An approach to predictive testing of contact sensitizers in vitro by monitoring their influence on endocytotic mechanisms.

Endocytotic activation of epidermal Langerhans cells (LC) by immunogenic haptens is an early event during development of allergic contact dermatitis. In this work a fast and objective flow-cytometric assay for predictive in vitro testing of contact sensitizers by monitoring their influence on endocytotic mechanisms in murine LC was developed. Epidermal cell suspensions were labelled with a monoclonal antibody directed to MHC class II molecules and pH-sensitive fluorochrome-coupled second-step reagents. For untreated LC a significant quenching of fluorescence intensity by internalization of the

[The ultrastructure of the parietal exocrinocytes of the glandulae propriae in the gastric mucosa in A avitaminosis].

Electron microscopic and morphometric methods were used to study the state of the parietal exocrinocytes of the proper glands of gastric mucosa in dynamics of A avitaminosis. The experiment was performed in 80 albino rats taken from mother's feeding with the initial body mass 40-50 g, divided into 2 groups. In animals of the first group A-avitaminosis was induced by feeding the animals with semisynthetic diet devoid of vitamin A. The 2nd control group was given the same semisynthetic diet but with retinol acetate (45 units a day for each animal). Ultrastructural findings showing the elevated p

Fermentation of wheat bran and gum arabic in rats fed on an elemental diet.

1. Gum arabic and wheat bran were added to an elemental diet (100 g/kg) in order to study their metabolism in the caeca of adult male albino Wistar rats. 2. Dry stool weight (g/d) over 12 weeks was 0.70 (SE 0.05) on the elemental control diet. Wheat bran increased mean dry stool weight to 1.09 (SE 0.08), an increase of 56%. There was no significant difference between faecal weights (0.65 (SE 0.08)) of the gum-arabic-supplemented group and the unsupplemented group. 3. Wet caecal-sac weight, dry caecal-contents weight, and faecal and caecal bacterial mass (measured by 2,6-diaminopimelic acid) al

Plasminogen activator-specific inhibitors produced by human monocytes/macrophages.

Human monocytes/macrophages produce plasminogen activator-specific inhibitors (PAIs) that form covalent complexes with urokinase-type plasminogen activator (uPA). We have characterized two functionally and antigenically related forms of PAIs produced by resting and phorbol myristate acetate (PMA)-treated U 937 cells: an Mr 40,000 form, presumably nonglycosylated, with a pI of 5.2, that is constitutively synthetized by these cells and that remains predominantly intracellular; a PMA-induced form of heterogeneous Mr (50,000-65,000) with a pI of 4.7, that is preferentially secreted; this PAI is gl

Vitamin A deficiency alters genomic expression for fibronectin in liver and hepatocytes.

Earlier work from our laboratory had shown that vitamin A-deficient rats had increased levels of fibronectin in their serum. To explain this result, we investigated the mechanism whereby vitamin A deficiency affects the production of fibronectin by liver and hepatocytes, since liver is the known source of plasma fibronectin. By use of cDNA specific for rat liver fibronectin, we showed that livers of vitamin A-deficient rats had a 2-4-fold increase in the level of fibronectin mRNA and also a higher transcription rate. Rate of synthesis and secretion of fibronectin was found to be increased 2-fo

High-performance liquid chromatographic analysis of the mycotoxin citrinin and its application to biological fluids.

Citrinin is a toxic metabolite produced by several species of Penicillium and Aspergillus. Citrinin is nephrotoxic and has been implicated in disease outbreaks in animals and humans. Citrinin was resolved in a sharp peak by reversed-phase high-performance liquid chromatography on a small-article (10 micrometers) column by elution in 4.25 min with a phosphoric acid (0.25 N)-acetonitrile-2-propanol solvent (55:35:10). Detection was by ultraviolet absorbance at 340 nm. The relationship between peak height and area and quantity injected was linear over a range of 2--50 ng at 340 nm and 5--200 ng a

Lyophilized rumen fluid from goats fed different roughage-based diets: fermentation characteristics, microbial communities, and post-reconstitution in vitro fermentation performance.

INTRODUCTION: Fresh rumen fluid is a promising donor material for rumen fluid transplantation (RFT), but poor storage stability and batch variability limit its use. METHODS: In this study, lyophilized rumen fluid samples from goats fed alfalfa hay or corn stover diets were designated AHP and CSP, respectively. For the in vitro fermentation assay, portions of lyophilized rumen fluid from each batch within each dietary group were pooled and reconstituted; the substrate-supplemented groups were designated AH and CS, whereas AH-CON and CS-CON served as the corresponding substrate-free controls.

A coculture system of Epicoccum nigrum with Mucor racemosus induces red pigment biosynthesis in vinegar mash.

Fungal pigments hold significant food industrial value due to their safe and sustainable properties. Here we identified a naturally occurring coculture pair Epicoccum nigrum SPDX618 and Mucor racemosus SPDX522 in vinegar mash that produced red pigments, which was not observed in either monoculture. Our results showed that E. nigrum acted as the pigment producer while M. racemosus served as the elicitor. Response surface methodology and pH stabilization strategy improved the yield of red pigment into 328.7 U/g. Through bioactivity-guided fractionation coupled with UHPLC-MS/MS and in silico stru

Synergistic mechanism of stir-baked curcumae radix with vinegar in dysmenorrhea rats based on UPLC-Q-TOF/MS metabolomics.

Curcumae Radix (i.e. Huangsiyujin: HSYJ), a well-known traditional Chinese medicine (TCM), has been widely used in clinical practice for many years to treat depression and primary dysmenorrhea. Modern pharmacological researches have demonstrated its anti-inflammatory, antidepressant, and dysmenorrhea relief effects. According to the processing theory of TCM, it is believed that stir-baked HSYJ with vinegar may enhance the ability to disperse stagnant hepatoqi and alleviate pain. However, whether the vinegar concoction of HSYJ can enhance the therapeutic effect on the Qi stagnation due to liver

Differing Effects of Vinegar on Pelagia noctiluca (Cnidaria: Scyphozoa) and Carybdea marsupialis (Cnidaria: Cubozoa) Stings-Implications for First Aid Protocols.

The jellyfish species that inhabit the Mediterranean coastal waters are not lethal, but their stings can cause severe pain and systemic effects that pose a health risk to humans. Despite the frequent occurrence of jellyfish stings, currently no consensus exists among the scientific community regarding the most appropriate first-aid protocol. Over the years, several different rinse solutions have been proposed. Vinegar, or acetic acid, is one of the most established of these solutions, with efficacy data published. We investigated the effect of vinegar and seawater on the nematocyst discharge p…

The Protective Role of Butyrate against Obesity and Obesity-Related Diseases.

Worldwide obesity is a public health concern that has reached pandemic levels. Obesity is the major predisposing factor to comorbidities, including type 2 diabetes, cardiovascular diseases, dyslipidemia, and non-alcoholic fatty liver disease. The common forms of obesity are multifactorial and derive from a complex interplay of environmental changes and the individual genetic predisposition. Increasing evidence suggest a pivotal role played by alterations of gut microbiota (GM) that could represent the causative link between environmental factors and onset of obesity. The beneficial effects of

Early luteal phase progestin concentration influences endometrial function in pregnant mares.

In the horse, it is still unclear if and to what extent low progestin concentration contributes to early conceptus loss. In the present study, we have investigated if reduced or elevated progestin concentration in the early luteal phase influences endometrial function and conceptus development. We hypothesized that reduced progestin concentration via delayed downregulation of endometrial progesterone receptors (PR) influences endometrial function in healthy fertile mares while progestin substitution does not. Genitally healthy estrous mares (n = 8; age 4-14 years) were inseminated and treated
