, hepatotoxicity occurred frequently. Research using alternative agents is warranted to examine the role of inflammation in HIV-related cardiovascul

Immune-Mediated Uveitis and Lifestyle Factors: A Review.

Uveitis is a leading cause of sight-threatening disease worldwide and is characterized by inflammation of the uvea, retina, and optic nerve. Among the forms of uveitis, immune-mediated uveitis represents the majority of cases in developed countries, while infectious uveitis is more common in developing countries. A Western lifestyle is thought to be one of the factors contributing to the difference and is responsible for the increasing incidence of immune-mediated uveitis. A vast range of studies have reported the importance of lifestyle factors, including smoking, gut microbiome, diet, and ph

Preoperative platelet-related inflammatory burden is associated with delayed early hemodynamic recovery following minimally invasive cardiac surgery.

BACKGROUND: Minimally invasive cardiac surgery (MICS) is associated with reduced surgical trauma and improved postoperative recovery; however, early hemodynamic recovery remains variable and may be influenced by patient-related biological factors. This study evaluated the association between preoperative inflammatory biomarkers and delayed early postoperative recovery following MICS. METHODS: This retrospective observational study included 166 consecutive patients undergoing thoracotomy-based MICS at a single tertiary center between January 2020 and January 2026. Inflammation may explain part of atherosclerotic cardiovascular disease risk conferred by elevated remnant lipoproteins: a cohort and Mendelian randomization study.

BACKGROUND AND AIMS: Elevated remnant lipoproteins are a causal risk factor for atherosclerotic cardiovascular disease. We tested the hypothesis that low-grade inflammation can explain part of the increased risk of peripheral artery disease (PAD) and myocardial infarction conferred by elevated remnant lipoproteins; we also explored the causal effect of genetically elevated remnant cholesterol on inflammatory cytokine levels. METHODS: In 90,789 individuals from the Copenhagen General Population Study (2003-2015), we estimated the fraction of the association from elevated remnant cholesterol to

Exploring the impact of pan-immune-inflammation-value and other traditional inflammation-based markers on coronary collateral circulation in patients with chronic total occlusion.

BACKGROUND: Coronary collateral circulation (CCC) plays a pivotal role in mitigating the adverse effects of chronic total occlusion (CTO) by providing an alternative blood supply to ischemic myocardium. In recent years, there has been a growing interest in understanding the relationship between inflammation and coronary collateral development. However, the predictive role of pan immune inflammation value (PIV), a new and expanded inflammatory response marker, on CCC has not yet been investigated. This article aims to provide a detailed comparison of the effect of PIV and other traditional infl

Systemic inflammatory and metabolic indexes for aortic dissection: a machine learning analysis.

BACKGROUND: Inflammation plays a crucial role in the development of aortic dissection. The systemic immune-inflammation index (SII) and systemic inflammation response index (SIRI) are widely used surrogate markers for systemic inflammation, while the neutrophil to high-density lipoprotein cholesterol ratio (NHR), platelet to high-density lipoprotein cholesterol ratio (PHR), lymphocyte to high-density lipoprotein cholesterol ratio (LHR), and monocyte to high-density lipoprotein cholesterol ratio (MHR) incorporate high-density lipoprotein, reflecting both inflammatory and metabolic status. Effect of perioperative repetitive transcranial magnetic stimulation on postoperative cognitive function and peripheral inflammation in elderly total knee arthroplasty patients: study protocol for a randomized controlled trial.

BACKGROUND: Postoperative cognitive dysfunction (POCD) is a serious and common complication after total knee arthroplasty (TKA) in the elderly. Studies have suggested that repetitive transcranial magnetic stimulation (rTMS) can reduce the levels of tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and interleukin-6 (IL-6) inflammatory factors in the hippocampus, inhibit neuroinflammatory responses in the brain, and reduce the damage to synapses, thereby improving cognitive dysfunction. However, the effectiveness of rTMS for POCD remains to be explored. Therefore, the aim of this
