Early Response-Based Therapy Stratification Improves Survival in Adult Early Thymic Precursor Acute Lymphoblastic Leukemia: A Group for Research on Adult Acute Lymphoblastic Leukemia Study.

Purpose Early thymic precursor (ETP) acute lymphoblastic leukemia (ALL) is an immunophenotypically defined subgroup of T-cell ALL (T-ALL) associated with high rates of intrinsic treatment resistance. Studies in children have shown that the negative prognostic impact of chemotherapy resistance is abrogated by the implementation of early response-based intensification strategies. Comparable data in adults are lacking. Patients and Methods We performed comprehensive clinicobiologic, genetic, and survival analyses of a large cohort of 213 adult patients with T-ALL, including 47 patients with ETP-A

Nodular lymphocyte-predominant Hodgkin lymphoma: a unique disease deserving unique management.

Nodular lymphocyte-predominant Hodgkin lymphoma (NLPHL) is a rare lymphoma entity with an incidence of 0.1 to 0.2/100 000/y. Compared with the more common subtypes of classical Hodgkin lymphoma, NLPHL is characterized by distinct pathological and clinical features. Histologically, the disease-defining lymphocyte predominant cells consistently express CD20 but lack CD30. Clinically, NLPHL mostly has a rather indolent course, and patients usually are diagnosed in early stages. The prognosis of early-stage NLPHL is excellent, with progression-free survival and overall survival rates exceeding 90%

Parathyroid hormone stimulates circulating osteogenic cells in hypoparathyroidism.

CONTEXT: The osteoanabolic properties of PTH may be due to increases in the number and maturity of circulating osteogenic cells. Hypoparathyroidism is a useful clinical model because this hypothesis can be tested by administering PTH. OBJECTIVE: The objective of the study was to characterize circulating osteogenic cells in hypoparathyroid subjects during 12 months of PTH (1-84) administration. DESIGN: Osteogenic cells were characterized using flow cytometry and antibodies against osteocalcin, an osteoblast-specific protein product, and stem cell markers CD34 and CD146. Changes in bone format

Revisiting hemostasis strategy: the impact of excessive monopolar electrocautery on periosteal-driven spinal fusion.

BACKGROUND CONTEXT: The common surgical approach in posterior spinal fusion (PSF) conflicts with basic principles of bone repair and may lead to high failure rates. In recent years, experimental data has shown that the periosteum plays an indispensable role in fracture bone repair both for skeletal stem/progenitor cells (SSPCs) and for angiogenesis promotion. However, the role of the periosteum in spinal fusion remains experimentally poorly defined. Furthermore, current surgical approach often involves electrocautery to achieve subperiosteal dissection. However, the impact of this electrocaute

Incidence and Outcomes of Neutropenic Enterocolitis Post-stem Cell Transplant for Hematological Malignancies: A Retrospective Study in Australia.

Neutropenic enterocolitis (NE) is a well-known occurrence in patients who receive chemotherapy and stem cell transplantation for hematological malignancies. Nonetheless, there is a lack of Australian research data available on this life-threatening condition. The purpose of this study is to determine the percentage of patients with multiple myeloma and lymphoma who develop NE after autologous stem cell transplant (ASCT) in an Australian setting and to examine the mortality rates in the selected group. Retrospective data were collected from a small pilot group of patients (N = 33) at a private

Extracellular vesicles secreted by bone marrow stem cells mediate angiogenesis for the treatment of diabetic ulcers: A systematic review and meta-analysis of preclinical studies.

BACKGROUND: Diabetic ulcers (DUs) typically occur in patients with vascular diseases and diabetes. Extracellular vesicles secreted by bone marrow-derived stem cells (BMSC-EVs) represent a cell-free therapy that has emerged as a promising alternative for treating DU, especially due to significant advancements in the understanding of their role in promoting angiogenesis; however, their application in DU treatment remains in the preclinical stage, and their effectiveness is still uncertain. Therefore, we conducted this meta-analysis to evaluate the therapeutic efficacy of BMSC-EVs in treating DU

Engineered Nanotechnology: An Effective Therapeutic Platform for the Chronic Cutaneous Wound.

The healing of chronic wound infections, especially cutaneous wounds, involves a complex cascade of events demanding mutual interaction between immunity and other natural host processes. Wound infections are caused by the consortia of microbial species that keep on proliferating and produce various types of virulence factors that cause the development of chronic infections. The mono- or polymicrobial nature of surface wound infections is best characterized by its ability to form biofilm that renders antimicrobial resistance to commonly administered drugs due to poor biofilm matrix permeability

Regulation of the Maturity of Dental Pulp-Derived Induced Neural Cells by Cell-Recognizing Cadherin-Fc Chimeric Antibody Improved Neurological Function after Cell Implantation in a Mouse Traumatic Brain Injury Model.

Biomaterial-based neural cell transplantation has displayed promise in regenerative medicine. E-cadherin and N-cadherin are cell-cell adhesion proteins with important roles in neuronal maturity. Therefore, cell culture on Petri dishes coated with a fusion protein combining the human E-cadherin or N-cadherin extracellular domain with an immunoglobulin G Fc region chimeric antibody (E-cad-Fc and N-cad-Fc) can regulate cell maturity through the process of neuronal differentiation in stem cells. This study explored the efficacy of dental pulp-derived induced neural cells (DPiNCs), which were cultu

Impact of Medications for Stable Angina Pectoris on Osteoporosis: A Review of Current Evidence.

Stable angina pectoris (SAP) and osteoporosis (OP) are both prevalent conditions among the elderly population. Compared to SAP, the prevention and management of OP are often neglected. Furthermore, certain medications used long-term for SAP may exert significant effects on bone metabolism. This review summarizes the impact of commonly prescribed SAP medications on OP. Extensive research indicates that nitrates not only promote vascular and osteogenic coupling via the nitric oxide (NO)-cyclic guanosine monophosphate (cGMP)-protein kinase G (PKG) pathway, enhancing the osteogenic effects of estr

A Systematic Review on Amnion as a Cell Delivery Scaffolding Material for Cartilage Regeneration in Pre-Clinical and Clinical Studies.

Cartilage is an important yet vulnerable tissue with limited self-healing capacity, where damage often progresses to joint degeneration, which eventually leads to severe osteoarthritis (OA). Current tissue engineering strategies focus on biocompatible scaffolds for cartilage regeneration, particularly amnion (or amniotic membrane), emerging as a promising biomaterial due to its wide availability, low immunogenicity, and naturally derived microenvironment that is advantageous for cartilage regeneration. This systematic review aims to evaluate the existing evidence on the efficacy of amnion as a

Effects of imidazole derivatives on cellular proliferation and apoptosis in myeloid leukemia.

BACKGROUND: Acute promyelocytic leukemia (APL) is the sub-type of Acute myeloid leukemia (AML) which is described by differentiation block at promyelocytic stage and t(15; 17) translocation with All trans retinoic acid (ATRA) and arsenic trioxide (ATO) as standard treatments. Chronic myeloid leukemia (CML) translocation t (19; 22) causes a rise in granulocytes and their immature precursors in the blood. Different mutations cause resistance to first-line tyrosine kinase therapies in CML. Beside drug resistance, leukemia stem cells (LSC) are critical resources for relapse and resistance in APL a

Human amnion cells for the prevention of bronchopulmonary dysplasia: a protocol for a phase I dose escalation study.

INTRODUCTION: Bronchopulmonary dysplasia (BPD), an important sequela of preterm birth, is associated with long-term abnormalities of lung function and adverse neurodevelopmental outcomes. Inflammation, inhibition of secondary septation and vascular maldevelopment play key roles in the pathogenesis of BPD. Human amnion epithelial cells (hAECs), stem-like cells, derived from placental tissues are able to modulate the inflammatory milieu and, in preclinical studies of BPD-like injury, restore lung architecture and function. Allogeneic hAECs may present a new preventative and reparative therapy fo

Teriparatide (PTH 1-34) treatment increases peripheral hematopoietic stem cells in postmenopausal women.

Cells of the osteoblast lineage play an important role in regulating the hematopoietic stem cell (HSC) niche and early B-cell development in animal models, perhaps via parathyroid hormone (PTH)-dependent mechanisms. There are few human clinical studies investigating this phenomenon. We studied the impact of long-term daily teriparatide (PTH 1-34) treatment on cells of the hematopoietic lineage in postmenopausal women. Twenty-three postmenopausal women at high risk of fracture received teriparatide 20 mcg sc daily for 24 months as part of a prospective longitudinal trial. Whole blood measuremen

Artificial intelligence virtual bone organoids (AIVBOs).

UNLABELLED: Global population aging has made skeletal disorders a major threat to human health. Solid bone organoids reproduce skeletal physiology and pathology more faithfully than traditional models, yet remain limited in standardisation, real-time monitoring, and high-throughput use. Integrating artificial intelligence (AI) with computational biology offers a route to ease these limits and motivates an emerging direction we develop here. We use virtual bone organoids (VBOs) for the in silico counterpart of a solid bone organoid, and artificial intelligence virtual bone organoids (AIVBOs) fo

Chemotherapy-Induced Cardiotoxicity: A Comprehensive Review of Current Understanding and Future Directions.

INTRODUCTION: Chemotherapy-Induced Cardiotoxicity (CIC) has become a growing concern in modern oncology. While chemotherapy remains one of the most effective treatments against cancer, its potential to harm the heart often limits its use. The underlying mechanisms involve oxidative stress, mitochondrial dysfunction, and inflammation, all contributing to the gradual weakening of cardiac muscle. As cancer survival improves, the burden of cardiovascular complications is more visible than ever, emphasizing the need for early recognition, prevention, and long-term management. METHODS: This review

ADSC-Exo-bFGF promotes skin wound healing and correlates with the PI3K/AKT signaling pathway activation.

Wound healing is a complex physiological process involving a variety of tissue repair cells. Adipose-derived stem cell (ADSC)-derived exosomes have demonstrated potential in promoting wound healing. Basic fibroblast growth factor (bFGF) is known to facilitate tissue regeneration. Nevertheless, the biological functions and detailed mechanisms of ADSC-Exo-bFGF in cutaneous wound healing remain largely unelucidated. The study aimed to construct a bFGF-glycosylphosphatidylinositol (GPI)-anchored exosome system (ADSC-Exo-bFGF) and further explore its therapeutic efficacy and underlying molecular me

Human cell-based models for urea cycle research: From gene regulation and ureagenesis to disease modeling and therapeutic development.

Development of cell-based model systems for studying the urea cycle has been motivated by the urgent need to develop and evaluate treatments for urea cycle disorders, such as gene therapies currently in clinical trials, and to conduct mechanistic studies of the regulation of the rate of ureagenesis and urea cycle gene expression. However, studying the urea cycle in cellular models has historically been challenging. Traditional in vitro models, primarily 2D cell cultures, often fail to fully recapitulate the complex physiological environment and metabolic zonation necessary for accurate urea cy

Diagnosis and Management of Secondary HLH/MAS Following HSCT and CAR-T Cell Therapy in Adults; A Review of the Literature and a Survey of Practice Within EBMT Centres on Behalf of the Autoimmune Diseases Working Party (ADWP) and Transplant Complications Working Party (TCWP).

Introduction: Secondary haemophagocytic lymphohistiocytosis (sHLH) or Macrophage Activation Syndrome (MAS) is a life-threatening hyperinflammatory syndrome that can occur in patients with severe infections, malignancy or autoimmune diseases. It is also a rare complication of haematopoetic stem cell transplantation (HSCT), with a high mortality. It may be associated with graft vs. host disease in the allogeneic HSCT setting. It is also reported following CAR-T cell therapy, but differentiation from cytokine release syndrome (CRS) is challenging. Here, we summarise the literature and present res
