Structure-Based Optimization of Triazole-Linked Sesamol Conjugates as Antifungal Agents Targeting CYP51 in Candida albicans.

The increasing prevalence of azole-resistant Candida albicans due to extensive and irrational use of antifungal agents demands the developme … e was not observed in a 30 day resistance induction assay. CH3 was nontoxic and capable of rescuing larvae (G. mellonella) from the Candida-infected larvae model. With a favorable safety profile and desirable potency in both in vitro and in vivo, CH3 is a promising lead molecule for future drug development against Candida infections.

Morphology and morphometry of Pseudoproleptus sp. (Nematoda: Cystidicolidae) parasitic on amazonian fish.

Nematodes of the family Cystidicolidae Skryabin, 1946 represent a complex group of parasites of aquatic organisms. They are present in freshwater, estuarine, and marine environments, with fish as intermediate, paratenic, and definitive hosts. This study adds relevant data on the morphology, morphometry, and parasitic ecology of third-stage larvae of Pseudoproleptus Khera, 1953 in fish consumed by riverside populations in the Amazon. A total of 92 fish specimens were acquired between March 2024 and January 2025 from artisanal fishing, distributed among five species: Astronotus ocellatus; Cichla…

Novel brood-site pollination mutualism between sympetalous Heterosmilax (Smilacaceae, Liliales) and a cecidomyiid gall midge (Cecidomyiidae, Diptera) breeding in fallen male flowers.

Heterosmilax is a unique dioecious genus of Smilacaceae (Liliales, Monocotyledon) in that both male and female flowers are sympetalous, ellipsoid, and almost closed. Our field observations in the Ryukyu Islands of Japan showed that H.japonica flowers are visited and pollinated exclusively by females of one cecidomyiid gall midge species, whose larvae breed in fallen male flowers and feed initially on pollen and later on floral tissue. This is the first example of obligate gall midge-associated brood-site pollination mutualism in which the pollinator brood site is fallen male flowers. The polli…

Comprehensive Management of Multisite Myiasis in a Critically Ill Patient: A Case Report Highlighting Challenges in Vulnerable Populations.

A 44-year-old unhoused male with a history of alcohol dependence was admitted to the emergency department with symptoms of tremors, agitation, and generalized pain. His condition rapidly deteriorated, revealing a subdural hematoma that necessitated an urgent craniotomy. Following surgery, he was diagnosed with multisite myiasis, with maggots present in his ears, nose, eye, and toenail bed. The treatment included surgical removal of the larvae, administration of ivermectin, and broad-spectrum antibiotics to prevent secondary infections. ENT evaluations confirmed further infestations, identifyin…

Ultrastructure of the larval rectum of the scorpionfly Panorpa liui (Mecoptera: Panorpidae).

The rectum is an important part of the alimentary canal responsible for ion and water reabsorption of insects. However, it has rarely been studied in the larvae of Panorpidae, the largest family in Mecoptera. Here, we investigated the ultrastructure of larval rectum of the scorpionfly Panorpa liui Hua, 1997 using light and transmission electron microscopy. The rectum comprises tracheal muscular layers, connective tissue, non-cellular basal lamina, junctional cells, rectal epithelium, cuticle with irregular outlines, and a central lumen. The rectal epithelium is infolded to form six longitudina…

Ultrastructural comparison of the larval midguts between Trypoxylus dichotomus (Linnaeus, 1771) and Anomala corpulenta (Motschulsky, 1854) (Coleoptera: Scarabaeidae).

Larvae are the most important feeding and developmental stage in the life cycle of insects. Correspondingly, the larval midguts, as the primary digestive organs, undergo diverse specialization among insect lineages. Larvae of Scarabaeoidae, commomly known as white grubs, exhibit diversity on feeding habits at the familial or subfamilial level. However, the ultrastructure of larval midguts is not yet satisfactorily understood. In this study, the larval midguts of Trypoxylus dichotomus and Anomala corpulenta were compared using light and transmission electron microscopy for the first time, to un…

Case Report: Re-Emergence of Cochliomyia hominivorax in Costa Rica: Report of a Human Myiasis Case 23 Years after Elimination.

The New World screwworm, Cochliomyia hominivorax Coquerel (Diptera: Calliphoridae), was officially eliminated from Costa Rica in 2000, but it was reintroduced in 2023. A myiasis by C. hominivorax in a 71-year-old man with a 4-month history of foot hyperkeratosis and interdigital ulcers is reported. The myiasis was detected before sampling for bacterial culture. Approximately 160 first- and second-instar larvae were recovered and identified as C. hominivorax. Morphological identification was based mainly on characteristics of the cephalopharyngeal skeleton, spiracles, and pigmented dorsal trach…

Synthesis of Esters Containing Cinnamoyl Motif with Potential Larvicide Action: A Computational, Ecotoxicity and in Vitro Cytotoxicity Assessments.

An increasing morbidity and mortality rate has been related to arboviruses transmitted by Aedes aegypti. Compounds with cinnamoyl moiety represent an alternative against mosquitos, considering their larvicidal activity. This study aimed to assess the larvicidal activity of cinnamic ester derivates against Aedes aegypti larvae, along with evaluating their toxicity effect to assess its safety as a larvicide. Ethyl cinnamate demonstrated larvicidal activity (LC50 =48.59 μg/mL). Morphological changes in larvae were detected, as a degenerative response in the thorax. Through molecular docking, the …

Tapeworms as pathogens of fish: A review.

Tapeworms (Cestoda) represents a species rich (about 5000 species) group of flatworms (Neodermata) parasitizing all groups of vertebrates including humans, with about 1000 species parasitizing elasmobranchs and almost 500 occurring in teleosts as adults. They are common parasites of cultured fish, both as adults and larvae (metacestodes), but only few adult tapeworms are actually pathogenic for their fish hosts. In contrast, cestode larvae can be harmful for fish, especially plerocercoids migrating throughout their tissue and internal organs. Current knowledge of host-parasite relationships, i…

Strongyloides Hyperinfection Associated with Enterococcus faecalis Bacteremia, Meningitis, Ventriculitis and Gas-Forming Spondylodiscitis: A Case Report.

An elderly Singaporean male with no travel history was hospitalized for fever and altered mental status. Blood cultures grew Enterococcus faecalis, and given a preceding history of steroid use and peripheral eosinophilia, Strongyloides hyperinfection was suspected. Stool specimens were positive for Strongyloides stercoralis larvae over four days, and larvae were also isolated in an early morning nasogastric aspirate specimen prior to initiation of ivermectin. A cerebrospinal fluid examination was consistent with partially treated bacterial meningitis and ventriculitis was demonstrated on neuro…

Antagonism between parasites within snail hosts impacts the transmission of human schistosomiasis.

Human disease agents exist within complex environments that have underappreciated effects on transmission, especially for parasites with multi-host life cycles. We examined the impact of multiple host and parasite species on transmission of the human parasite Schistosoma mansoni in Kenya. We show S. mansoni is impacted by cattle and wild vertebrates because of their role in supporting trematode parasites, the larvae of which have antagonistic interactions with S. mansoni in their shared Biomphalaria vector snails. We discovered the abundant cattle trematode, Calicophoron sukari, fails to devel…

Early life exposure of zebrafish (Danio rerio) to synthetic pyrethroids and their metabolites: a comparison of phenotypic and behavioral indicators and gene expression involved in the HPT axis and innate immune system.

Ecotoxicological studies have revealed the association between synthetic pyrethroid (SP) exposure and aquatic toxicity in fish; however, research on the toxic effects of SP metabolites is still limited. In this study, the toxicity of two SPs (permethrin (PM) and β-cypermethrin (β-CP)) and their three metabolites (3-phenoxybenzoic alcohol (PBCOH), 3-phenoxybenzaldehyde (PBCHO), and 3-phenoxybenzoic acid (PBCOOH)) towards zebrafish embryos and larvae was evaluated. Both SPs and their metabolites exhibited significant developmental toxicities, caused abnormal vascular development, and changed loc…
