Chemosensitizer Effects of Coencapsulation of Curcumin and Cabazitaxel in Nanostructured Lipid Carriers in Glioblastoma Cells.

Gliomas are the most common primary tumors of the central nervous system and are associated with poor prognosis. Current therapy with Temozolomide shows limited efficacy, highlighting the need for new treatment options. Cabazitaxel (CBZ), a chemotherapeutic agent approved for prostate cancer, has shown efficacy in preclinical glioblastoma models. However, there is a need for strategies to reduce toxicity and enhance delivery. Curcumin (CUR), known for its antitumor, anti-inflammatory, and antioxidant properties, has been investigated as a potential chemosensitizer. Here, we developed and chara

MARCHF6 orchestrates hepatic lipid homeostasis by targeting SREBP1 for ER-associated degradation.

BACKGROUND & AIMS: Increased de novo lipogenesis, largely mediated by sterol regulatory element-binding protein 1 (SREBP1), is a hallmark of metabolic dysfunction-associated steatotic liver disease (MASLD). However, the post-translational mechanisms regulating SREBP1 turnover remain poorly understood. The endoplasmic reticulum-associated degradation (ERAD) pathway ensures protein quality and quantity control, yet its role in hepatic lipid metabolism remains elusive. Here, we investigate the function of the ERAD-specific E3 ubiquitin ligase MARCHF6 in hepatic lipid homeostasis and MASLD pathoge

[Research progress in mechanisms of dietary resistant starch for regulating glucose and lipid metabolism].

The global prevalence of metabolic diseases such as obesity, diabetes, and cardiovascular diseases is closely related to overnutrition and imbalanced dietary patterns. As an important carbohydrate, starch directly affects the homeostasis of glucose and lipid metabolism due to its digestion characteristics. Resistant starch (RS) with unique anti-digestive properties and prebiotic functions has become the current hotspot in dietary nutrition research for improving glucose and lipid metabolism disorders. This review summarizes the digestive characteristics of starch and the comprehensive effects

Peroxisome deficiency impacts metabolites of lysine, lipid, and polyamine metabolism in Saccharomyces cerevisiae.

Peroxisomes are eukaryotic organelles primarily known for their conserved roles in fatty acid β-oxidation and hydrogen peroxide detoxification. These organelles are also involved in a diverse range of other metabolic and non-metabolic functions. We recently compared the transcriptome and proteome of Saccharomyces cerevisiae wild-type and peroxisome-deficient (pex3) cells. This study uncovered the major processes and metabolic pathways that are influenced by peroxisomes. Here we performed a mass spectrometry-based analysis of intracellular metabolites of the same two strains. This led to the id

Lipid dysregulation as a central contributor of neurodegenerative diseases: Emerging therapeutic targets and strategies.

Lipid homeostasis is essential for preserving the structural integrity and functional capacity of the brain. A diverse array of lipids, including cholesterol, phospholipids, and sphingolipids, has been identified as playing pivotal roles. Dysregulation of lipid metabolism is increasingly recognized as a central pathological mechanism in neurodegenerative diseases, including Alzheimer's Disease, Parkinson's Disease, Amyotrophic Lateral Sclerosis, Huntington's Disease, and Cerebrotendinous Xanthomatosis, though much of the existing evidence comes from associative studies, and causal relationship…

Charge-driven lipid nanoparticle encapsulation of enzymes for enhanced ERT with reduced immunogenicity.

Efficient intracellular delivery of therapeutic enzymes remains a central limitation of protein-based therapies. In lysosomal storage disorders such as Pompe disease, classical enzyme replacement therapy (ERT) is particularly constrained by inefficient delivery to skeletal muscle, limited cellular uptake, and immunogenicity. Here, we introduce a lipid nanoparticle (LNP) platform designed for charge-mediated enzyme encapsulation, enabling high enzyme loading (up to 100 enzymes per LNP) while preserving catalytic activity. Developed enzyme-loaded LNPs achieve efficient delivery to muscular tissu

Tissue-specific fibroblast lipid cues impose the rate of epithelial cancer invasion.

Squamous cell carcinomas (SCCs) originate in epithelial tissues of older individuals who have been exposed to environmental carcinogens. Despite overlapping clinical hallmarks, SCCs from different anatomic sites have different prognoses. Here we show that fibroblasts confer site-specific cues that determine SCC proliferation and invasion. Oral and lung fibroblasts have distinct lipid metabolism, transferring unique lipids to SCC cells that promote epithelial-to-mesenchymal transition, and oral and lung SCC invasion. Whereas oral fibroblasts transfer sphingomyelins, which activate the ceramide-…

A novel NAMPT activator ameliorates obesity by repressing ACSL1-dependent lipid synthesis.

Excessive incorporation of long-chain fatty acids (LCFAs) into triglycerides in adipose tissue is a key contributor to obesity and related metabolic disorders, and pharmacologically modulating this process remains challenging. Here, we synthesized a library of β-indoquinazolinone derivatives via palladium-catalyzed oxidative addition complex chemistry and identified compound b2b as a potent and selective anti-obesity candidate. b2b inhibited triglyceride accumulation in adipocytes in vitro and significantly reduced adiposity, body weight, and lipid metabolic disturbances in diet-induced obese

Lipid metabolism, neuroinflammation, and oxidative stress in Alzheimer disease: an integrated mechanistic review.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterised by the accumulation of amyloid-β plaques, tau tangles, and extensive synaptic and neuronal loss. Increasing evidence suggests that the condition develops through the combined effects of protein misfolding, lipid dysregulation, oxidative stress, and chronic neuroinflammation. Among these processes, the APOE ε4 allele has a central role by linking disrupted lipid metabolism to impaired amyloid clearance, abnormal tau phosphorylation, and heightened neuronal vulnerability. This relationship highlights lipidopathy a

Lipid and Carbohydrate Modifications Enhance Potency of Antisense Oligonucleotides Targeting Oncogenic KRAS.

One prominent open question in the field of antisense oligonucleotide remains the intracellular delivery of active sequences. While different strategies have been developed for addressing this issue, including formulation and/or conjugation approaches, multiconjugations with both sugars and nucleolipids have not been investigated so far. In this contribution, we are interested in a new hypothesis that focuses on the double conjugation of oligonucleotides with both glucose-thymine and nucleolipid moieties. The lipid carbohydrate modified antisense oligonucleotides were the most efficient in dow…

Magnetic Lipid Nanoparticles for Targeted mRNA Delivery and Enhanced Organ-Specific Expression.

Targeted delivery of mRNA therapeutics offers immense potential for improving treatment efficacy while reducing off-target effects. Despite extensive efforts to develop both passive and active targeting strategies, achieving precise and efficient mRNA delivery remains a significant challenge due to rapid clearance, endosomal entrapment, and nonspecific tissue distribution. This study explores magnetic lipid nanoparticles (mLNPs) as an advanced platform for in vivo mRNA delivery. By incorporating an externally applied magnetic field to the heart and lung regions, the impact of mLNPs on the tiss

Lipid nanoparticles with aptamers enable targeted mRNA delivery to CD4⁺ T cells.

In vivo genetic engineering of T cells could overcome the logistical, biological, and safety challenges of ex vivo modification, but effective and safe delivery systems remain limited by a lack of cellular specificity. Here, we developed aptamer-functionalized lipid nanoparticles (LNPs) for targeted mRNA delivery to CD4 + T cells, employing both a validated CD4-binding aptamer (Apt62) and novel aptamers generated using our proprietary transformer-based AI language model, AptaBLE. LNPs formulated with ionizable lipid SM102 or MC3 and conjugated with aptamers at controlled densities were physioc

Impaired adipogenesis and lipid storage capacity in subcutaneous adipose tissue of patients with PMOS.

Women with PMOS (formally termed PCOS) have an overall increased prevalence of metabolic syndrome (MetS) and central obesity. To help determine whether there might be changes in s.c. adipose tissue (SAT) associated with these abnormalities, we performed single-nuclei and scRNA-seq on SAT biopsies from 15 premenopausal PMOS women with signs of insulin resistance and 17 healthy BMI-matched controls. In SAT from PMOS versus control we observed a higher ratio of fibrotic versus insulin sensitive adipocytes and a higher ratio of mesenchymal stem cells (MSCs) to preadipocytes. Further in silico anal

Lipid-nanoparticle-mediated base editing of the trabecular meshwork rescues glaucoma in vivo.

Mutations in MYOC, the most common genetic cause of glaucoma, cause misfolded myocilin to accumulate in the endoplasmic reticulum (ER), leading to trabecular meshwork (TM) dysfunction, elevated intraocular pressure, and progressive vision loss. While gene editing offers curative potential, current delivery methods rely on viral vectors, which are limited by inflammation, off-target effects, and poor translatability. Here, we report a nonviral lipid nanoparticle (LNP) platform that enables selective in vivo delivery of mRNA encoding an adenine base editor and single guide RNA (LNP-ABE) to TM ce

Granulated bovine oocytes represent a lipid rich phenotype with enhanced developmental competence.

Immature bovine cumulus-oocytes complexes (COCs) with several layers of tightly packed cumulus cells and a homogeneous, darkish ooplasm are considered to be of good quality, whereas COCs exhibiting cytoplasmatic granulation are frequently observed but routinely discarded. This study compared the protein and lipid landscape, as well as the developmental competence, of immature cattle oocytes with either granulated or homogeneous ooplasm but similarly compact cumulus investment. Nile Red staining revealed increased lipid droplet accumulation, greater heterogeneity in droplet size, and formation

Counterion-Engineered Redox-Active Lipid Cubic Phases for Stable Enzyme Bioelectrodes.

The fabrication of stable and highly performing enzyme-based electrodes is key for the effective generation of biodevices and bioelectronics, such as electrochemical biosensors. In this context, redox-active lyotropic liquid crystals based on 3D nanomaterials, known as lipid cubic phases (LCP), hold great potential due to the large specific surface area and the possibility of being functionalized. In this study, we functionalized a monoolein (MO) LCP matrix by incorporating an amphiphilic redox shuttle within its matrix with the aim of enhancing the electrochemical performance of a glucose oxi
