The effect of tobacco smoke on the metabolism and function of rat alveolar macrophages.

Alveolar macrophages harvested by bronchopulmonary lavage from rats exposed to tobacco smoke for 30 days ("smokers") showed alterations in oxidative metabolism, lactate production and phagocytosis of inert starch particles when compared with control macrophages. Phagocytosis of viable Staphylococcus aureus was unaffected by tobacco smoke. Glucose oxidation measured by conversion of glucose-1-14C to 14CO2 moderately affected while oxidation of glucose-6-14C to 14CO2 was not. Smokers routinely yielded fewer cells than controls, though these cells contained approximately 17% more protein than did

Ethylene formation by polymorphonuclear leukocytes. Role of myeloperoxidase.

Ethylene formation from the thioethers, beta-methylthiopropionaldehyde (methional) and 2-keto-4-thiomethylbutyric acid by phagocytosing polymorphonuclear leukocytes (PMNs) was found to be largely dependent on myeloperoxidase (MPO). Conversion was less than 10% of normal when MPO-deficient PMNs were employed; formation by normal PMNs was inhibited by the peroxidase inhibitors, azide, and cyanide, and a model system consisting of MPO, H2O2, chloride (or bromide) and EDTA was found which shared many of the properties of the predominant PMN system. MPO-independent mechanisms of ethylene formation

Neurotoxicogenomic impact of 4-nonylphenol on Heteropneustes fossilis via molecular, histopathological and bioinformatic analysis.

Exposure to 4-Nonylphenol at concentrations of 64 µg/L (low) and 160 µg/L (high) for 30 to 60 days, spanning the pre-spawning to spawning phases in H. fossilis, induces significant adverse effects in the brain, particularly within the telencephalon and cerebellum regions. Additionally, protein content in brain decreases in treated group of 4-Nonylphenol (4-NP), brain acetylcholinesterase (AChE) activity decreases in a manner contingent upon the amount of dose and exposure period. In the brain tissue of H. fossilis, exposure to 4-Nonylphenol (4-NP) results in a dose-dependent decrement of antio

A Phase-Resolved Geometric Deep Learning Framework Maps Structural Determinants of Disease-Associated Protein Aggregation and Guides Suppressor Design.

Protein aggregation drives major neurodegenerative diseases, yet most computational predictors collapse assembly into static risk scores and do not resolve the distinct structural determinants of nucleation and elongation. Here, we present SKALE 2.0, a phase-resolved geometric deep learning framework that represents proteins as multimodal structural graphs and learns mutation-induced aggregation phenotypes directly from three-dimensional topology. Across SOD1, TDP-43, MAPT, and PRNP, SKALE 2.0 recovered a conserved latent transition from nucleation to elongation while resolving distinct mutati

Optimization of ultrasound-assisted deep eutectic solvents extraction, enrichment, and anti-diabetic of glycosides from Anemarrhena asphodeloides.

Steroidal glycosides and xanthone C-glycosides in Anemarrhena asphodeloides have anti-diabetic, antifungal, anti-inflammatory, anti-platelet aggregation, antioxidant and anticancer properties. This study employed a choline chloride/methylurea deep eutectic solvent (DES) for sustainable and efficient extraction of these glycosides. Optimal conditions-64% water content, 29 mL/g solvent-to-solid ratio, and 42 min extraction time-were established using single-factor and Box-Behnken design, yielding 118.51 ± 0.39 mg/g total glycosides, a 2.34-fold increase over ethanol extraction. Subsequent enrich

L-Proline protects human sperm from heat stress-induced oxidative damage.

L-proline, an amino acid with antioxidant properties, has demonstrated significant potential in counteracting oxidative stress and preserving sperm quality. This study evaluated the protective effects of L-proline against heat stress-induced damage during sperm preparation incubation, a critical step in assisted reproductive technologies (ART). Semen samples from 30 normozoospermic men were divided into two groups: a control group and a treatment group supplemented with 2 mmol/L L-proline. Following initial incubation at 37 °C for 30 min, samples were further subdivided and subjected to contro

Detection and cross-organ characterization of physiological response to microplastic stress in Panax ginseng based on hyperspectral imaging assisted with machine learning.

Microplastic pollution can affect growth and quality of medicinal plants, yet rapid detection of microplastic stress responses remains underexplored. We treated ginseng with polyethylene microplastics, acquired leaf hyperspectral images (HSI) on day 23, and constructed machine learning models for identifying stress levels and predicting physiological indicators. Furthermore, the applicability of successive projections algorithm (SPA) and competitive adaptive reweighted sampling (CARS) for characteristic wavelength selection was compared. Results showed that polyethylene stress significantly af

Cellular redox disruption and apoptosis: Differential effects of RFR frequencies on Leydig cells.

Radiofrequency radiation (RFR), widely emitted from modern wireless devices, has raised questions regarding its possible impact on male reproductive health. In this comparative study, we examined the redox and apoptotic responses of TM3 Leydig cells following exposure to mobile phone radiation, as well as 2450 MHz, and 1800 MHz frequencies for 15, 30, 45, 60, 90 & 120 min, and redox imbalance was assessed by quantifying nitric oxide (NO) and intracellular superoxide (SO) levels. Apoptotic cell percentages were evaluated by dual labeling with Annexin V-FITC/PI using flow cytometry. Mobile phone

Bacterial Contribution to Hydrogen Peroxide Decomposition in Lake Erie Cyanobacterial Harmful Algal Bloom Communities Is Dependent on Bloom Stage.

Cyanobacterial harmful algal blooms (cHABs) are expanding globally and often involve toxin-producing species such as Microcystis aeruginosa that pose ecological and public health risks. Some cyanobacteria, including some strains of M. aeruginosa, lack antioxidant enzymes such as catalase, making them more reliant on associated bacteria for reactive oxygen species (ROS) decomposition. To evaluate bacterial contribution to oxidative stress defence in Microcystis blooms across stages of bloom development, we conducted a time-series incubation after a pulse of elevated hydrogen peroxide spanning t

Behavioral and redox effects of lithium and curcumin in a planarian model of sublethal stress.

Freshwater ecosystems are increasingly contaminated by neuroactive compounds, including lithium (Li) from battery waste and curcumin (CUR) from consumer products, but their sublethal effects on aquatic invertebrates are poorly characterized. Linking behavioral alterations to biochemical responses under sublethal exposure remains a challenge. We used Girardia tigrina to assess behavioral and redox responses to mechanical, photic, and predator odor stress, followed by Li or CUR treatment. Mechanical and predator-odor stress increased time in the illuminated compartment by 40-60% (p < 0.0001), wh

Mechanistic insights into profenofos interactions with antioxidant enzymes in Labeo rohita.

Profenofos pesticide (Organophosphate) is one of the most commonly used pesticide products. It has been detected in many freshwater ecosystems and can impact the survival of non-target organisms. Its exposure causes oxidative stress and compromises the ability of fish to regulate its defense mechanisms. While there is significant interest in understanding the effects of profenofos on fish, the exact molecular mechanisms are not yet fully understood. This study is focused on determining the interactions of profenofos with key enzymes of the antioxidant defense system of Labeo rohita (freshwater

1-MCP pretreatment combined with SO2 preservation pad delays postharvest senescence and softening of 'Shine Muscat' grapes by regulating cell-wall metabolism and antioxidant activity.

Quality deterioration of 'Shine Muscat' grapes involves tissue softening, aroma loss, and flavor alteration. This study investigated the effects of single and combined treatments of 1-methylcyclopropene (1-MCP) and preservation pads (SO2-pad) on physiological activities and quality changes in grapes during storage. The combined treatment exhibited delayed quality deterioration, with maintained fruit color, reduced respiration rate and weight loss. Moreover, the combined treatment best preserved characteristic terpenoid aroma compounds such as linalool while inhibiting off-flavor formation. Fur

A multifaceted insight into dinuclear salicylatocopper(II) complexes containing 2,2'-bipyridine: Syntheses, structural characterization, spectral properties and biological activity.

The present study reports the syntheses, structural characterization, and biological evaluation of five dinuclear copper(II) complexes containing substituted salicylato ligands and 2,2'-bipyridine as a neutral N,N´-donor ligand: [Cu₂(bpy)2(3-F-Sal)2] (1), [Cu₂(bpy)2(4-Cl-Sal)2] (2), [Cu₂(bpy)2(3-Cl-Sal)2] (3), [Cu₂(bpy)2(4-MeO-Sal)2] (4), and [Cu₂(bpy)2(3,5-I-Sal)2] (5a). This work demonstrates that variations in preparation methods can lead to solid-state structural variability. Single-crystal XRD diffraction and complementary spectroscopic methods revealed a conserved CuN₂O₃ coordination env

Drosophila: An Emerging New Approach Method (NAM) for Studying Amyotrophic Lateral Sclerosis (ALS).

Drosophila melanogaster (D. melanogaster), or fruit flies, are a commonly used model organism in the study of neurodegenerative diseases (NDs). Their short lifespan, low cost, genetic tractability, and conserved signaling and developmental pathways make them ideal for studying NDs and associated biochemical pathways. Further, flies offer the advantage of high-throughput exploratory drug and genetic screening without stringent ethical constraints. Therefore, D. melanogaster serves as an ideal organism for preliminary drug screening before transitioning to toxicity and efficacy studies in verteb
