Design and Investigation of Carbazole-Dithienylethene Compounds for Switchable Organic Radical Systems.

Photoswitchable radicals incorporating dithienylethene (DTE) moieties offer unique opportunities for dynamic control in various chemical and material applications. By exploiting light-triggered isomerization processes, researchers can achieve precise modulation of radical reactivity, enabling spatiotemporal control of chemical transformations. Despite growing interest in this field, challenges remain in controlling spin-orbit coupling and spin dynamics, which influence the electronic structure and transitions induced by light absorption as well as achieving photostability for repeated photoiso

From crops to clinic: the impact of dual azole use on antifungal resistance in Candida and Candida associated yeasts.

Invasive fungal infections caused by pathogenic yeasts are an escalating global health crisis that demands urgent attention within a One Health framework. This review critically examines mounting evidence that widespread agricultural azole fungicide use is a key driver accelerating antifungal resistance in pathogenic yeast. We dissect the shared molecular targets and resistance pathways that underpin dangerous cross-resistance between environmental fungicides and clinical azoles. Traditionally viewed as human commensals, we provide a comprehensive account of the evidenced environmental reservo

Calcium transport systems as virulence determinants: Mechanistic insights into fungal pathogenicity and antifungal resistance.

This review examines the molecular mechanisms through which calcium transport proteins modulate virulence and drug resistance in human pathogenic fungi, synthesizing recent advances in calcium homeostasis research. Emerging evidence from calcium signaling pathway analyses reveals that fungal calcium transporters (including PMC1, VCX1, and CCH1/MID1 complexes) orchestrate critical stress adaptation processes through calcineurin-dependent and calcineurin-independent pathways, highlighting their critical involvement in fungal physiology, adaptive stress responses, and pathogenicity. Through a sys

Recognizing a MIS-Chievous Cause of Acute Viral Gastroenteritis.

Historically, children evaluated for vomiting and diarrhea secondary to viral enteritis have symptoms lasting 2-4 days and respond to supportive care, including oral rehydration and anti-emetics if required. Recently, within a 14-day timespan, we encountered three children with severe diarrhea who rapidly became dehydrated and went into hypotensive shock. Although SARS-CoV-2 molecular tests were negative by nasopharyngeal swab, all were later found to have MIS-C. This small case series underscores features reported in previous larger studies and emphasizes the rapid clinical evolution of this

Antimicrobial Functions of Lactoferrin Promote Genetic Conflicts in Ancient Primates and Modern Humans.

Lactoferrin is a multifunctional mammalian immunity protein that limits microbial growth through sequestration of nutrient iron. Additionally, lactoferrin possesses cationic protein domains that directly bind and inhibit diverse microbes. The implications for these dual functions on lactoferrin evolution and genetic conflicts with microbes remain unclear. Here we show that lactoferrin has been subject to recurrent episodes of positive selection during primate divergence predominately at antimicrobial peptide surfaces consistent with long-term antagonism by bacteria. An abundant lactoferrin pol

Adaptive cyanogenesis clines evolve recurrently through geographical sorting of existing gene deletions.

Identifying the genetic basis of parallel phenotypic evolution provides insight into the process of adaptation and evolutionary constraint. White clover (Trifolium repens) has evolved climate-associated adaptive clines in cyanogenesis (the ability to produce hydrogen cyanide upon tissue damage) in several world regions where it has been introduced. Gene-deletion polymorphisms at the CYP79D15 and Li loci underlie the presence/absence of the cyanogenic phenotype. Both loci have undergone multiple independent gene-deletion events, which are identifiable through molecular signatures in flanking re

Chronic myeloid leukemia stem cell biology.

Leukemia progression and relapse is fueled by leukemia stem cells (LSC) that are resistant to current treatments. In the progression of chronic myeloid leukemia (CML), blast crisis progenitors are capable of adopting more primitive but deregulated stem cell features with acquired resistance to targeted therapies. This in turn promotes LSC behavior characterized by aberrant self-renewal, differentiation, and survival capacity. Multiple reports suggest that cell cycle alterations, activation of critical signaling pathways, aberrant microenvironmental cues from the hematopoietic niche, and aberra

Transformation of diffuse large B-cell lymphoma into pre-B acute lymphoblastic leukemia: clinicopathologic features and clonal relationship.

A patient with fibrosing alveolitis developed a diffuse large B-cell (DLBC) lymphoma that expressed CD20 and CD30. After an initial response, the lymphoma relapsed and was salvaged with further chemotherapy. After another remission of 3 years, a pre-B-cell acute lymphoblastic leukemia (ALL), which expressed CD10, CD19, CD22, CD79a, CD34 and terminal deoxyribonucleotidyl transferase, developed and led to death. Molecular analysis of the immunoglobulin heavy-chain gene showed that the initial lymphoma and its relapse were clonally related. At leukemic relapse, 2 clones related to the initial and

Gene-driven self-morphing microtubule-based active matter.

Materials that change shape or flow typically rely on external fields, photochemical cues or compositions fixed at assembly. Here we introduce a self-morphing active material whose mechanical behaviour is genetically programmed and executed autonomously. By embedding cell-free gene expression within a dense microtubule network, DNA instructions synthesize molecular motors, cross-linkers and transcription factors that continuously generate and regulate active stresses. This experimental platform enables gene-circuit architectures to directly program active-matter dynamics, producing steady, pul

Unlocking the molecular basis of the unique jujube aroma in white tea: pile-withering driven aroma formation.

Jujube-aroma white tea (JWT) can be produced via pile-withering, yet the key compounds and mechanisms responsible for its characteristic aroma remain poorly understood. This study systematically investigated the dynamic evolution of volatile and non-volatile compounds during JWT processing, identifying 107 volatile compounds and 421 differential metabolites. Pile-withering promoted the accumulation of alcohols, aldehydes, and ketones, while reducing hydrocarbons and esters. Through comparative analyses of samples from different processing stages, as well as red jujubes and aged white tea, seve

Homogeneous copper-catalyzed aerobic oxidation of alcohols and aldehydes: mechanistic insights and recent advances.

Copper-catalyzed aerobic oxidation of alcohols and aldehydes has become a key transformation for the sustainable synthesis of carbonyl compounds, combining high efficiency with the use of molecular oxygen as a benign terminal oxidant. Owing to its earth-abundance, low cost, low toxicity, and versatile redox properties, copper offers a compelling alternative to traditional noble metal catalysts and stoichiometric oxidation methods. This review highlights the evolution of copper-catalyzed aerobic oxidation of alcohols, emphasizing two distinct mechanistic regimes: nitroxyl radical-mediated syste

Clonal Evolution and Lineage Switch from T-Cell Acute Lymphoblastic Leukemia to Acute Myeloid Leukemia in Therapy-Resistant PICALM::MLLT10 Leukemia.

BACKGROUND: PICALM::MLLT10-positive T-ALL is rare and associated with poor prognosis. Lineage switch to AML is exceptionally uncommon, particularly after long-term remission. CASE PRESENTATION: We report an adolescent PICALM::MLLT10-positive T-ALL with a cortical thymocyte, non-ETP phenotype. The patient achieved complete remission but relapsed as AML 6 years later. Cytogenetics revealed del(5q), del(17p), and 17q gain. Mutational profiling demonstrated mutations with LOH in NF1 and EZH2, a hemizygous SMC1A mutation, and a hemizygous PHF6 mutation detected only after subsequent therapy. CONC

Decoding the medicinal aromatic characteristic of Zhuyeqing and gaining new insight into sotolon.

Zhuyeqing is made from Fenjiu base liquor infused with extracts from 12 Chinese herbs. Its medicinal aroma is a unique feature of Zhuyeqing. However, this particular characteristic has not yet been thoroughly studied. Utilizing a normal phase liquid chromatography-sensomics-integrated strategy, we sequentially identified 110 potential odorants in the fraction with characteristic medicinal aroma via GC × GC-TOF MS, then these compounds were narrowed down to 25 sensorially active compounds through GC-O validation. Quantitative analysis identified 10 odorants with odor activity values (OAVs) ≥ 1.

Molecular Bridge Engineering at the Anode/Electrolyte Interface: Synergistic MPS/ZnO Hybrid Corrosion Inhibition for High-Utilization Alkaline Al-H2O Batteries.

Aluminum-water (Al-H2O) batteries, enabling on-demand high-purity hydrogen production and simultaneous power output, hold great promise for hydrogen energy and electrochemical energy storage applications. However, severe self-corrosion of aluminum anode and parasitic hydrogen evolution reaction (HER) in highly alkaline electrolytes hinder their practical deployment. This work proposes a novel organic-inorganic hybrid corrosion inhibitor comprising sodium 3-mercapto-1-propanesulfonate (MPS) and zinc oxide (ZnO). Combined experimental and computational investigations demonstrate that a dense, ro

Template-Encoded Assembly and Adaptive Function in Selenium-Based Molybdenum Chalcoxide Rings.

Controlling both structure and function in inorganic self-assembly remains a major challenge. Here we establish a programmable strategy for constructing selenium-based molybdenum chalcoxides using the [Mo2O2Se2]2+ dimer as a universal building block. The template-directed assembly process produces a complete family of ring architectures (n = 8-16), with nuclearity encoded by the template's geometry and acidity. Selenium incorporation introduces electronic softness that imparts unusual structural flexibility and enables adaptive behaviour. NMR titration experiments reveal host-guest exchange in

Zebrafish sox9b+ cells function as intestinal stem cells for epithelial homeostasis and regeneration.

Intestinal stem cells (ISCs) are essential for maintaining epithelial homeostasis and driving tissue regeneration. The zebrafish intestine features a distinctive furrow-ruga architecture and lacks the crypt structures characteristic of mammalian intestines, leaving its ISC identity elusive. Characterizing zebrafish stem cell populations and their molecular markers is informative for intestinal physiology and evolution. Here, we identify sox9b+ cells residing within the proliferative zone of intestinal furrows as functional zebrafish ISCs. These cells contribute to epithelial turnover during ho

Conception-calibrated male pediatric tumor mitotic clocks.

Molecular clocks can reconstruct tumorigenesis, but their calibration is limited by uncertainty in cancer ages. Pediatric cancers simplify this problem because age ranges are narrowly bounded by conception and a minimum of ~30 divisions. We developed mitotic clocks from rapidly fluctuating CpG (fCpG) DNA methylation on the X chromosome in male cancers, applying a two-state Markov model to estimate mitotic age and epimutation rates. Across acute lymphoblastic leukemia, acute myeloid leukemia, neuroblastoma, and embryonal brain tumors, modeled and observed methylation data were highly concordant

Multifocal papillary thyroid carcinoma with RET p.V804M, EML4-ALK fusion, and BRAF V600E positivity.

Multifocal papillary thyroid cancer (PTC) can arise from independent primaries with discordant drivers in parallel clonal evolution rather than a single-clone pattern. We present a 31-year-old female with multifocal PTC harboring 3 distinct oncogenic alterations: a germline RET p.V804M mutation, low-frequency EML4-ALK fusion, and BRAF V600E mutation. The RET and ALK alterations were identified in a midpole nodule, whereas BRAF positivity was seen in a separate lower pole tumor. Ultrasound revealed multiple right-lobe thyroid nodules; the dominant 2.1-cm lesion was hypoechoic with calcification

DNAzyme-Based Point-of-Care Diagnosis of Aggressive Periodontitis.

The rapid and accurate detection of Aggregatibacter actinomycetemcomitans (AA), a key pathogen in aggressive periodontitis, remains a critical challenge in point-of-care diagnostics. Here, we report the development of a portable lateral flow device (LFD) using an RNA-cleaving DNAzyme (RCD-AA5) as a highly specific molecular recognition element. Selected via systematic evolution of ligands by exponential enrichment (SELEX) against AA crude extracellular mixture (CEM-AA), RCD-AA5 demonstrated exceptional specificity, distinguishing the target from seven other Gram-negative and Gram-positive bact
