BACKGROUND: Impulse control-related behavioral disorders refer to a group of compulsive behaviors seen in patients with Parkinson's disease (PD), comprising impulse control disorders (ICD), dopamine dysregulation syndrome (DDS), and punding. Over the years, numerous studies have been published on their prevalence, risk factors, neuroimaging, genetics, and management. More evidence now suggests that each disorder involves a distinct neurobiological mechanism. Although our understanding has improved, early recognition and appropriate treatment remain a significant challenge for clinicians, patie

Recent research indicates a strong link between synaptic dysfunction and Parkinson's disease (PD). This study utilizes summary data-based Mendelian randomization (SMR) to explore genetic associations and causal relationships between synaptic genes and the risk of developing PD. This study utilized the GeneCards database to gather synaptic genes. Subsequently, we integrated QTL data related to these genes, encompassing DNA methylation (mQTLs), gene expression (eQTLs), and protein expression (pQTLs). GWAS data on PD were acquired from the GWAS Catalog, with validation datasets from the FinnGen d

INTRODUCTION: Hemophagocytic lymphohistiocytosis (HLH) is a life-threatening hyperinflammatory syndrome caused by excessive cytokine release from activated T cells and macrophages. Primary HLH, or familial HLH (FHL), results from genetic mutations affecting cytotoxic lymphocyte function.
CASE REPORT: We present a case of FHL Type 2 (FHL2) caused by compound heterozygous variants in the PRF1 gene, including one novel missense variant of p.Ala21Val (A21V). A 5-month-old boy presented with persistent fever, pancytopenia, coagulopathy, hepatosplenomegaly, and elevated ferritin, meeting the HLH-20

Genome-wide association studies (GWASs) have identified multiple genetic regions that confer risk for juvenile idiopathic arthritis (JIA). However, identifying the single-nucleotide polymorphisms (SNPs) that drive disease risk has been impeded by the fact that the SNPs used to identify risk loci are in linkage disequilibrium (LD) with hundreds of other SNPs. Since the causal SNPs remain unknown, it is difficult to identify target genes and thus use genetic information to elucidate disease biology and inform patient care. We next used existing genotyping data from 3,939 children with JIA and 14

Cleft lip with or without cleft palate (CL/P) is one of the most common congenital craniofacial anomalies worldwide and presents significant functional, esthetic, and psychosocial challenges. Despite advances in multidisciplinary care and surgical reconstruction, complications such as impaired wound healing, scar formation, and growth disturbances warrant the development of novel regenerative and surgical strategies, which heavily rely on animal models at the pre-clinical stage. For the current narrative review, the literature search was performed by combining cleft phenotype terms with modeli

BACKGROUND: Acute myeloid leukemia (AML) is driven by complex interactions among genetic alterations, metabolic reprogramming, and immune microenvironment dysregulation. Ferroptosis, a form of iron-dependent regulated cell death linking iron metabolism with immune regulation, has been implicated in AML; however, its causal interplay with immune cells remains unclear. Therefore, this study employed a Mendelian randomization (MR) approach to systematically investigate this relationship.
METHODS: We conducted a two-sample MR study to investigate the causal effects of ferroptosis-related genes an

BACKGROUND: Familial hypocalciuric hypercalcemia (FHH) can mimic primary hyperparathyroidism (PHPT), yet commonly used 24-hour urine calcium (UCa) and calcium:creatinine clearance ratio (CCCR) thresholds may not reliably identify patients with FHH before parathyroidectomy.
STUDY DESIGN: We retrospectively reviewed a prospectively maintained single-institution database of adults undergoing initial or reoperative parathyroidectomy for presumed PHPT from January 2012 through December 2023. Patients required concurrent 24-hour urine calcium and creatinine measurements and serum calcium and creati

The 5' untranslated region (5' UTR) of messenger RNAs (mRNAs) plays a central role in regulating protein synthesis initiation, particularly through the Kozak sequence and upstream open reading frames (uORFs). Genetic variants within these regulatory elements could affect translation, altering gene expression and contributing to clinical phenotypes in humans. We developed a computational method called 5ULTRA (5' Untranslated Region Annotation) for analysis of whole-exome sequencing and whole-genome sequencing data to detect, annotate, and prioritize 5' UTR variants with potential translation im

Phages are ubiquitous and diverse, playing a key role in maintaining microbial ecosystem balance. However, their diversity, potential applications, and their interactions with hosts and other phages remain largely unexplored. To address this, we collected 59,652,008 putative viral genomes from our laboratory, 45 public viral datasets, and an integrated public viral genome database (IGN), covering seven habitats. We obtained 741,692 phage genomes with completeness ≥50% (PGD50), and most (93.83%, 695,938/741,692) of these phage genomes were classified into the Caudoviricetes class. We found that
