Involvement of LTD(4)in allergic pulmonary inflammation in mice: modulation by cysLT(1)antagonist MK-571.

Cysteinyl leukotrienes are potent inflammatory molecules playing a major role in asthma. The involvement of these mediators in hypersensitivity in mice is not well known. This study aimed at elucidating their implication by using MK-571, a cysLT(1)receptor antagonist. Mice were sensitized with a suspension of ovalbumin (8 microg) adsorbed to alum (2 mg) and were challenged with an aerosolized ovalbumin solution (0.5%). Inflammatory cell infiltration in the bronchoalveolar lavage (mostly eosinophils) following antigen challenge was inhibited by dexamethasone (0.1, 1 and 5 mg kg(-1)s.c.) and MK-

Inhibition of Dectin-1 alleviates inflammation in early diabetic retinopathy by regulating microglia phenotype.

BACKGROUND: Diabetic retinopathy (DR) is a major factor in vision loss in diabetic patients, triggering a series of pathological changes. At present, the treatment methods for diabetic retinopathy are limited. There is an urgent need to further explore its mechanism to bring more treatment options to patients. There is increasing evidence that microglia activation plays a crucial role in inflammatory DR. The C type lectin receptor Dectin-1 is known to play an important role in the inflammatory regulation of microglia, however, its role and mechanism in DR remains unclear. This study aims to elu

[The role of vitamin D in the carcinogenesis of breast and ovarian cancer].

The review evaluates the role of vitamin D in carcinogenesis. Based on ecological studies, the incidence of many cancers has been shown to be higher in northern countries, suggesting an association with latitude and solar radiation. Vitamin D produced in skin under the influence of sun exposure may play a protective role in the process leading to cancer. Vitamin D deficiency is now recognized as a pandemic, mainly due to lack of knowledge that sun exposure in moderation is the major source of vitamin D for most humans. After vitamin D was discovered to be the necessary element of nourishment t

Mechanistic insights on lignin-mediated plant defense against pathogen infection.

Lignin, a pivotal constituent of plant secondary cell walls, plays a versatile role as a structural and biochemical defense compound against biotic stress. This phenolic biopolymer is integral in defending plants and neutralizing invading pathogens, including necrotrophs, biotrophs, and hemibiotrophs. Lignin monomer units vary from plant to plant and it has been observed that this variability plays a crucial role in the resistance and susceptibility of a particular plant type. The complex network and cross-linking of lignin units have been observed to disintegrate under the influence of a fung

Recent advances in small molecule Nav 1.7 inhibitors for cancer pain management.

The dorsal root ganglion (DRG) is the primary neuron responsible for transmitting peripheral pain signals to the central nervous system and plays a crucial role in pain transduction. Modulation of DRG excitability is considered a viable approach for pain management. Neuronal excitability is intricately linked to the ion channels on the neurons. The small and medium-sized DRG neurons are chiefly engaged in pain conduction and have high levels of TTX-S sodium channels, with Nav1.7 accounting for approximately 80% of the current. Voltage-gated sodium channel (VGSC or Nav) blockers are vital targe

The zinc finger domain of IKKγ (NEMO) protein in health and disease.

Inhibitor of κB kinase (IKK) gamma (IKKγ), also known as nuclear factor κB (NF-κB) essential modulator (NEMO), is a component of the IKK complex that is essential for the activation of the NF-κB pathway. The NF-κB pathway plays a major role in the regulation of the expression of genes that are involved in immune response, inflammation, cell adhesion, cell survival and development. As part of the IKK complex, IKKγ plays a regulatory role by linking the complex to upstream signalling molecules. IKKγ contains two coiled-coil regions, a leucine zipper domain and a highly conserved zinc finger doma

[Liver-bone axis: novel mechanisms and strategies for MAFLD regulation].

Bones not only play an important role in providing structural support in the human body but also play a critical role in regulating the pathophysiological processes of various organs by secreting specific cytokines (osteokines). Growing evidence confirms the existence of extensive and close interaction between the liver and bone, known as the liver-bone axis. The regulatory role of the liver-bone axis in chronic liver diseases offers a novel research concept, particularly for understanding the complex metabolic regulatory network of metabolic dysfunction-associated fatty liver disease (MAFLD).

The role of trauma in low back pain: a review.

Trauma plays an important role in the production of low back pain. This review emphasizes how the forces to which the spine is subjected are modified by many factors. Attention is directed to the role of mechanical stress in the etiology of spondylolisthesis, subtle spinal fractures, as well as the relevance of stress to the degenerative process. It is suggested that trauma plays a very major role in low back pain but, at present, the problems in diagnostic technology create difficulty in establishing the precise degree of that role. The drawbacks of over-reliance on plane radiography are emph

Viral Infection in Endometritis: Is There an Important Role or Not?

Chronic endometritis (CE) is a frequent pathological condition that is defined as localized inflammation in the endometrium. Some adverse fertility consequences such as recurrent miscarriage and failure of implantation are associated with chronic endometritis. On the one hand, inflammation plays an important role in the pathogenesis of endometritis, and on the other hand, the role of viral infections in inducing inflammation can make this review strongly attractive and practical. We set out to provide an overview of viral infections as a potential etiology of CE pathophysiology through the alt

A Survey of the United Kink-dom: Investigating Five Paraphilic Interest Groups and Their Demographic and Psychological Correlates.

We explored demographic, psychological, and behavioral characteristics of five paraphilic interest groups in the UK: BDSM, pet play, age play/ABDL, furries, and balloon fetishists - chosen for their prevalence, power dynamics, and varied activities. Through an anonymous survey (N = 470), we assessed role identities, engagement patterns, and relational dynamics. Groups were more likely to have non-heterosexual, non-monogamous, and non-cisgender identities, with high levels of co-occurrence across paraphilic interests. Participants were predominantly cisgender men, though over 25% identified as

The influence of BACE1 on macrophage recruitment and activity in the injured peripheral nerve.

Following peripheral nerve injury, multiple cell types, including axons, Schwann cells, and macrophages, coordinate to promote nerve regeneration. However, this capacity for repair is limited, particularly in older populations, and current treatments are insufficient. A critical component of the regeneration response is the network of cell-to-cell signaling in the injured nerve microenvironment. Sheddases are expressed in the peripheral nerve and play a role in the regulation if this cell-to-cell signaling through cleavage of transmembrane proteins, enabling the regulation of multiple pathways

Research Progress on the Role of Ubiquitination in Eye Diseases.

The occurrence and development of ophthalmic diseases are related to the dysfunction of eye tissues. Ubiquitin is an important form of protein post-translational modification, which plays an essential role in the occurrence and development of diseases through specific modification of target proteins. Ubiquitination governs a variety of intracellular signal transduction processes, including proteasome degradation, DNA damage repair, and cell cycle progression. Studies have found that ubiquitin can play a role in eye diseases such as cataracts, glaucoma, keratopathy, retinopathy, and eye tumors.

Diuretics in the management of chronic heart failure: when and how.

Heart failure is an increasingly prevalent condition resulting in recurrent hospitalisations and significant mortality and morbidity. The management of heart failure has evolved, and multiple drugs have an established mortality benefit in heart failure with reduced ejection fraction. Although the focus should be on ensuring that patients are treated with the maximum tolerated doses of these guideline-directed therapies, diuretics continue to play a key role in the management of clinical congestion in all forms of heart failure. Clinicians play a key role in heart failure management. Familiarit

Interaction of low-density neutrophils with other immune cells in the mechanism of inflammation.

Low-density neutrophils (LDNs) are a unique subpopulation of neutrophils, play a significant role in regulating innate and adaptive immunity in various inflammation-related diseases. Emerging evidence suggests that LDNs play a significant role in the pathogenesis and progression of various diseases, including infections, autoimmune disorders, and cancer. In this review, we address the origin, development, and heterogeneity of LDNs, and the roles of LDNs in system homeostasis and diseases. We will focus on the findings of the interaction between LDNs and other immune cells. We will then discuss

Central mechanisms of fever.

The possible role of various potential chemical mediators in the production of fever is reviewed. A major problem in this field is the very considerable conflict of evidence, let alone interpretation. On the existing evidence, it appears unlikely that monoamines, acetyl choline, or alterations in relative concentrations of sodium and calcium play any major role in the production of fever. Recent evidence makes it unlikely that prostaglandins have a direct role in this mechanism, though the involvement of other metabolites of arachidonic acid has not been excluded. It is possible that protein s

The effect of a training program on maternal role adaptation and self-esteem of mothers with preterm infants: a quasi-experimental study.

BACKGROUND: Admission of preterm infants in the neonatal intensive care unit limits the mother's interaction with their infants, delaying accepting and playing the motherhood role. Besides, mothers of preterm infants have low self-esteem due to their infants' condition. Accordingly, the present study explored the effect of implementing the training program on maternal role adaptation and self-esteem of mothers of preterm infants admitted to the neonatal intensive care unit. METHODS: This study employed a quasi-experimental design with two groups. The participants were 80 mothers of preterm in

Direct Intercellular Communications and Cancer: A Snapshot of the Biological Roles of Connexins in Prostate Cancer.

Tissue homeostasis is the result of a complex intercellular network controlling the behavior of every cell for the survival of the whole organism. In mammalian tissues, cells do communicate via diverse long- and short-range communication mechanisms. While long-range communication involves hormones through blood circulation and neural transmission, short-range communication mechanisms include either paracrine diffusible factors or direct interactions (e.g., gap junctions, intercellular bridges and tunneling nanotubes) or a mixture of both (e.g., exosomes). Tumor growth represents an alteration
