[Post-thyrotoxic arterial hypertension].

The paper is concerned with the results of a study of the state of central hemodynamics, sympathetic-adrenal and renin-angiotensin systems to reveal their role in the development of postthyrotoxic arterial hypertension. Altogether 147 patients with diffuse toxic goiter before therapy and 114 patients after surgery (subtotal subfascial thyroidectomy after O. V. Nikolaev or its modification) were investigated. In 35 (30.7%) of 114 operated on patients BP values in the long-term postoperative period were above 160/95 mm Hg. Hemodynamic indices and the state of the vasoactive system were correlate

Effects of beta-agonists on b- and c-waves implicit for adrenergic mechanisms in cat retina.

Nylidrin (buphenine) is a beta-adrenergic agonist known to dilate peripheral vessels and used therapeutically in retinal degeneration and glaucoma. We studied retinal function under beta-agonists in arterially perfused cat eyes and observed a dose-dependent, reversible increase in b-wave amplitude and a decrease in c-wave amplitude in concentrations from 4.5 to 120 microM. A half maximal response was obtained at 40 to 50 microM. The optic nerve response to light showed dose-dependent reversible changes under nylidrin. Standing potential, light peak, intraocular pressure, vascular resistance, a

Centrally induced vasopressor responses to ouabain in DOCA-salt hypertensive rats.

To investigate the effect of inhibition of cerebral Na+, K+-ATPase on cardiovascular regulation ouabain was injected into the lateral ventricle or the posterior hypothalamus in either conscious or urethane anaesthetised, deoxycorticosterone-salt hypertensive (DOCA) and sham operated (sham) rats. Ouabain injected intracerebroventricularly produced dose dependent vasopressor responses and tachycardia in the conscious rat; the magnitude of the pressor response was consistently larger in DOCA than in sham rats. In anaesthetised rats the pressor responses were accompanied by corresponding increases

Barbiturate inhibition of endothelium-dependent dilatation of blood- and Krebs-perfused rat tail arteries.

Rat tail artery segments were cannulated both ends, immersed in an organ bath filled with Krebs solution, and perfused at a constant 3 ml/min with Krebs solution from a reservoir or arterial blood from a donor conscious rat. Donor rats had chronic indwelling exteriorized silastic cannulas in a carotid artery and jugular vein. Blood withdrawn from the carotid artery returned to the donor rat via the jugular vein cannula after perfusing the tail artery segment. Arteries were constricted and perfusion pressure increased by infusing noradrenaline into the perfusate. In Krebs perfused arteries vaso

Hypothalamic beta 2-adrenoceptor control of renal sympathetic nerve activity and urinary sodium excretion in conscious, spontaneously hypertensive rats.

The contributions of beta 1-, B2-, and alpha 2-adrenoceptors in the posterior hypothalamus to the increased renal sympathetic nerve activity and decreased urinary sodium excretion resulting from environmental stress (air jet) in conscious spontaneously hypertensive rats were examined. Air stress increased mean arterial pressure and renal sympathetic nerve activity (54% from 7.0 +/- 0.7 integrator resets/min), and decreased urinary sodium excretion (44% from 2.7 +/- 0.4 microEq/min per 100 g body weight). After bilateral injection of ICI 118,551 (beta 2-adrenoceptor antagonist) into the posteri

Studies on bethanidine and meobentine: direct and indirect effects of antifibrillatory drugs.

The electrophysiological effects of bethanidine and meobentine were studied on isolated canine cardiac tissues and the in situ dog heart using standard techniques. The "direct" electrophysiological effects of bethanidine (in the beta-adrenergic-blocked Purkinje fiber) resemble the effects of meobentine in the normal canine Purkinje fiber; both drugs produce use-dependent decreases of the maximum rate of depolarization of phase 0 and action potential amplitude. In addition, meobentine prolongs action potential duration (100%) of Purkinje fibers. In ventricular muscle cells, the only significant

Control of ocular blood flow.

The eye has a dual vascular supply. The uveal vessels are distributed within the choroid, the ciliary body, and the iris; and the retinal vessels within the inner parts of the retina. The vascularization of the uvea is very rich and the blood flow is high, which stabilizes the temperature of the eye. The vascularization of the retina is relatively sparse, which is an obvious advantage for the optics. The perfusion pressure in the eye can be defined as the local arterial blood pressure minus the intraocular pressure. Reductions in perfusion pressure, caused by increments in intraocular pressure

Reduced uptake of noradrenaline into storage vesicles from the pregnant uterus.

A noradrenaline (NA) storage vesicle fraction was isolated from the uteri of virgin as well as pregnant guinea-pigs. The uptake of 3H-NA into respective vesicle fractions was compared. Total uptake of 3H-NA in the presence of Mg2+ (2.5mM) and ATP (2.5 mM) at 30 degrees C after 20 min was 6.0 +/- 1.4 pmol . mg-1 in uterus fractions from virgin guinea-pigs as compared to 1.87 +/- 0.41 pmol . mg-1 in uterus fractions from pregnant animals. When desipramine (5mM) was added in order to block neuronal uptake the corresponding figures were 4.75 +/- 0.92 and 0.51 +/- 0.18 pmol . mg-1, respectively. Th

The effect of anaesthesia on the haemodynamic and sympathoadrenal responses of the dog in experimental haemorrhagic shock.

Studies were carried out to demonstrate the effect of sodium pentobarbital and morphine-pentobarbital anaesthesia on the haemodynamic and sympathoadrenal responses of dogs in haemorrhagic shock. The results were compared to those of conscious dogs and of dogs receiving only morphine. The reactivity of the cardiovascular system to exogenous adrenaline was also studied in every group. Pentobarbital did not change the resting plasma level of catecholamines but reduced the increase of plasma catecholamines in shock. In the pentobarbital-anaesthetized dogs in which shock was less severe, the reacti

Central effect of intravenous phenylephrine on baroreflex control of renal nerves.

Responses of renal sympathetic nerve activity were determined in eight chloralose-anesthetized rabbits during sustained (1-3 minutes) increases in arterial pressure induced by phenylephrine infusion, and as arterial pressure returned to control. In four of the eight experiments, aortic baroreceptor traffic was also recorded. When arterial pressure was raised from 81 +/- 5 to 110 +/- 7 mm Hg, renal nerve activity decreased from 30 +/- 7 to 1 +/- 1 imp/sec. Aortic nerve activity increased from 208 +/- 35 to 346 +/- 49 imp/sec. When pressure returned to control (81 +/- 5 mm Hg), renal nerve activ

Alpha adrenoceptors and autonomic mechanisms in perinephritis hypertension in the rabbit.

Increased pressor responses to norepinephrine and other pressor agents have been reported to occur in human essential hypertension and in several animal models of experimental hypertension. These increased responses might be related to the development of hypertension or could be a secondary consequence of the elevation in blood pressure. We have examined pressor responses to alpha-adrenoceptor agonists and to angiotensin II in male New Zealand White rabbits with perinephritic hypertension. Increased pressor responses were observed for the alpha 1 adrenoceptor agonist phenylephrine and the mixe

The effects of stimulation of autonomic nerves on carotid body blood flow in the cat.

1. The effects of electrical stimulation of the distal ends of cut preganglionic cervical sympathetic trunks and cut sinus nerves on carotid body total blood flow, local blood flow and tissue PO2 (Pt,O2) were studied in anaesthetized cats. 2. Sympathetic stimulation caused reductions, often marked, of the total blood flow through the carotid body, yet did not influence local flow or Pt,O2 recorded by electrodes whose tips lay in deep locations within the carotid body. Intraglomeral electrodes did respond to reductions of perfusion pressure and to perfusions of the carotid body with saline solu

Plasma Somatostatin-like immunoreactivity in chemically sympathectomized dogs.

The present study was designed to determine the role of the adrenergic nerves upon basal and postprandial gastric and pancreatic SLI release. In 19 chemically sympathectomized dogs peripheral venous plasma SLI levels in the basal and postprandial state were significantly below those of 30 controls for the first 135 min after the ingestion of a fat-protein meal. To determine the origin of this reduction, the SLI release from fundus, antrum and pancreas was studied in anesthetized dogs during the gastric phase of a meal at either pH 7 or pH 2. In response to a liver meal at pH 7 fundic, antral a

An examination of factors influencing adrenergic transmission in the pithed rat, with special reference to noradrenaline uptake mechanisms and post-junctional alpha-adrenoceptors.

Adrenergic mechanisms were analysed in the pithed rat to determine to what extent the actions of drugs observed in vitro are relevant in situ. The drugs examined were those which are known to block the neuronal or extraneuronal uptake of noradrenaline (cocaine, desipramine and corticosterone) or to be antagonists at post and/or pre-junctional alpha-adrenoceptors (prazosin and yohimbine) together with the antidepressant, mianserin, which has been implicated in several of these actions. These drugs were tested against the arterial diastolic pressor, cardiac chronotropic and vas deferens response

Activation of afferent cardiac sympathetic nerve fibers of the cat by pain producing substances and by noxious heat.

Afferent discharges of 64 single units were recorded from the left cardiac sympathetic nerve of anesthetized cats. Mechano-sensitive terminals of the afferent fibers were localized in the extrapulmonary part of the pulmonary artery, left atrium, left ventricle and left pericardium, as determined by direct mechanical probing of the heart after death of the animals. Conduction velocity of the fibers ranged from 2.5 to 14.6 m/s. Excitation of these Adelta-fibers with mechanically excitable endings was produced by intravenous injections of acetylcholine, 5-hydroxytryptamine, bradykinin, histamine

The importance of adrenergic receptors in disorders of micturition.

A review is presented of the adrenoreceptors in the lower urinary tract, and their relationship to disorders of micturition. It is suggested that they may be of importance of neurogenic bladder disease, bladder neck obstruction, benign enlargement of the prostate, and certain drug and hormonal effects on the lower urinary tract, and that modification of their activity can be of benefit in the management of these conditions.

[Sympathetic-adrenergic and hypophysial response to different anesthesia-analgesia technics].

The authors used techniques of pure anesthesia analgesia with Fentathienyl-Pavulon, or anesthesia and analgesia potentialised with Fentathienyl-Flunitrazepam, or Pentothal-Fentathienyl-Pavulon on various series of patients. The study was oriented on changes in the sympathetico-adrenalin pituitary response, and the use of glucose under the effect of these techniques. There was noted a fall in plasma cortisol levels, a fall in urinary excretion of catecholamines, an increase in growth hormone and better peripheral use of glucose. A comparative study was carried out with similar techniques using
