Serotonin and brain development.

The role of the serotonergic system in the neuroplastic events that create, repair, and degenerate the brain has been explored. Synaptic plasticity occurs throughout life and is critical during brain development. Evidence from biochemical, pharmacological, and clinical studies demonstrates the huge importance of an intact serotonergic system for normal central nervous system (CNS)function. Serotonin acts as a growth factor during embryogenesis, and serotonin receptor activity forms a crucial part of the cascade of events leading to changes in brain structure. The serotonergic system interacts

Pineal metabolic reaction to retinal photostimulation in ganglionectomized rats.

The aim of the present work was to test the pineal gland metabolic reactivity to nocturnal retinal short term photic stimulation in superior … acetylserotonin and an increase of the amount of serotonin, 5-hydroxyindoleacetic acid and 5-hydroxytryptophan. This inhibitory process brought into action by retinal illumination is dependent on the direct central neural connections to the pineal gland, since rats that were lesioned in the DP, previously to ganglionectomy, did not show any alteration on the …

Interaction of angiotensin II with the cholinergic and noradrenergic systems in the rat pineal gland: regulation of indole metabolism.

The pineal gland, angiotensin, and noradrenergic and cholinergic systems are involved in the regulation of tissue indole metabolism. Angiotensin II increased noradrenaline release and the production of hydroxy- and methoxyindoles by pineal slices. Electrical field stimulation (EFS) of pineal slices released angiotensin II and reproduced many of the actions of exogenous angiotensin II on serotonin and melatonin biosynthesis and release. Both sarcosine-isoleucine-angiotensin II ([Sar, Ile]-ANG II) and atropine blocked, and nadolol increased, the effect of EFS and exogenous angiotensin II on sero

Localisation of esteroproteases in 'resting' salivary glands from different species and the effects of the organophosphorus inhibitor E600.

The distribution of reaction sites for esteroproteases has been assessed in 'resting' salivary glands from rats, guinea-pigs, cats, dogs and … se' is discussed. Ductal activity in rats, cats and guinea-pigs had some relationship with sites of tryptophan staining but not in rabbits or dogs. Any relationship with acinar staining was less evident. Mast cells, showing strong esteroprotease staining, were present in variable numbers in the different glands. Preliminary attempts to qualify the enzymes be …

The Gut-Brain Axis and Neurodegenerative Diseases: From Mechanistic Pathways to Therapeutic Interventions.

As the global population ages, the incidence of neurodegenerative diseases (NDDs) has increased. Neurodegenerative diseases have complex pat … ferences in gut microbiota metabolites (such as short-chain fatty acids (SCFAs), neurotransmitters, tryptophan metabolites, etc.). These changes influence the CNS through multiple pathways, including neuroendocrine, vagus nerve, and immune regulation, ultimately contributing to the onset and progression of the diseases. Furthermore, we have collected and sum …

Microbiota-Brain Interactions in Dementia: Role of Diet in Neuroinflammation.

Aging is one of the risk factors for dementia, a group of neurocognitive disorders characterized by a progressive decline in cognitive funct … ier (BBB). Short-chain fatty acids (SCFAs), such as butyrate, have anti-inflammatory effects, while tryptophan derivatives, such as indole-3-acetic acid (IAA), support intestinal barrier function and modulate neuroinflammatory pathways. Microbial-derived metabolites may serve as potential therapeutic targets to mitigate neurodegeneration, highlighting the in …

Expression, Purification, and Characterization of Recombinant Isocitrate Dehydrogenase (IDH) from Yarrowia lipolytica and its Application as a Protein Template for In Situ Synthesis of Gold Nanoparticles.

In this study, isocitrate dehydrogenase (IDH) was employed exclusively as a structural protein template for the synthesis of gold nanoparticles, rather than relying on its enzymatic catalytic function. The reduction of Au³⁺ to Au⁰ is accomplished through the intrinsic reducing capability of amino acid residues (particularly tryptophan and tyrosine) within the protein scaffold, which simultaneously acts as a capping and stabilizing agent. This protein-templating strategy provides a straightforward, biocompatible, and cofactor-free approach for developing AuNP-based biosensing platforms. So, the…

Intrinsic Fluorescence as a Probe of Structural Dynamics and Interactions of Human Adenovirus with Surfactants and Liposomes.

In this work, the intrinsic fluorescence of Human Adenovirus serotype 2 (HAdV-2), arising from tryptophan (Trp) and tyrosine (Tyr) residues present in viral proteins, was used to monitor structural changes in the three-dimensional organization of the viral particle as a function of the inactivation method employed (UV irradiation or paraformaldehyde (PFA) treatment), temperature, and the presence of negatively and positively charged surfactants. Steady-state fluorescence anisotropy was used to investigate the interaction between HAdV-2 and zwitterionic or negatively charged liposomes by monito…

Bifidobacterium longum alleviation of metabolic dysfunction-associated steatotic liver disease: A multi-omics landscape of microbiota and metabolome reconfiguration.

The gut microbiome-host metabolism axis plays a critical role in the pathogenesis of metabolic dysfunction-associated steatotic liver diseas … ntrinsically linked to a reconfigured serum metabolome, highlighted by the enrichment of protective tryptophan-derived metabolites (e.g., indole-3-propionic acid) and the reduction of detrimental ones (e.g., 17α-methyltestosterone, 7-HDoHE). Collectively, our results suggest that B. longum mitigates MASLD through modulation of the gut microbiota and host ser …
