Investigational drugs in PTSD.

Post-traumatic stress disorder (PTSD) remains one of psychiatry's most challenging disorders, common, disabling, and biologically complex. Despite decades of research, only sertraline and paroxetine are FDA-approved, both with modest efficacy. Even first-line trauma-focused psychotherapies leave up to half of patients with persistent symptoms. However, advances in neurobiology are reframing PTSD as a disorder of maladaptive stress circuitry, neuroplasticity, and memory reconsolidation, opening new therapeutic possibilities. This comprehensive review examines current PTSD pharmacotherapy, emerg

Methyl divanillate enhances doxorubicin efficacy in TNBC cell lines through redox imbalance and apoptotic reprogramming.

Triple-negative breast cancer (TNBC) represents one of the most aggressive breast cancer subtypes, lacking effective targeted therapies and relying primarily on chemotherapy. In this study, we investigated the cytotoxic and molecular effects of doxorubicin (DOX) and methyl divanillate (DMV), alone or in combination, in three-dimensional spheroid models derived from MDA-MB-231 (claudin-low) and HCC70 (basal-like) TNBC cell lines. Combined DOX/DMV treatment significantly reduced spheroid viability and was associated with increased reactive oxygen species (ROS) accumulation and thiol depletion, t

Prenatal-lactational alcohol exposure induces sex-specific CX3CL1/CX3CR1 dysregulation linked to neuroendocrine imbalance and cardiovascular risk.

Fetal alcohol spectrum disorder is associated with lasting neurodevelopmental and cardiovascular dysfunctions. The fractalkine axis CX3CL1/CX3CR1, a chemokine and its sole known receptor expressed in microglia and myeloid/endothelial cells, coordinates neuroimmune and vascular responses. We tested whether prenatal-lactational alcohol exposure (PLAE) is associated with sex-specific dysregulation of this axis along with integrated behavioral, neuroendocrine, inflammatory, and cardiovascular signatures. Pregnant C57BL/6 dams consumed 20% ethanol using a drinking-in-the-dark (DID) paradigm through

Lipid dysregulation in renal cancer: Drivers of tumor growth and determinants of treatment-induced toxicity.

BACKGROUND: Clear cell renal cell carcinoma (ccRCC) exhibits a distinctive metabolic signature marked by the excessive buildup of cholesterol and neutral lipids. This phenotype stems largely from the loss of the von Hippel-Lindau (VHL) tumor suppressor and the resulting stabilization of hypoxia-inducible factors (HIFs). The altered lipid environment enables ccRCC cells to sustain growth, evade immune surveillance, and withstand contemporary systemic therapies. Increasing evidence indicates that similar disturbances in lipid metabolism contribute to kidney injury during cancer treatment, sugges

ROS-responsive nanodiscs for STING-NF-κB pathway inhibition and glycosaminoglycan layer restoration in interstitial cystitis/bladder pain syndrome therapy.

Interstitial cystitis/bladder pain syndrome (IC/BPS) is a chronic, refractory bladder disorder characterized by urothelial barrier disruption, chronic inflammation, and oxidative stress. Generally, the deficiency of the glycosaminoglycan (GAG) layer constitutes a core pathological feature of IC/BPS. Although bladder instillation with chondroitin sulfate (CS), a sulfated polysaccharide and a member of the GAG family, has been employed as a conventional replenishment therapy, the clinical efficacy remains limited. This is primarily attributed to its poor mucosal adhesion and retention, rapid cle

Numerical Simulation of Failure Modes of Solid Propellants with Internal Cavities Under Various Loading Conditions.

The reliability of solid rocket motors depends primarily on the structural integrity of their propellants. Internal cavity defects in the widely used hydroxyl-terminated polybutadiene (HTPB) propellant, formed during manufacturing and service, significantly degrade its mechanical properties and compromise motor safety. This study developed a constitutive model for HTPB propellant based on the generalized incremental stress-strain damage model (GISSMO). The validity of the constitutive model was verified through uniaxial tensile tests conducted at various tensile rates. Based on this constituti

Functional mechanism of SptP from Salmonella Typhimurium in modulating environmental adaptability, intracellular proliferation and intestinal inflammation in Wenchang chickens.

Salmonella enterica subsp. enterica serovar Typhimurium (S.Typhimurium) is a critical foodborne zoonotic pathogen that impairs poultry production and threatens public health. The virulence effector SptP encoded by Salmonella Pathogenicity Island 1 (SPI-1) exerts unclear pathogenic functions in indigenous Wenchang chickens, a high-susceptible local broiler breed in China. Here, wild-type (WT) S. Typhimurium and an isogenic ΔSptP knockout mutant were combined with in vitro stress assays, HD11 chicken macrophage infection, in vivo oral challenge of Wenchang chicks, and ileal RNA sequencing (RNA-s

Tanshinone-IIA Mediated Attenuation of Post-Infarction Myocardial Fibrosis via IL-33/ST-2 Signaling Changes in Rat Epicardial Adipose Tissue.

Myocardial infarction (MI), a severe form of coronary artery disease, often results in myocardial fibrosis, a key contributor to cardiac dysfunction and heart failure. Epicardial adipose tissue (EAT) is increasingly implicated in cardiovascular pathology through cytokine-mediated modulation of cardiac function. Tanshinone IIA (TanIIA), a lipophilic compound derived from Salvia miltiorrhiza, exhibits anti-inflammatory, antioxidant, and anti-fibrotic properties. This study aimed to evaluate the therapeutic potential of TanIIA in attenuating post-MI myocardial fibrosis and to explore its underlyi

Proteomic Profiling of Cryopreservation-Induced Alterations in Rhesus Macaque Sperm.

BACKGROUND: Sperm cryopreservation is a fundamental technique in assisted reproductive technologies. While human sperm freezing is clinically routine, preserving nonhuman primate sperm remains challenging due to species-specific characteristics and low post-thaw viability. OBJECTIVES: This study aims to investigate the proteomic changes in rhesus macaque sperm before and after cryopreservation, focusing on identifying the molecular mechanisms behind cryoinjury. MATERIALS AND METHODS: Semen was collected from six sexually mature male rhesus macaques (6-10 years) using rectal electroejaculatio

Ambient Temperature Shapes Skeletal Muscle Growth and Fiber-Type Plasticity in Mice.

Skeletal muscle development and physiological homeostasis are profoundly influenced by environmental cues. Among these factors, ambient temperature represents a critical determinant of growth performance and metabolic adaptation in mammals. However, the effects of different ambient temperature ranges on skeletal muscle characteristics and on responses across multiple visceral tissues remain poorly understood. In this study, five ambient temperature conditions (16 °C, 20 °C, 24 °C, 28 °C, and 32 °C) were established to investigate their physiological impacts in a mouse model. Our results demons

Impact of renal lipid droplet dynamics on tubular health.

Lipid droplets (LDs) are dynamic organelles that regulate lipid storage, metabolism, and organelle crosstalk. In renal tubular cells, which rely heavily on fatty acid oxidation for energy, LDs play a dual role. The transient accumulation of LDs during acute stress may protect against excess fatty acid accumulation and reduce lipotoxicity, but persistent LD overload contributes to mitochondrial dysfunction, oxidative stress, inflammation, and fibrotic remodeling. This review synthesizes recent advances in LD biology in the kidney, highlighting mechanisms of biogenesis, turnover via lipolysis an

FABP4 as an immunometabolic hub in preeclampsia: from maternal-fetal interface to systemic inflammation.

Preeclampsia (PE) is a multisystem vascular disease that occurs specifically during pregnancy, and the understanding of its pathogenesis is gradually shifting from the traditional "placental ischemia-endothelial injury" model to immunometabolic disorders. In this article, we propose a conceptual framework positioning fatty acid binding protein 4 (FABP4) as an immunometabolic hub in the pathogenesis of PE. Clinical studies suggest that FABP4 is significantly elevated in preeclamptic placentas and maternal circulation; epigenetic derepression via miR-148a/152-mediated DNMT1 downregulation contri

Emerging biomodels to understand the pathophysiology of sepsis and evaluate new therapeutic strategies targeting extracellular histones.

BACKGROUND: Extracellular histones (extH) have emerged as key damage-associated molecular patterns (DAMPs) driving multi-organ failure in sepsis. Despite their correlation with disease severity, organ-specific mechanisms of extH toxicity and targeted therapeutic strategies remain underexplored. MAIN BODY: This review dissects extH-mediated pathophysiology across vital organs including heart, lungs, kidneys, liver, and brain, through convergent pathways including TLR activation, NLRP3 inflammasome signaling, NETosis, oxidative stress, calcium influx, pyroptosis, and microvascular thrombosis. Vitamin D and L-Cysteine as Potential Regulators of Adiponectin in Alzheimer's Disease: A Narrative Review.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder marked by amyloid-β buildup, tau pathology, neuroinflammation, and declining cognition. Adiponectin, a hormone produced by adipose tissue with insulin-sensitizing, anti-inflammatory, and antioxidant effects, may link peripheral metabolic health to brain function. Studies show that adiponectin helps neurons survive, improves synaptic plasticity, maintains blood-brain barrier integrity, and reduces amyloid-β and tau damage via AdipoR1/R2 signaling. Clinical studies evaluating circulating adiponectin have yielded inconsistent fi

Respiratory health outcomes following co-exposure to subchronic high temperature, episodic wildfire smoke exposure, and high-cholesterol diet.

BACKGROUND: Increased ambient temperature alongside concomitant heatwaves and wildfire smoke imperil human health. We examined whether long-term episodic exposure to wildfire smoke induced greater respiratory injury/inflammation in male rats subchronically housed at elevated temperature or consuming unhealthy, cholesterol-rich diets. Respiratory effects of wildfire eucalyptus smoke exposure (WFES; ∼7mg/m3 particulate matter) were assessed 2h and 24h post-exposure in rats housed at 22 °C (room temperature; RT) consuming normal diet (ND) and exposed to filtered air (FA) or WFES for 1h. Proposal for a new definition of health: an anthropological reappraisal based on a cross-sectional survey.

BACKGROUND: The definition of health proposed by the World Health Organization (WHO) in 1948 ("a state of complete physical, mental and social well-being and not merely the absence of disease or infirmity") remains influential but increasingly criticized for its idealistic, static, and potentially unattainable nature. Contemporary societies are confronted with aging populations, chronic diseases, environmental disruptions, psychosocial stress, and changing perceptions of normality and pathology. This study aimed to explore current perceptions of disease and health among French-speaking respond
