Molecular identification of zoonotic hookworms in environmentally collected canine fecal samples in Northern Philippines.

BACKGROUND: Hookworm infections remain an important public health and veterinary concern in tropical and subtropical regions, particularly in communities with close human-animal interactions and limited sanitation. Dogs are recognized as reservoirs of zoonotic hookworms, yet species-level infection data in endemic areas remain lacking because the widely utilized microscopic examination of stool samples cannot differentiate morphologically indistinguishable hookworm eggs. METHODS: Environmentally deposited canine fecal samples (n = 72) were collected from five barangays in Gonzaga, Cagayan, No

Detection of Ehrlichia spp., Candidatus Neoehrlichia sp. (FU98) and Anaplasma phagocytophilum in red foxes (Vulpes vulpes) in Germany.

Vector-borne diseases pose a growing challenge to public health. Effective surveillance, interdisciplinary cooperation in line with the One Health approach, and prevention and control strategies are therefore essential to reduce the risk of future outbreaks and burden of disease in the long term. Identifying and investigating sentinels can help in this regard. In this study, 537 red fox (vulpes) spleen samples from three regions in Germany (Berlin, Brandenburg, Arnsberg administrative region) were tested for Anaplasmataceae and Rickettsia spp. using PCR. Only 3% of the samples (n = 15) tested

Genetic diversity of Trichomonads from Milu deer (Elaphurus davidianus) in China.

The Milu deer, or Père David's deer (Elaphurus davidianus), a rare endemic species in China, represents a case of successful reintroduction of a species previously considered extinct in the wild. Trichomonads, protozoan symbionts capable of infecting vertebrates, are transmitted via the fecal-oral route; they are a subgroup of Parabasalia and include some pathogenic species that pose zoonotic risks. Until now, data on the diversity and prevalence of trichomonads in Chinese Milu deer have not been reported. To better understand the colonization status of trichomonads, fecal samples from 112 Mil

Development and validation of a real-time PCR assay for Cochlosoma anatis in turkeys.

Cochlosoma anatis is a flagellated protozoan parasite in the Trichomonadidae family that causes an enteric disease of turkeys and ducks known as cochlosomosis. Following the withdrawal of effective antiprotozoal medications from commercial poultry production, this organism has become of increased concern for the turkey industry. Historically, this organism has been detected by light microscopic evaluation of enteric mucosal scrapings. However, live trophozoites must be present in the sample for this method of detection because the parasite is differentiated from other protozoa by its character

Practice Versus Potential: The Impact of the BioFire FilmArray Pneumonia Panel on Antibiotic Use in Children.

BACKGROUND: The BioFire FilmArray Pneumonia Panel (BFPP), a multiplex PCR panel for the diagnosis of lower respiratory tract infections, has been proposed as a tool for antimicrobial stewardship. Few studies evaluate real-world implementation of the BFPP and no studies focus exclusively on children. Our institution implemented BFPP testing without restrictions. METHODS: We conducted a retrospective cohort study in children hospitalized at St. Louis Children's Hospital to (1) characterize the use of the BFPP in pediatric patients and (2) assess how results impacted antibiotic use. We included

Human leukocyte antigen-DQA1*04:01 and rs2040406 variants are associated with elevated risk of childhood Burkitt lymphoma.

Burkitt lymphoma (BL) is responsible for many childhood cancers in sub-Saharan Africa, where it is linked to recurrent or chronic infection by Epstein-Barr virus or Plasmodium falciparum. However, whether human leukocyte antigen (HLA) polymorphisms, which regulate immune response, are associated with BL has not been well investigated, which limits our understanding of BL etiology. Here we investigate this association among 4,645 children aged 0-15 years, 800 with BL, enrolled in Uganda, Tanzania, Kenya, and Malawi. HLA alleles are imputed with accuracy >90% for HLA class I and 85-89% for class

Accuracy of quantitative viral secondary standards: a re-examination.

UNLABELLED: Interlaboratory agreement of viral load assays depends on the accuracy and uniformity of quantitative calibrators. Previous work demonstrated poor agreement of secondary cytomegalovirus (CMV) standards with nominal values. This study re-evaluated this issue among commercially produced secondary standards for both BK virus (BKV) and CMV, using digital polymerase chain reaction (dPCR) to compare the materials from three different manufacturers. Overall, standards showed an improved agreement compared to prior work, against nominal values in both log10 copies/mL and log10 internationa

High prevalence and genetic heterogeneity of adenoviruses at a psittacine breeding facility.

A polymerase chain reaction (PCR) survey was performed at an amateur parrot breeding facility in Italy to investigate the presence and molecular characteristics of adenoviruses. Eighty psittacine birds, belonging to seven parrot species, were sampled by cloacal swabs; in addition, 15 livers were collected from specimens that were found dead. Seventy-two out of 95 samples collected were positive for adenoviruses, with a prevalence rate of 75.8%. All seven psittacine species tested positive for at least one genus of the family Adenoviridae; notably, adenoviral infection was found for the first t

Expression activation of over 70% of Chlamydia trachomatis genes during the first hour of infection.

The obligate intracellular bacterium Chlamydia has a unique developmental cycle that alternates between two contrasting cell types. With a hardy envelope and highly condensed genome, the small elementary body (EB) maintains limited metabolic activities yet survives in extracellular environments and is infectious. After entering host cells, EBs differentiate into larger and proliferating reticulate bodies (RBs). Progeny EBs are derived from RBs in late developmental stages and eventually exit host cells. How expression of the chlamydial genome consisting of nearly 1,000 genes governs the chlamy

Identification of Burkholderia cepacia Complex by PCR: A Simple Way.

United States Pharmacopeia (USP) General Chapter <60> for the detection of Burkholderia cepacia complex (Bcc) members in nonsterile products became official in December 2019. This isolation method requires confirmation of the identity of any growth found on Burkholderia cepacia Selective Agar (BCSA) by additional identification tests (refer to the Interpretation section). This article presents a singleplex polymerase chain reaction (PCR) method to rapidly confirm the membership of any microbial grown on BCSA (and other nutrient medium) in the Bcc group. This method is cost effective as it does

Correlation of cytomegalovirus viral load between whole blood and plasma of congenital cytomegalovirus infection under valganciclovir treatment.

BACKGROUND: Congenital cytomegalovirus (CMV) infection (cCMV) can cause sensorineural hearing loss and neurodevelopmental disabilities in children. Oral valganciclovir (VGCV) therapy has been reported to improve long-term audiological and neurodevelopmental outcomes in patients with cCMV. The levels of CMV DNA in whole blood have been monitored in previous studies. However, quantitative methods using whole blood have not been standardized. Recently, the plasma viral load has been standardized and widely used in CMV-associated diseases. METHODS: CMV viral loads in whole blood and plasma were s

Optimizing congenital cytomegalovirus detection by pool testing in saliva by a rapid molecular test.

Universal congenital cytomegalovirus (cCMV) screening in saliva is increasingly recommended. The aim of our study was to correlate the performance of a point-of-care rapid molecular test with CMV real time PCR (CMV RT-PCR) detection, using saliva pool-testing in newborns under a universal screening strategy. Saliva swabs were prospectively collected from newborns < 21 days old and tested by Alethia-LAMP-CMV assay in pools of 5 samples. In positive pools, subjects were tested individually and by saliva and urine CMV RT-PCR. A subset of negative pools were studied with both techniques and viral

Comparison of molecular methods for Bartonella henselae detection in blood donors.

The Bartonella genus consists of neglected pathogens associated with potentially transfusional-transmitted and fatal human diseases. We aimed to evaluate Bartonella sp. prevalence in 500 blood donors and compare the results with the data already published about these samples. We used molecular diagnostic methods to detect Bartonella sp.-DNA from blood and liquid culture samples: (A) conventional PCR for two gene regions, the ITS targeting the genus Bartonella and the specific gltA Bartonella henselae; (B) nested PCR for the ftsZ gene and (C) qualitative real-time PCR for the gltA gene, both B.
