In-depth analysis of cystic fibrosis cases caused by CFTR gene variation and research on the prediction and simulation of the impact on protein function.

BACKGROUND: Cystic fibrosis (CF) is caused by CFTR gene mutations. Its diagnosis mainly depends on genetic and sweat chloride tests, but the complexity of these mutations challenges diagnosis. METHODS: This paper reports a new case of a Chinese child with cough and wheezing, suspected of having CF. Trio whole-exome sequencing for the pedigree was carried out to detect CFTR gene mutations. Five tools, namely Mutation Taster, PolyPhen-2, SIFT, FATHMM, and PROVEAN, were used to predict the impacts of mutations on protein function. AlphaFold was employed to predict protein structures, and GROMACS

Predicting high-risk clinical missense variants of SMARCB1 in rare neurogenetic disorder schwannomatosis (nerve tumor) through sequence, structure, and molecular dynamics analyses.

The SMARCB1 gene codes for a key element of the SWI/SNF chromatin-modifying complex, which plays a vital role in controlling gene expression by modifying chromatin architecture. Alterations in SMARCB1 have been linked to several neurological disabilities, including schwannomatosis, a condition marked by the formation of numerous benign tumors affecting the nerve sheaths. Present study explore the effects of nonsynonymous single nucleotide polymorphisms (nsSNPs) within the SMARCB1 gene on its protein structure and functionality. We utilized both sequence-based and structure-oriented predictive

Directed evolution of hydrolytic enzymes for enhanced biodegradation of spinal surgery scaffolds.

Enzymes are nature's tiny workers, breaking down complex molecules with precision. Directed evolution a technique that mimics natural selection in the lab allows scientists to enhance enzymes for specific tasks. In this study, we applied directed evolution to improve four key enzymes PHB Depolymerase, Cutinase, Lipase, and Esterase so they could efficiently break down biomaterial-based spinal scaffolds after surgery. Using advanced computational tools like molecular docking and molecular dynamics (MD) simulations, along with high-throughput screening, we developed enzyme variants with superior

Current Mechanobiological Pathways and Therapies Driving Spinal Health.

Spinal health depends on the dynamic interplay between mechanical forces, biochemical signaling, and cellular behavior. This review explores how key molecular pathways, including integrin, yeas-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ), Piezo, and Wingless/Integrated (Wnt) with β-catenin, actively shape the structural and functional integrity of spinal tissues. These signaling mechanisms respond to physical cues and interact with inflammatory mediators such as interleukin-1 beta (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor alpha (TNF-α),

Molecular regulatory networks of microplastics and cadmium mediated hepatotoxicity from NAFLD to tumorigenesis via integrated approaches.

Microplastics (MPs), as emerging environmental pollutants, not only possess inherent toxicity but also adsorb or carry other hazardous substances, such as the carcinogen cadmium (Cd), leading to more complex public health issues. Exposure to MPs and Cd has been shown to induce liver injury, with each pollutant triggering distinct toxicological pathways and pathological processes. In this study, Cd and MPs were selected as "stressors" to simulate the transformation from inflammation to tumorigenesis, including non-alcoholic fatty liver disease (NAFLD), liver cirrhosis, and hepatocellular carcin

Molecular Imaging in Parathyroid Carcinoma Management: A Comprehensive Review.

Parathyroid carcinoma (PC) is an exceedingly rare endocrine malignancy, accounting for less than 1% of all primary hyperparathyroidism (pHPT) cases. It typically presents with pronounced hypercalcemia and markedly elevated parathyroid hormone (PTH) levels. Accurate imaging plays a pivotal role in diagnosis, staging, surgical planning, and long-term surveillance, although differentiating PC from benign disease on imaging remains a significant challenge. A multimodal imaging strategy combining cervical ultrasonography (US) and nuclear medicine techniques provides high sensitivity for lesion dete

Repositioning Triazoles as Phosphodiesterase-4 Inhibitors to Suppress COVID-19 Cytokine Storms and Fungal Co-Infections via Docking and Simulation.

Severe COVID-19 cases are often characterised by a hyperinflammatory cytokine storm, which leads to immune dysregulation and increased mortality. Simultaneously, opportunistic fungal infections such as mucormycosis have been increasingly reported, especially in immunocompromised individuals. Triazole antifungals are widely used to treat such infections, but their potential immunomodulatory effects remain underexplored. This study aimed to investigate the off-target potential of commonly used antifungal triazoles itraconazole, ketoconazole, posaconazole and voriconazole against human phosphodie

In-silico core proteome analysis for chimeric vaccine development against tick-borne tularemia.

Tularemia is an extremely contagious zoonotic illness resulting from infection with the intracellular bacterium Francisella tularensis. It is transmitted primarily via vector bites particularly from ticks, flies, and mosquitoes and is a severe public health threat. Because of its high virulence, low infective dose, aerosol transmissibility, and potential for mass casualties, F. tularensis is also considered a potential biological warfare agent. Despite its severity, there is presently no licensed vaccine against this pathogen. In the present work, a subtractive proteomics pipeline was implemen
