Differential effects of sound repetition rate on auditory cortex development and behavior in fragile X syndrome mouse model.

Fragile X syndrome (FXS) is a leading genetic form of autism and intellectual disability that is associated with a loss-of-function mutation in the Fragile X messenger ribonucleoprotein 1 (Fmr1) gene. The Fmr1 knockout (KO) mouse model displays many aspects of FXS-related phenotypes and is used to study FXS pathophysiology. Sensory manipulations, such as sound exposure, are considered as a non-invasive approach to alleviate FXS phenotypes. However, it is unclear what specific sound attributes may have beneficial effects. In this study, we examined the effects of sound repetition rate on audito

Whole-ear finite element analysis of superior semicircular canal dehiscence and its impact on inner-ear responses.

OBJECTIVE: To establish a high-fidelity finite element method (FEM) model of the human inner ear and explore the biomechanical effects of superior semicircular canal dehiscence (SCD) on both cochlear and vestibular function. METHODS: A detailed FEM model of the entire human ear was reconstructed from high-resolution computed tomography (CT) data. The model was validated through comparison with established experimental data, including basilar membrane (BM) displacement patterns, cochlear tonotopy, inner ear impedance, and middle-ear transfer function. After validation, the model was adapted to

Prevention and treatment of noise-induced hearing loss and cochlear synapse degeneration by potassium channel blockers in vivo.

Noise can induce hearing loss. In particular, noise can induce cochlear synapse degeneration leading to hidden hearing loss, which is the most common type of hearing disorders in the clinic. Currently, there is no pharmacological treatment, especially no post-exposure (i.e., therapeutic) treatment available in the clinic. Here, we report that systemic administration of K+ channel blockers by intraperitoneal injection before or after noise exposure significantly ameliorated noise-induced hearing loss (NIHL) and cochlear synapse degeneration. After systemic administration of a general K+-channel

Integrated stress response inhibition provides sex-dependent protection against noise-induced cochlear synaptopathy.

Noise-induced hearing loss (NIHL) is a common health concern with significant social, psychological, and cognitive implications. Moderate levels of acoustic overstimulation associated with tinnitus and impaired speech perception cause cochlear synaptopathy, characterized physiologically by reduction in wave I of the suprathreshold auditory brainstem response (ABR) and reduced number of synapses between sensory hair cells and auditory neurons. The unfolded protein response (UPR), an endoplasmic reticulum stress response pathway, has been implicated in the pathogenesis and treatment of NIHL as w

Kv3 K+ currents contribute to spike-timing in dorsal cochlear nucleus principal cells.

Exposure to loud sound increases burst-firing of dorsal cochlear nucleus (DCN) fusiform cells in the auditory brainstem, which has been suggested to be an electrophysiological correlate of tinnitus. The altered activity of DCN fusiform cells may be due to down-regulation of high voltage-activated (Kv3-like) K+ currents. Whole cell current-clamp recordings were obtained from DCN fusiform cells in brain slices from P15-P18 CBA mice. We first studied whether acoustic over-exposure (performed at P15) or pharmacological inhibition of K+ currents with tetraethylamonium (TEA) affect fusiform cell act

Auditory Spatial Deficits in the Early Stage of Ischemic Cerebral Stroke.

BACKGROUND: Clinical research, together with computed tomography/magnetic resonance imaging findings, proves that ischemic stroke (IS) that damages auditory pathways can cause hearing loss and impairment of higher auditory processes such as sound localization. The goal of the study was to find possible correlations between the IS risk factors, ischemic lesion volume and localization, neurologic status, and the sound localization capability in acute IS patients. METHODS: We consecutively enrolled 61 IS patients into the study. The control group consisted of 60 healthy volunteers. All neuro-oto

A motor learning-based intervention to ameliorate freezing of gait in subjects with Parkinson's disease.

Freezing of gait (FOG) is an episodic gait disturbance that is commonly seen in Parkinson's disease (PD). To date, treatment efficacy is limited. We tested the hypothesis that an intervention that utilizes motor learning provided through intensive cueing can alleviate this symptom. Fifteen subjects with PD suffering from FOG participated in a 6 week progressive motor learning program (three training sessions per week--open trial). A training session included FOG-provoking situations (e.g., turns). Prior to each presumed FOG provocation (e.g., just before a turn), rhythmic auditory stimulation

Early post-stressor intervention with minocycline, a second-generation tetracycline, attenuates post-traumatic stress response in an animal model of PTSD.

We assessed the effects of minocycline, a tetracycline with anti-inflammatory, anti-apoptotic and neuroprotective capacities, in an animal model of post-traumatic stress disorder (PTSD). Rats were exposed to psychogenic stress and treated 1h later with minocycline or saline. Behavioral measures included the elevated plus-maze (EPM) and acoustic startle response (ASR) 7 days post stress-exposure. One day after behavioral testing, animals were exposed to a trauma cue and freezing response was assessed. Local levels of cytokines interleukin-1 (IL-1), interleukin-6 (IL-6) and tumor necrosis factor

Immediate ketamine treatment does not prevent posttraumatic stress responses in an animal model for PTSD.

Clinical studies suggest that administration of ketamine hydrochloride-an antagonist at the N-methyl-d-aspartate ionophore-provides short-term amelioration for depressive symptoms. The effects of a brief course of ketamine given immediately following exposure to psychogenic stress on the behavioral stress responses were assessed in an animal model of posttraumatic stress disorder. Animals exposed to stress were treated 1h later with ketamine (0.5, 5, and 15 mg/kg) or vehicle for three days (N = 107). Outcome measures included behavior in the elevated plus maze (EPM) and acoustic startle respon

Stimulation rate reduction and auditory development in poorly performing cochlear implant users with auditory neuropathy.

OBJECTIVE: Patients with auditory neuropathy spectrum disorder (ANSD) exhibit altered neural synchrony in response to auditory stimuli. It has been hypothesized that a slower rate of electrical stimulation in programming strategies for cochlear implant (CI) users with ANSD may enhance development of neural synchrony and speech perception abilities. STUDY DESIGN: Retrospective case series. SETTING: Tertiary otologic practice. PATIENTS: Twenty-two patients with ANSD underwent CI. Patients with complete postoperative audiometric data and at least 2 years of follow-up were included in further a

Growth of suppression using distortion-product otoacoustic emission measurements in hearing-impaired humans.

Growth of distortion-product otoacoustic emission suppression was measured in 65 subjects with mild-to-moderate sensorineural hearing loss (HI). Measurements were made at four probe frequencies (f(2)) and up to five L(2) levels. Eleven suppressor frequencies (f(3)) were used for each f(2), L(2) combination. These data were compared to data from normal-hearing (NH) subjects (Gorga et al., 2011a). In both NH and HI subjects, growth of suppression depended on the relation between f(2) and f(3), such that the slope was close to one when f(3) ≈ f(2), steeper than one when f(3) < f(2), and shallower

Individual differences in behavioral estimates of cochlear nonlinearities.

Psychophysical methods provide a mechanism to infer the characteristics of basilar membrane responses in humans that cannot be directly measured. Because these behavioral measures are indirect, the interpretation of results depends on several underlying assumptions. Ongoing uncertainty about the suitability of these assumptions and the most appropriate measurement and compression estimation procedures, and unanswered questions regarding the effects of cochlear hearing loss and age on basilar membrane nonlinearities, motivated this experiment. Here, estimates of cochlear nonlinearities using te

The effects of age and cochlear hearing loss on temporal fine structure sensitivity, frequency selectivity, and speech reception in noise.

Temporal fine structure (TFS) sensitivity, frequency selectivity, and speech reception in noise were measured for young normal-hearing (NHY), old normal-hearing (NHO), and hearing-impaired (HI) subjects. Two measures of TFS sensitivity were used: the "TFS-LF test" (interaural phase difference discrimination) and the "TFS2 test" (discrimination of harmonic and frequency-shifted tones). These measures were not significantly correlated with frequency selectivity (after partialing out the effect of audiometric threshold), suggesting that insensitivity to TFS cannot be wholly explained by a broaden

Accuracy of velocity distortion product otoacoustic emissions for estimating mechanically based hearing loss.

Distortion product otoacoustic emissions (DPOAEs) measured as vibration of the human eardrum have been successfully used to estimate hearing threshold. The estimates have proved more accurate than similar methods using sound-pressure DPOAEs. Nevertheless, the estimation accuracy of the new technique might have been influenced by endogenous noise, such as heart beat, breathing and swallowing. Here, we investigate in an animal model to what extent the accuracy of the threshold estimation technique using velocity-DPOAEs might be improved by reducing noise sources. Velocity-DPOAE I/O functions wer

Ensemble spontaneous activity in the guinea-pig cochlear nerve.

Spectral analysis of electrical noise recorded from the round window (RW) of the cochlea is referred to as the ensemble spontaneous activity (ESA) of the cochlear nerve. The ESA is considered to represent the summed spontaneous activity of single fibers of the auditory nerve and changes in the spectral characteristics of the ESA have been observed in humans with tinnitus. Experiments were undertaken to determine the relationship of the ESA to auditory neurotransmission. The ESA consisted of energy centered at approximately 900 Hz, similar to the spectral peak of single auditory neuron discharg

Deficits of spatial and non-spatial memory and of auditory fear conditioning following anterior thalamic lesions in mice: comparison with chronic alcohol consumption.

This study was aimed at determining (i) whether or not bilateral subtotal lesions of the anterior thalamic nuclei (ATH) in rodents produced memory deficits for spatial and/or non-spatial information and of auditory fear conditioning, and (ii) if these eventual deficits resemble those produced by chronic alcohol consumption (CAC). Working memory was assessed using both spatial (spontaneous alternation) and non-spatial (temporal alternation) delayed response tasks. Results showed that ATH lesions induced delay-dependent memory impairments in both spatial and non-spatial alternation tasks, as wel
