BACKGROUND: Poorly differentiated (PDTC) and anaplastic thyroid (ATC) cancer cells are characterized by the acquisition of epigenetic abnormalities, leading to the silencing of both the sodium iodide co-transporter and the Coxsackie adenovirus receptor. As aberrant histone acetylation and DNA methylation represent epigenetic mechanisms involved in neoplastic transformation, our study investigated the anticancer properties of epigenetic modifiers in thyroid carcinoma.
MATERIALS AND METHODS: The cytotoxicity and gene expression modulation of histone deacetylase and DNA methyltransferase inhibit

Glycogen phosphorylase (GP) and cytochrome oxidase (CO) activities were mapped histochemically in the brain of the turtle Trachemys dorbigni. In the telencephalon, both activities occurred in the olfactory bulb, in all cortical areas, in the dorsal ventricular ridge, striatum, primordium hippocampi and olfactory tubercle. In the diencephalon, they were identified in some areas of the hypothalamus, and in rotundus and geniculate nuclei. Both reactions were detected in the oculomotor, trochlear, mesencephalic trigeminal nuclei, the nucleus of the posterior commissure, torus semicircularis, subst

We present a protocol to generate and analyze 3D cancer models composed of tumor cells, cancer-associated fibroblasts (CAFs), and immune cells that mimic the complexity of the pancreatic ductal adenocarcinoma (PDAC) stroma. The protocol includes steps and conditions for establishing 3D co-cultures, embedding for histological analysis, and flow cytometry analysis for the immunophenotyping and characterization of tumor-infiltrating immune cells. These models are valuable for testing strategies and studying the roles of CAFs and immune cells in tumor growth, chemoresistance, and immunosuppression

The B cells within the germinal centres of lymphoid organs undergo affinity maturation of their antigen receptors, a critical event for antibody memory. Follicular dendritic cells within the germinal centres retain immune complexes that select the developing B cells for which they have a higher affinity. We have now identified a subset of CD4+ CD11c+ CD3- dendritic cells in the germinal centres. These are strong antigen-presenting cells for T cells, but do not co-stimulate CD40-activated B cells. These dendritic cells probably stimulate germinal centre T cells and aid the complicated processes

We previously showed that inhibition of the NF-κB signaling pathway in mesenchymal stromal cells (MSCs) (NKI-MSCs) induces quiescence in co-cultured hematopoietic stem cells (HSCs). This led us to investigate whether NKI-MSCs exert similar growth-inhibitory effects on leukemic cells. We found that both NKI-MSCs and their secretome induce cell cycle arrest in KG1a cells, a cell line of acute myeloid leukemia (AML) origin. Surprisingly, the extracellular vesicles (EVs) isolated from NKI-MSCs supported the proliferation of KG1a cells. This is perhaps the first report showing the opposite effects

Dendritic cells (DCs) play a pivotal role in initiating robust T cell responses against tumors, yet effective strategies to enhance DC activation remain limited. This study explores the synergistic effects of Lycium barbarum polysaccharide (LBP) and the tumor cell lysate of endothelial progenitor cells (EPC-TCL) to boost DC activation and anti-tumor immunity. Using flow cytometry, the enzyme-linked immunosorbent assay, western blot analysis, and in vivo tumor models, we demonstrate that the combination treatment significantly enhanced expression of co-stimulatory molecules (CD40, CD80, CD86, a

Breast cancer (BC) exhibits significant heterogeneity and complexity and is leading causes of mortality in women globally. Paclitaxel (PTX) is commonly utilized as the primary medication for BC. However, the resistance of BC to PTX poses a significant challenge in clinical treatment. This study aimed at to explore whether carboxy-terminal domain small phosphatase like 2 (CTDSPL2) affected PTX resistance in BC cells. PTX resistant BC cell lines, including MCF-7/PTX and MDA-MB-231/PTX, were developed by continuously increasing PTX concentration, and we found that CTDSPL2 was upregulated in BC ce

BACKGROUND: Oral lichen planus (OLP) is a chronic immune-mediated inflammatory disease with malignant potential, and its local inflammatory microenvironment is closely related to mucosal-associated invariant T cells (MAIT). Our prior work established OLP-associated fibroblasts (OLP/AFs) in creating the inflammatory milieu, but their interaction with MAIT cells remains unexplored. This study aims to investigate how OLP/AFs interact with MAIT cells.
MATERIALS AND METHODS: Oral mucosal samples from OLP patients and healthy controls were selected for flow cytometry to detect MAIT cells. MR1 expre

Antibody-dependent cellular phagocytosis is a key mechanism by which macrophages engulf antibody-opsonized cancer cells. This protocol describes a time-lapse live-cell imaging method to visualize antibody-dependent macrophage phagocytosis of green fluorescent protein-labeled, CD24-expressing triple-negative breast cancer cells. Macrophages are derived from human peripheral blood mononuclear cells, differentiated with macrophage colony-stimulating factor, and polarized using interferon gamma and lipopolysaccharide to generate a pro-inflammatory phenotype. Cancer cells are opsonized with an anti

Cuproptosis is a type of cell death that depends on the energy production process inside cells, and it is important in cancer treatment. This study aimed to identify cuproptosis-associated genes in triple-negative breast cancer (TNBC) and reveal regulatory pathways. We applied weighted gene co-expression network analysis to screen gene expression data from TNBC patients. Cuproptosis-associated genes were identified at the intersection of the co-expression modules and the cuproptosis gene dataset. G protein-coupled receptor 68 (GPR68) was identified as a cuproptosis-associated gene that is high

Alzheimer's Disease (AD) is marked by the accumulation of pathology, neuronal loss, and gliosis and frequently accompanied by decline in cognition. Understanding brain cell interactions is key to identifying new therapeutic targets to slow its progression. Here, we used systems biology methods to analyze single-nucleus RNA sequencing (snRNASeq) data generated from dorsolateral prefrontal cortex (DLPFC) tissues of 424 participants in the Religious Orders Study or the Rush Memory and Aging Project (ROSMAP). We identified modules of co-regulated genes in seven major cell types and assigned them t

Vitamin C, also known as L-ascorbic acid (AA), functions as a pro-oxidant in cancer at high doses and exerts anticancer effects by generating reactive oxygen species (ROS) and selectively inducing damage to cancer cells. However, AA at low doses promotes cancer cell proliferation. The efficacy of high-dose AA therapy is frequently restricted by inadequate intracellular AA uptake, resulting from low expression of sodium-dependent vitamin C transporter 2 (SVCT2). In this study, we investigated whether valproic acid (VPA), a histone deacetylase inhibitor, could circumvent this constraint by incre

Syphilis, caused by the extracellular bacterium Treponema pallidum ssp. pallidum, is a multi-stage and systemic infection that is lifelong in the absence of treatment. Two host cell types that frequently encounter T. pallidum during infection are endothelial cells and macrophages; treponemes disseminate through the vasculature and cross the blood-brain and placental barriers by traversing endothelial cell barriers, and macrophages are known to be critical for clearance of T. pallidum. Despite the importance of macrophages in treponemal clearance and endothelial cells in treponemal disseminatio
