Ginsenoside Rg1 Restores Sirt2/Foxo1 Expression and Alleviates Autism-Like Behaviors in a Valproic Acid Induced Male Mouse Model.

This study investigated whether Ginsenoside Rg1 (Rg1) alleviates autism-like behaviors in mice prenatally exposed to valproic acid (VPA) via Sirt2/Foxo1 signaling. Pregnant C57BL/6J mice received a single intraperitoneal injection of VPA (600 mg/kg) on embryonic Day 12.5 to establish an autism model. At 8 weeks of age, male offspring were randomly divided into four groups: Normal, VPA, VPA + Rg1 (5 mg/kg), and VPA + Rg1 (10 mg/kg). Rg1 was administered once daily for 28 days. Behavioral assessments included grooming, rearing, locomotor activity, social interaction, novel object recognition, op

LLM-Guided Pain Management: Examining Socio-Demographic Gaps in Cancer vs non-Cancer cases.

Large language models (LLMs) offer potential benefits in clinical care. However, concerns remain regarding socio-demographic biases embedded in their outputs. Opioid prescribing is one domain in which these biases can have serious implications, especially given the ongoing opioid epidemic and the need to balance effective pain management with addiction risk. We tested ten LLMs-both open-access and closed-source-on 1,000 acute-pain vignettes. Half of the vignettes were labeled as non-cancer and half as cancer. Each vignette was presented in 34 socio-demographic variations, including a control g

The Role of TDP-43 in SARS-CoV-2-Related Neurodegenerative Changes.

The coronavirus disease 2019 (COVID-19) pandemic has been linked to long-term neurological effects with multifaceted complications of neurodegenerative diseases. Several studies have found that pathological changes in transactive response DNA-binding protein of 43 kDa (TDP-43) are involved in these cases. This review explores the causal interactions between severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) and TDP-43 from multiple perspectives. Some viral proteins of SARS-CoV-2 have been shown to induce pathological changes in TDP-43 through its cleavage, aggregation, and mislocaliz

Polyphenolic extracts from Diospyros kaki and Vitis vinifera by-products stimulate cytoprotective effects in bacteria-cell host interactions by mediation of transcription factor Nrf2.

BACKGROUND: The intestinal and skin epithelium play a strong role against bacterial stimuli which leads to inflammation and oxidative stress when overwhelmed. Polyphenols from fruit-rich diets and by-products show promise against bacterial deleterious effects; however, their antibacterial and health-promoting effects remain understudied. PURPOSE: This study aimed to assess the impact of polyphenolic extracts of grape (GrPE), persimmon (PePE) and pomegranate (PoPE) by-products on bacterial pathogen-host interactions, focusing beyond growth inhibition to explore their effects on bacterial adhes

An integrated experimental-computational framework for elasto-plastic characterization and spatial mapping of canine bone properties.

Bone is a hierarchical and heterogeneous bio-composite material, with cortical and trabecular regions exhibiting distinct mechanical behaviors across scales. Patient-specific Finite Element Analysis (FEA) derived from medical images is increasingly used in both human and veterinary medicine to predict bone mechanical behavior under various loading conditions, assess fracture risk, and evaluate treatment outcomes. However, due to the complexity associated with measuring full elasto-plastic bone properties across multiple scales and bridging microscale properties to macroscale models, simplified

Targeting TOMM40 unleashes immunogenic mitophagy to facilitate antigen presentation and immunotherapy sensitization.

BACKGROUND: Mitophagy is a mitochondrial quality control process that maintains cellular homeostasis in cancer, yet whether its dysregulation can be exploited to induce tumor immunogenicity remains unclear. METHODS: We integrated pancancer single-cell transcriptomic analyses with genetic perturbation strategies in hepatocellular carcinoma models, including CRISPR/Cas9-mediated gene depletion, in vivo syngeneic tumor systems, and RNA-based lipid nanoparticle delivery. Mechanistic investigations combined mitochondrial functional assays, imaging-based mitophagy analysis, flow cytometry, and tran

Evolutionary Divergence, Predicted Interaction Interface, and Regulatory Specialization of MTB as a Non-Catalytic Scaffold in the Plant m6A Writer Complex.

N6-methyladenosine (m6A) is the most prevalent internal modification of eukaryotic mRNA and a central regulator of plant development and stress adaptation. The plant m6A writer complex requires two MT-A70 family proteins, the catalytic subunit MTA70 and its non-catalytic partner MTB, yet the evolutionary basis and structural logic underlying this functional division remain unresolved across land plant lineages. Here, we present an integrative computational analysis of MTA70 and MTB across 15 phylogenetically representative species spanning bryophytes, lycophytes, charophyte algae, monocots, an

Protective effect of ischemic preconditioning against hepatic ischemia-reperfusion injury and associated remote organ damage.

Hepatic ischemia-reperfusion injury is a significant complication of liver surgery, including major hepatectomy, trauma surgery and liver transplantation. It is a key factor in postoperative organ failure, which negatively affects prognosis and overall patient survival. Beyond its localized hepatic effects, ischemia-reperfusion injury is increasingly recognized as a potent trigger of the systemic inflammatory response and remote organ damage. The cellular and molecular mechanisms involved are highly complicated and have yet to be entirely elucidated. The core pathophysiological mechanisms of h

Immune imbalance between T helper 1, T helper 17 and regulatory T cells fuels amyotrophic lateral sclerosis pathogenesis: disease trajectory, diagnosis and therapeutic implications.

Amyotrophic lateral sclerosis (ALS) is a fatal, rapidly progressive neurodegenerative disease, causing motor neuron loss and with limited treatment options. Although traditionally considered non-immune in origin, accumulating evidence implicates the adaptive immune system, particularly CD4+ T cell subsets, as key modulators of disease progression. Among these, T helper (Th)1 and Th17 cells are elevated in ALS blood, cerebrospinal fluid and central nervous system tissues, and drive pro-inflammatory cascades. By contrast, regulatory T cells (Tregs) suppress pathogenic inflammation and maintain i

Repurposing Alzheimer's and ovarian cancer drugs as sonosensitizers for glioblastoma via a positive-unlabeled learning and 3D bioprinting-based new approach methodology (NAM).

Glioblastoma (GBM) remains a lethal primary brain tumor, in part because therapeutic efficacy is limited by the blood-brain barrier (BBB) and the complex tumor microenvironment (TME). Sonodynamic therapy (SDT), i.e., use of ultrasound to activate chemical sensitizers and generate cytotoxic stress, offers a non-invasive strategy for treating deep-seated intracranial disease, but progress is constrained by the scarcity of validated sonosensitizers and the inefficiency of conventional in vitro screening methods. Here, we introduce a New Approach Methodology (NAM) that couples a neural network-bas

Maternal transfer of bisphenol A impacts the ontogeny of cortisol stress response in rainbow trout.

Bisphenol A (BPA) is widespread in the aquatic environment, and early life-stage exposure to this chemical affects growth and development in fish. We tested the hypothesis that BPA accumulation in eggs, mimicking maternal transfer, impacts the development and function of the hypothalamus-pituitary-interrenal (HPI) axis in rainbow trout (Oncorhynchus mykiss). To mimic maternal transfer, oocytes were loaded with 0 (control), 1, 4 and 40ng BPA embryo(-1). We measured the temporal changes in whole body cortisol content, transcript levels of corticosteroidogenesis-related genes and glucocorticoid r

Induction of cell surface peptidase activity: a global response to cell stress correlated with apoptosis.

We have previously characterized the stimulation of HeLa cell surface peptidase activity directed toward a nonapeptide substrate in response to low fluences of ultraviolet irradiation [Brown et al. (1993): J Cell Biochem 51:102-115]. To explore the hypothesis that this comprised a global response to cell stress featuring the interruption of DNA synthesis, a variety of agents affecting macromolecular synthesis were applied to HeLa cell cultures. Actinomycin D, 5,6-dichloro-1 beta-ribofuranosyl benzimadazole, mitomycin C, ultraviolet light, and cycloheximide at doses which inhibited cell growth,

Differential Effects of Five Rearing Systems on Immune-Related Gene Expression in the Blood and Spleen of Termond White Rabbits.

BACKGROUND/OBJECTIVES: Improving rabbit welfare through alternative housing systems requires a better understanding of how environmental conditions modulate physiological and immune responses at the molecular level. This study aimed to evaluate the influence of different rearing systems on the expression of genes associated with inflammation, immune regulation, and stress response in Termond White rabbits. METHODS: After weaning (35 days of age), Termond White females (n = 16 per group) were allocated to five housing systems differing in space allowance and activity opportunities: hutches wit

Cigarette smoke-induced injury induces distinct sex-specific transcriptional signatures in mice tracheal epithelial cells.

The airway epithelial barrier is crucial for defending against respiratory insults and diseases. Disruption of epithelial integrity contributes to respiratory diseases, and sex-specific differences in susceptibility and severity have been observed. However, sex-specific differences in the context of respiratory diseases are often overlooked, especially in murine models. In this study, we investigated the in vitro transcriptomics of male and female murine tracheal epithelial cells (mTECs) in response to chronic cigarette smoke (CS) exposure using an International Organization for Standardizatio

Early-life microbial contact with adult hens modulates cecal microbiota and mitigates host responses to lipopolysaccharide-induced stress in broilers.

BACKGROUND: In commercial production, the absence of adult hens disrupts natural gut colonization, forcing chicks to rely on environmental microbes and potentially compromising the ecological succession essential for health. This study investigated whether early-life adult hen microbial contact could optimize the intestinal community and metabolite profiles, thereby mitigating lipopolysaccharide (LPS)-induced immunological stress in broilers. Accordingly, 128 one-day-old broilers were assigned to control or adult hen microbial contact groups. Hens were removed on D 10, and LPS was administered

Distinct 24-hour movement behaviour profiles and their associations with negative affect among Chinese university students.

BACKGROUND: University students experience a high prevalence of depression, anxiety and stress, while simultaneously undergoing substantial changes in daily movement behaviours. From a 24-hour movement behaviour perspective, sleep, sedentary behaviour and physical activity are interdependent components of a fixed time budget, yet little is known about how distinct combinations of these behaviours cluster among Chinese university students and how such patterns relate to negative affect. METHODS: A cross-sectional online survey was conducted from October 2024 to April 2025 using stratified clus

An integrated Msr-antioxidase-host gene circuit maintains redox homeostasis in legume-rhizobium symbiosis.

Methionine sulfoxide reductases (Msrs) play a critical role in oxidative stress resistance; however, their functions in rhizobium-legume symbiotic nitrogen fixation (SNF) are not well understood. In this study, we systematically characterized four Msrs (MsrA1, MsrB1, MsrA2, MsrB2) from Mesorhizobium huakuii 7653R, a symbiotic partner of Astragalus sinicus. Sequence and phylogenetic analyses confirmed the presence of conserved catalytic domains and revealed genus-specific clustering of these Msrs. Expression profiling demonstrated distinct patterns: msrA1 and msrB1 were transiently induced duri
