SIVA1 is a multifunctional protein involved in processes that are critical for tumor progression. In breast cancer, SIVA1 is frequently overexpressed; however, its functional role remains poorly defined. This study aimed to elucidate the cellular mechanisms regulated by SIVA1 and to characterize its molecular network in breast cancer. Integrated in silico analyses and in vitro functional and molecular assays in breast cancer cell lines were conducted to investigate SIVA1 expression and associated signaling pathways. Cell proliferation, migration, invasion, and drug sensitivity were assessed in

Immune checkpoint inhibitors (ICIs) have transformed cancer therapy, yet clinical responses remain highly variable, and microbiome-associated findings lack reproducibility across studies. Increasing evidence implicates the gut microbiome in modulating ICI efficacy; however, findings remain inconsistent at the taxonomic level, raising the possibility that functionally convergent immunological mechanisms may underlie this apparent variability. To address this, a critical synthesis was conducted, integrating bibliometric mapping of publications indexed in the Web of Science Core Collection (2013-

INTRODUCTION: Diabetic foot diseases (DFD) severely impair quality of life, yet specific biomarkers and effective therapies remain limited. Identifying causal molecular targets is critical to address this unmet need.
METHODS: We integrated plasma proteome and genome-wide data with Mendelian randomization (MR) to screen therapeutic targets for DFD. Genetic instruments (pQTLs/e- QTLs) proxied plasma protein and gene expression levels. Causal effects on three DFD outcomes (peripheral atherosclerosis, neuropathy, charcot foot) were analyzed via inverse variance weighting and summary-data-based MR

Diabetes is a well-known risk factor for pancreatic adenocarcinoma (PAAD), yet the underlying molecular mechanisms remain unclear. This study employs single-cell sequencing to analyze gene expression patterns and uses Mendelian randomization to assess the association between genetic variations in specific genes and the risk of PAAD. Our findings reveal a significant reduction in the proportion of monocytes in patients with both diabetes and PAAD. Monocytes play a crucial role in the progression of both diseases. Notably, we identified an increase in intermediate monocytes (CD14 + + CD16+) in b

Establishing plasma biomarkers in the veterinary field has always been a challenge, due to a lack of significant understanding of pathophysi … ical signs become evident, which is a significant outcome for fHCM. These proteomic changes suggest critical pathways for earlier intervention and could potentially lead to more effective treatment outcomes. Furthermore, having significant similarity to human disease strengthens the case for using cats as a potential translational model for hHCM.

BACKGROUND: Infectious bronchitis virus (IBV) is a highly contagious virus in poultry causing significant economic losses. Despite the implementation of various vaccines, the continuous emergence of regional variants had made complete protection challenging. T cell responses are critical for immunity against viral infections; however, a deeper understanding in avian T cell-mediated immune responses to IBV is still needed. RESULTS: In this study, we isolated splenocytes form 4–to 8-week-old specific-pathogen free (SPF) chickens and investigated their immune responses to the IBV strain K047-12.

UNLABELLED: Aging is a major risk factor for cardiac diseases, including heart failure, myocardial infarction, and arrhythmias. Activation of p38 MAPKs regulates cardiac remodeling and contributes to age-related cardiac dysfunction. However, the isoform-specific roles of p38 kinases in the aging heart remain poorly understood. Although p38β has been reported to exert cardioprotective effects in models of doxorubicin-induced cardiotoxicity and ischemia-reperfusion, its role in cardiac aging remains unclear. Here, we investigated the role of p38β using p38β germline knockout (p38β -/- ) mice. Ag

Hepatocellular carcinoma (HCC) is distinguished by profound metabolic reprogramming and a highly immunosuppressive tumor microenvironment, two hallmarks that cooperatively drive tumor progression and therapeutic resistance. While metabolic rewiring and immune escape have traditionally been studied as parallel processes, the molecular regulators that integrate these pathways remain incompletely defined. Circular RNAs (circRNAs) are a type of covalently closed non-coding RNAs with exceptionally stable and regulatory versatility, and have recently emerged as critical modulators of both cancer met

Lumpy skin disease virus (LSDV) is a critical transboundary pathogen that causes devastating infections in cattle, buffalo, and other ruminants. The virus induces characteristic clinical manifestations, including cutaneous nodules, marked reduction in milk yield, and impaired production performance, leading to severe economic losses in the global livestock sector. Although LSDV exhibits remarkable multi-tissue tropism and persistent viral shedding in various organs, posing significant challenges for disease control, the molecular mechanisms underlying its tissue-specific adaptation remain poor

The pathogenesis of prevalent metabolic diseases such as obesity, atherosclerosis, metabolic dysfunction-associated steatotic liver disease, and diabetes is intricately linked to dysregulated lipid metabolism. Peroxisome proliferator-activated receptor gamma (PPARγ) is a key transcriptional regulator of lipid homeostasis and a well‑researched therapeutic target. Although biochemical signaling pathways have been the traditional focus, recent studies now highlight the mechanical microenvironment (matrix stiffness, fluid shear stress, and tensile strain) as a pivotal physical metabolism regulator

Aluminum (Al) is the most widely distributed neurotoxic metallic element in the environment, and prolonged exposure can lead to cognitive impairment and increase the risk of developing Alzheimer's disease (AD). This article systematically reviews the critical mechanisms underlying aluminum-induced neurotoxicity: mediating neuronal apoptosis through activation of oxidative stress pathways and induction of mitochondrial dysfunction; engaging programmed cell death modalities such as necroptosis via activation of the RIP1/RIP3/MLKL signaling pathway; disrupting the homeostasis of neurotransmitter

Evolutionary divergence in gene regulation is a major source of phenotypic novelty, yet understanding how deeply conserved, pleiotropic signaling pathways evolve under selective constraint while maintaining essential cellular functions remains an important challenge. Here, we investigate the evolutionary divergence in transcriptional regulation mediated by the mechanistic target of rapamycin (mTOR) pathway in two related species, Drosophila melanogaster and Drosophila simulans. mTOR is a highly conserved central regulator of cellular growth and metabolism, making it well-suited for the study o

This study explores the contribution of ferroptosis, a tightly regulated form of cell death, to the development of high-altitude pulmonary edema (HAPE) under hypoxic conditions. We focused on the central ferroptosis regulator glutathione peroxidase 4 (GPX4) and its interaction with neutrophil extracellular trap (NET) formation. Utilizing a murine model of high-altitude hypoxia and in vitro hypoxia/reoxygenation models, we employed a multi-omics approach to map the molecular landscape of HAPE. Transcriptomic and metabolomic analyses of lung tissues confirmed a significant downregulation of GPX4

Brain-derived growth factor, BDNF, has critical roles in a wide variety of neuronal aspects, including cell survival, differentiation, and synaptic function after their maturation. TrkB, a high-affinity receptor for BDNF, is a major contributor in these neuronal aspects, and its functions are exerted via stimulating intracellular signaling pathways including the mitogen-activated protein kinase (MAPK) pathways. As a family of MAPKs, the functions of ERK1/2, p38MAPK, and JNKs have been extensively studied using in vivo and in vitro neuronal systems. ERK 1/2, a major serine-threonine kinase and

Diabetic kidney disease (DKD) is one of the most common microvascular complications of diabetes and a leading cause of end-stage renal disease. Recently, immune and inflammatory mechanisms have been recognized as critical in the pathogenesis of DKD, among which CD4+ T cell subset imbalance serves as a key contributor to renal inflammation and fibrosis. This narrative review synthesizes evidence from PubMed, Web of Science, and EMBASE (inception to March 2026) to provide a comprehensive overview of the immunopathological roles and molecular regulatory networks of major CD4+ T cell subsets, incl

Pulsatile secretion of gonadotropin-releasing hormone (GnRH) is essential for normal reproductive function in mammals. GnRH pulses from the hypothalamus drive pulsatile gonadotropin release from the anterior pituitary, thereby regulating gonadal activity. Classic studies in rhesus monkeys have demonstrated that intermittent physiological GnRH administration maintains luteinizing hormone and follicle-stimulating hormone secretion, whereas continuous infusion suppresses pituitary responsiveness, establishing the necessity of pulsatile GnRH release. The neural mechanism governing this rhythmic se

Recent technological advances are reshaping the health care landscape and fueling innovation within and outside health systems. Physicians bring critical stewardship to this transformation by launching companies, advising start-ups, and shaping investment decisions; indeed, more than 25% of the 105 health care companies that achieved valuations of more than US$1 billion between 2015 and 2024 had at least one founder who is a clinician. Clinical training rarely fully prepares physicians to engage in these types of initiatives. While formal educational pathways, such as Doctor of Medicine/Master

Cancer screening is increasingly promoted as a public health intervention, yet its effective translation with demonstrable impact on disease burden remains limited in low resource settings due to the suboptimal implementation of guidelines. While evidence from high-income countries and settings have informed international guidelines, the direct application of these recommendations often overlooks the realities of under-resourced settings, where diagnostic infrastructure, referral pathways, and treatment capacity are fragmented and inadequate. Moreover, the societal and health belief systems in

Single-cell RNA sequencing data rely heavily on dimensionality reduction methods to uncover underlying structures and patterns. Nonlinear dimensionality-reduction methods such as uniform manifold approximation and t-distributed stochastic neighbor embedding have been routinely used for this task. However, gene expression data alone often fails to capture and identify changes in cellular pathways, protein complexes, and TF-targets, which are more enlightening at the regulation level. To address this limitation, we present PVAED, a dimensionality reduction framework that integrates biological pr
