Naturally plant-derived compounds: role in bone anabolism.

From a nutritional point of view, several factors are involved in ensuring optimal bone health. The most documented of these are calcium and vitamin D. However, it is now well acknowledged that some phytochemicals, also known as phytonutrients, which are plant-based compounds that are present in our daily diet, can positively regulate a number of physiological functions in mammalian systems involved in chronic diseases such as osteoporosis. Indeed, emerging data in animal models of postmenopausal osteoporosis has shown that exposure to some of these naturally plant-derived compounds (e.g. flav

The emerging role of IL-17 in the pathogenesis of psoriasis: preclinical and clinical findings.

Although the histological changes seen in psoriasis have long been well characterized, the underlying cellular and molecular mechanisms have only begun to be elucidated over the past 20 years. Proinflammatory factors such as tumor necrosis factor (TNF)-α have a central role in psoriasis pathogenesis, and many T-helper 1 (Th1) cytokines and messenger RNAs are elevated in psoriatic lesions. IL-17A, IL-17F, and other Th17 cell-derived cytokines have been shown in murine models to induce features that mimic human psoriasis. This review focuses on the emerging biology of the IL-17 cytokine family i

Hypoxia and muscle maintenance regulation: implications for chronic respiratory disease.

PURPOSE OF REVIEW: Muscle wasting and impaired muscle oxidative metabolism are common extrapulmonary features of chronic respiratory failure (CRF) that significantly increase disease burden. This review aims to address the question whether hypoxia, an obvious consequence of this disease, actually plays a causal role in these muscle impairments. RECENT FINDINGS: In experimental models, a causal role for hypoxia in muscle atrophy and metabolic impairments has clearly been shown. Although the hypoxia-inducible factors and nuclear factor kappa B are putative mediators of these hypoxia-induced alt

Analysis of arthritic lesions in the Del1 mouse: a model for osteoarthritis.

Osteoarthritis (OA) is characterised by progressive erosion of articular cartilage with a number of associated degenerative processes within the joint. Animal models of OA provide the only feasible way to systematically study the development and progression of OA, in order to understand the molecular events, and to develop tools for prevention and therapy of OA. Gene manipulation techniques have provided opportunities to generate transgenic mouse models for OA. In heterozygous Dell mice, incorporation of Col2a1 transgenes with a short deletion mutation results in production of shortened proalp

The link between low-LET dose-response relations and the underlying kinetics of damage production/repair/misrepair.

PURPOSE: To review current opinion on the production and temporal evolution of low-LET radiobiological damage. METHODS: Standard cell survival models which model repair/misrepair kinetics in order to quantify dose-response relations and dose-protraction effects are reviewed and interrelated. Extensions of the models to endpoints other than cell survival, to multiple or compound damage processing pathways, and to stochastic intercellular damage fluctuations are surveyed. Various molecular mechanisms are considered, including double strand breaks restitution and binary misrepair. CONCLUSIONS:

Aqueous extract of Solanum nigrum L. restrains colorectal cancer progression by blocking cancer-associated fibroblast-induced STAT3-Annexin A2 signaling.

BACKGROUND: Colorectal cancer (CRC) remains a significant clinical challenge, with progression largely driven by tumor microenvironment (TME) remodeling. Cancer-associated fibroblasts (CAFs), key stromal components, promote CRC progression and metastasis via paracrine signaling. Solanum nigrum L. (SNL), a traditional medicinal plant rich in steroidal glycoalkaloids, is recognized for its health-promoting potential. PURPOSE: This study aimed to systematically investigate the bioactive chemical constituents of an aqueous SNL extract, evaluate its biological effects in multiple CRC models, and l

Automated full-process pipeline via multi-parametric MRI for tumor segmentation, molecular subtype classification and prognostic factor analysis in breast cancer.

BACKGROUND: Accurate identification of molecular subtypes and prognostic factors is crucial for personalized breast cancer management. Multi-parametric magnetic resonance imaging (mp-MRI) offers comprehensive, non-invasive characterization of tumors by capturing complementary information on morphology, perfusion, and tissue microstructure, but conventional radiologic assessment is often inadequate for reliably inferring molecular subtype or prognostic factors. Existing deep learning (DL) approaches depend on manually delineated tumor regions to define inputs, which is difficult to scale in rou

Alternative splicing-based therapeutics for neurodegenerative diseases: a dual-database bibliometric and NLP-driven analysis (2000-2025).

BACKGROUND: Neurodegenerative diseases (NDDs) are driven by complex molecular dysregulation, among which aberrant alternative splicing (AS) has emerged as a critical pathogenic mechanism. Despite the rapid development of splicing-targeted therapeutics, including antisense oligonucleotides (ASOs) and small molecules, comprehensive big-data syntheses mapping this translational landscape remain scarce. This study systematically analyzes the global research trends, conceptual frameworks, and clinical evolution of AS-based therapeutics for NDDs using an integrated bibliometric and natural language

Identifying Therapeutic Targets for Amyotrophic Lateral Sclerosis Through Modeling of Multi-Omics Data.

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease that primarily affects motor neurons, leading to loss of muscle control, and, ultimately, respiratory failure and death. Despite some advances in recent years, the underlying genetic and molecular mechanisms of ALS remain largely elusive. In this respect, a better understanding of these mechanisms is needed to identify new and biologically relevant therapeutic targets that could be developed into treatments that are truly disease-modifying, in that they address the underlying causes rather than the symptoms of ALS.

Baicalin affects the progression of diabetic retinopathy through the RAGE/PXDN/PI3K/AKT pathway.

BACKGROUND: Diabetic retinopathy (DR) is the leading cause of blindness-related eye diseases in adults. The pathogenesis of DR is attributed to the excessive proliferation of microvessels, which leads to vitreous haemorrhage and retinal traction, thereby significantly damaging the patient's vision. In this study, we investigated the role and molecular mechanism of Peroxidasin (PXDN) and the active ingredient baicalin (BAI) of traditional Chinese medicine in regulating the senescence and pathological angiogenesis of human retinal microvascular endothelial cells (HRMECs) to identify the key gene

A practical post-Hartree-Fock approach describing open-shell metal cluster-support interactions. Application to Cu3 adsorption on benzene/coronene.

Current advances in synthesizing and characterizing atomically precise monodisperse metal clusters (AMCs) at the subnanometer scale have opened up fascinating possibilities in designing new heterogeneous (photo)catalysts as well as functional interfaces between AMCs and biologically relevant molecules. Understanding the nature of AMC-support interactions at molecular-level is essential for optimizing (photo)catalysts performance and designing novel ones with improved properties. Møller-Plesset second-order perturbation theory (MP2) is one of the most cost-efficient single-reference post-Hartre

Sampling is decisive to determination of Leishmania (Viannia) species.

BACKGROUND: Accuracy of molecular tools for the identification of parasites that cause human cutaneous leishmaniasis (CL) could largely depend on the sampling method. Non-invasive or less-invasive sampling methods such as filter paper imprints and cotton swabs are preferred over punch biopsies and lancet scrapings for detection methods of Leishmania based on polymerase chain reaction (PCR) because they are painless, simple, and inexpensive, and of benefit to military and civilian patients to ensure timely treatment. However, different types of samples can generate false negatives and there is

A fetal wound healing program after intrauterine bile duct injury may contribute to biliary atresia.

BACKGROUND & AIMS: Biliary atresia (BA) is an obstructive cholangiopathy that initially affects the extrahepatic bile ducts (EHBDs) of neonates. The etiology is uncertain, but evidence points to a prenatal cause. Fetal tissues have increased levels of hyaluronic acid (HA), which plays an integral role in fetal wound healing. The objective of this study was to determine whether a program of fetal wound healing is part of the response to fetal EHBD injury. METHODS: Mouse, rat, sheep, and human EHBD samples were studied at different developmental time points. Models included a fetal sheep model

A Sustained Immune Response Supports Long-Term Antiviral Immune Priming in the Pacific Oyster, Crassostrea gigas.

Over the last decade, innate immune priming has been evidenced in many invertebrate phyla. If mechanistic models have been proposed, molecular studies aiming to substantiate these models have remained scarce. We reveal here the transcriptional signature associated with immune priming in the oyster Crassostrea gigas Oysters were fully protected against Ostreid herpesvirus 1 (OsHV-1), a major oyster pathogen, after priming with poly(I·C), which mimics viral double-stranded RNA. Global analysis through RNA sequencing of oyster and viral genes after immune priming and viral infection revealed that
