Stem cell treatment improves post stroke neurological outcomes: a comparative study in male and female rats.

BACKGROUND AND PURPOSE: The therapeutic potential of different stem cells for ischaemic stroke treatment is intriguing and somewhat controversial. Recent results from our laboratory have demonstrated the potential benefits of human umbilical cord blood-derived mesenchymal stem cells (MSC) in a rodent stroke model. We hypothesised that MSC treatment would effectively promote the recovery of sensory and motor function in both males and females, despite any apparent sex differences in post stroke brain injury. METHODS: Transient focal cerebral ischaemia was induced in adult Sprague-Dawley rats b

Non-immunogenic Induced Pluripotent Stem Cells, a Promising Way Forward for Allogenic Transplantations for Neurological Disorders.

Neurological disorder is a general term used for diseases affecting the function of the brain and nervous system. Those include a broad range of diseases from developmental disorders (e.g., Autism) over injury related disorders (e.g., stroke and brain tumors) to age related neurodegeneration (e.g., Alzheimer's disease), affecting up to 1 billion people worldwide. For most of those disorders, no curative treatment exists leaving symptomatic treatment as the primary mean of alleviation. Human induced pluripotent stem cells (hiPSC) in combination with animal models have been instrumental to foste

Interleukin-6 is important for regulation of core body temperature during long-term cold exposure in mice.

Interleukin-6 (IL6) is a cytokine important for inducing the fever response during infection and has been reported to uphold core body tempe … ndirect calorimetry at room temperature (20°C), and during the first and last day of cold exposure. Brain stem, hypothalamus and white and brown adipose tissues from the cold-exposed mice were subjected to gene expression analysis. After 6 days in 4°C, the IL6-/- mice exhibited significantly lower body temperature and oxygen consumption compared with Wt mice …

Extended magnetic resonance imaging studies on the effect of classically activated microglia transplantation on white matter regeneration following spinal cord focal injury in adult rats.

Spinal cord injuries are still a serious problem for regenerative medicine. Previous research has demonstrated that activated microglia accu … low-up. The lesions were characterized by means of magnetic resonance imaging (MRI). Neurons in the brain stem and motor cortex were labeled with FluoroGold (FG). A total of 12 weeks after surgery, spinal cords and brains were collected and subjected to histopathological and immunohistochemical examinations. Lesion sizes in the spinal cord were measured and …

Functional genomics reveals an essential and specific role for Stat1 in protection of the central nervous system following herpes simplex virus corneal infection.

Innate immune deficiencies result in a spectrum of severe clinical outcomes following infection. In particular, there is a strong associatio … by death (WT infection of Stat1(-/-) mice). Through the use of functional genomics on the infected brain stems, we determined gene signatures that were representative of the three infection outcomes. We demonstrated a pathological signature in the brain stem of Stat1-deficient mice characterized by upregulation of transcripts encoding chemokine receptors, i …

Therapeutic benefit of intravenous transplantation of mesenchymal stem cells after experimental subarachnoid hemorrhage in rats.

BACKGROUND: Subarachnoid hemorrhage (SAH) usually occurs when an aneurysm ruptures and bleeds into the subarachnoid space. However, no information is available regarding the therapeutic potency of transplanted mesenchymal stem cells (MSCs) for SAH. Therefore, our aim was to investigate whether MSC transplantation therapy may cause stem cell activation and improve neurologic functional recovery after induction of SAH. METHODS: Female rats were divided into 2 groups of SAH plus phosphate-buffered saline (PBS; control) and SAH plus MSCs (experimental). Both control and experimental groups receiv

Effect of serotonin inhibition on glucocorticoid and mineralocorticoid expression in various brain structures.

Many studies have shown the existence of functional interactions between central neurotransmitter systems and the hypothalamo-pituitary adre … ion of their respective messenger ribonucleic acids (mRNA) in the rat hippocampus, hypothalamus and brain stem. The study was performed in bilaterally adrenalectomized animals, in order to avoid potential drug-induced changes in plasma corticosterone levels, which could secondarily regulate MR and GR. Promoter methylation-associated brain-enriched long noncoding RNAs in glioblastoma: a multi-cohort public epigenomic re-analysis.

BACKGROUND: Glioblastoma (GBM) is epigenetically heterogeneous, and previous long noncoding RNA (lncRNA) methylation studies have often begun from tumor-wide screens. We asked whether brain-enriched lncRNAs show promoter methylation-associated suppression in GBM. RESEARCH DESIGN AND METHODS: Brain-enriched lncRNAs were defined using GENCODE v49 and GTEx/UCSC Xena. Primary discovery used TCGA-GBM primary tumors with matched expression and promoter methylation; 52 patients had both data types. Spearman correlations identified methylation-associated candidates (ρ ≤-0.30; false discovery rate [FD

Exploring Human Brain Metabolism via Genome-Scale Metabolic Modeling with Highlights on Multiple Sclerosis.

Cerebral dysfunctions give rise to a wide range of neurological diseases due to the structural and functional complexity of the human brain stemming from the interactive cellular metabolism of its specific cells, including neurons and glial cells. In parallel with advances in isolation and measurement technologies, genome-scale metabolic models (GEMs) have become a powerful tool in the studies of systems biology to provide critical insights into the understanding of sophisticated eukaryotic systems. In this study, brain cell-specific GEMs were reconstructed for neurons, astrocytes, microglia, …

Bradycardia and sinus arrest following saline irrigation of the brain during epilepsy surgery.

Adverse cardiac events during the intraoperative period are life-threatening. The authors report three episodes of severe bradycardia and sinus arrest that occurred in a patient undergoing anterior temporal lobectomy and amygdalo-hippocampectomy for the treatment of epilepsy. The first episode occurred during resection of the amygdala; the other two episodes were observed during subsequent irrigation of the exposed brain structures, most likely the brain stem structures, because of a rent that the surgeon had deliberately made into the basilar cistern for better anatomic appreciation of the st…

Tissue distribution of ryanodine receptor isoforms and alleles determined by reverse transcription polymerase chain reaction.

The tissue distribution of mRNA for ryanodine receptor (ryr) isoforms in various porcine tissues has been determined using the reverse trans … g the parietal, frontal, and temporal lobes of the cerebrum, thalamus/hypothalamus, cerebellum, and brain stem. The normal (Arg615) and mutant (Cys615) ryr1 alleles were expressed in the brains of normal and malignant hyperthermia susceptible pigs, respectively. These results thus demonstrate expression of two ryr isoforms in each type of striated muscle, an …

Multichannel recordings of the human brainstem frequency-following response: scalp topography, source generators, and distinctions from the transient ABR.

Brainstem frequency-following responses (FFRs) probe the neural transcription of speech/music, auditory disorders, and plasticity in subcortical auditory function. Despite clinical and empirical interest, the response's neural basis remains poorly understood. The current study aimed to more fully characterize functional properties of the human FFR (topography, source locations, generation). Speech-evoked FFRs were recorded using a high-density (64 channel) electrode montage. Source dipole modeling and 3-channel Lissajous analysis was used to localize the most likely FFR generators and their or
