Effects of ionizing radiation on incisor development of the prenatal mouse.

The effects of 100 rad of X-irradiation of incisor development in CD1 mice were studied. 24 pregnant mice were irradiated on the 12th day post coitum and sacrificed from the 14th through the 20th gestational days. The 191 irradiated fetuses were smaller than those not irradiated, their crania and necks were malformed and their lower extremities were poorly developed. The developing incisors of the irradiated animals were retarded, the pulpal vessels were enlarged and the vessels walls did not maintain their structural integrity. The cells of the future pulp were necrotic, and the basement memb

The big challenge of SARS-CoV-2 latency: testes as reservoir.

In the efforts to explain COVID-19 pathophysiology, studies are being carried out on the correspondence between the expression of SARS-CoV-2 cell receptors and viral sequences. ACE2, CD147 and TMPRSS2 receptors expression could indicate poorly explored potential infection targets. For the genomic analysis of SARS-CoV-2 receptors, using BioGPS information was decided, which is a portal that centralizes genetic annotation resources, in combination with that of The Human Protein Atlas, the largest portal of human transcriptome and proteome data. We also reviewed the most recent articles on the su

Altered endocrine, cytokine signaling and oxidative stress: A plausible reason for differential changes in testicular cells in diet-induced and genetically-inherited - obesity in adult rats.

Obesity is emerging as a potential risk factor for male infertility. It is a multifactorial disorder with primarily genetic and/or environmental factors. Our earlier studies have shown differential effects of genetically inherited-and high fat diet induced-obesity on hormones, fertility and spermatogenesis in adult male rats. In the present study, we assessed the effect of high fat diet induced - and genetically inherited - obesity on the underlying molecular mechanisms affecting spermatogenesis. The expression of hormone receptors, cytokines and markers of oxidative stress as well as cell cyc

TLR5-mediated sensing of gut microbiota is necessary for antibody responses to seasonal influenza vaccination.

Systems biological analysis of immunity to the trivalent inactivated influenza vaccine (TIV) in humans revealed a correlation between early expression of TLR5 and the magnitude of the antibody response. Vaccination of Trl5(-/-) mice resulted in reduced antibody titers and lower frequencies of plasma cells, demonstrating a role for TLR5 in immunity to TIV. This was due to a failure to sense host microbiota. Thus, antibody responses in germ-free or antibiotic-treated mice were impaired, but restored by oral reconstitution with a flagellated, but not aflagellated, strain of E. coli. TLR5-mediated

Novel intrinsic factor Yun maintains female germline stem cell fate through Thickveins.

Germline stem cells (GSCs) are critical for the reproduction of an organism. The self-renewal and differentiation of GSCs must be tightly controlled to avoid uncontrolled stem cell proliferation or premature stem cell differentiation. However, how the self-renewal and differentiation of GSCs are properly controlled is not fully understood. Here, we find that the novel intrinsic factor Yun is required for female GSC maintenance in Drosophila. GSCs undergo precocious differentiation due to de-repression of differentiation factor Bam by defective BMP/Dpp signaling in the absence of yun. Mechanist

Myeloid progenitors in remission bone marrow in patients with malignant blood diseases: clues for slow hematopoietic regeneration after ABMT?

Slow hematopoietic recovery is a well-known feature in patients undergoing autologous bone marrow transplantation (ABMT), and here we demonstrate that compared with patients with other malignant blood diseases, the thrombocytopenia in acute myeloid leukemia (AML) patients stands out, with the median time to recovery of > 50 x 10(9) platelets being 101 days for AML patients and 22-37 days for other patient groups. We have consequently evaluated the content and fate of myeloid progenitor cells (BFU-E, CFU-GM, and CFU-GEMM) in bone marrow preparations from cancer patients in an attempt to uncover

Generation of human induced pluripotent stem cell lines derived from a patient carrying an intragenic deletion in the NFIA gene.

Brain malformations with or without urinary tract defects (BRMUTD) are caused by heterozygous variants in the NFIA gene. BRMUTD is a neurodevelopmental disorder characterized by hypoplasia or absence of the corpus callosum, hydrocephalus or ventriculomegaly, and developmental delay, which may or may not be accompanied by urinary tract defects. Here, we report the successful generation of induced pluripotent stem cells (hiPSCs) from a 3-year-old male BRMUTD patient using Sendai virus-based non-integrating reprogramming technology. This patient-derived cell line harbors an intragenic deletion wi

Nodal histiocytic sarcoma mimicking metastatic carcinoma, a challenging diagnosis - Case report of an extremely rare hematolymphoid neoplasm with recent updates.

Histiocytic sarcoma (HS), a rare malignant neoplasm of hematolymphoid origin, belongs to the World Health Organization (WHO) classification of histiocytic and dendritic cell neoplasms. This rare aggressive malignancy occurs predominantly in adults and accounts for less than 1% of all hematolymphoid neoplasms. It arises from the monocytic/macrophage lineage and shares the histomorphologic and immunohistochemical features of mature tissue histiocytes. Its predominant presentation is extranodal, involving the intestinal tract, skin, and soft tissue, and it rarely presents with lymphadenopathy. Th

Advancements in three-dimensional bioprinting for reproductive medicine: a systematic review.

Reproductive failure due to age, genetics and disease necessitates innovative solutions. While reproductive tissue transplantation has advanced, ongoing research seeks superior approaches. Biomaterials, bioengineering and additive manufacturing, such as three-dimensional (3D) bioprinting, are harnessed to restore reproductive function. 3D bioprinting uses materials, cells and growth factors to mimic natural tissues, proving popular for tissue engineering, notably in complex scaffold creation with cell distribution. The versatility which is brought to reproductive medicine by 3D bioprinting all

Generation of TWO iPSC lines (CRICKi009-A; CRICKi010-A) from patients with type 1 von Hippel-Lindau (VHL) and histopathologically confirmed renal cell carcinoma (RCC).

VHL disease is an inherited and autosomal dominant disorder affecting 1 in 36,0000 individuals worldwide. It is caused by von Hippel-Lindau (VHL) gene mutations and can affect both genders and all ethnic backgrounds (Nordstrom-O'Brien et al., 2009; Maher, 2004). Here, we generated and characterised two iPSC lines derived from patients with histopathologically confirmed clear cell renal cell carcinoma (ccRCC) and VHL Type 1 enrolled in the TRACERx Renal (TRAcking Renal Cell Carcinoma Evolution Through Therapy (Rx)). PBMCs were reprogrammed to pluripotency using a genome non-integrating Sendai v

Infertility revealing intratubular germinal cell neoplasia of the testis: Case report and review of the literature.

INTRODUCTION: Testicular cancer is the most frequent solid tumor in young people. Most germ cell tumors of the testis, with the exception of vitelline tumors and teratomas in children, and spermatocytic seminoma in the elderly, derive from a common precursor. This precursor, initially described as carcinoma in situ (CIS), is now referred to as intratubular Germinal Cell Neoplasia (ITGCN). CASE PRESENTATION: We report the case of a 37-year-old man with intratubular Germinal Cell Neoplasia (ITGCN) on a testis already treated for cryptorchidism in a context of infertility. We proposed active sur

Testis cancer gene D40 expression and its relationship with clinicopathological features in infertile men.

We previously identified a novel human cancer/testis gene, D40, which is dominantly expressed in testicular germ cells, various cancer cell lines and primary human tumors. The expression of D40 mRNA and proteins in various testicular tissues was quantified using the conventional reverse transcription polymerase chain reaction (RT-PCR), real-time quantitative RT-PCR and western blot analysis. The relationship between levels of D40 expression, serum follicle stimulating hormone (FSH) level and Johnsen's score was examined. D40 mRNA expression was observed in the testes of infertile men, except t…

The transforming growth factor-beta superfamily in early spermatogenesis: potential relevance to testicular dysgenesis.

Regulated transforming growth factor-beta (TGFbeta) superfamily signalling is an integral part of normal testicular development and the processes that enable the production of fertile sperm. Through shared utilization of receptors, signal transduction components and inhibitors, many ligands in this family exhibit functional overlaps; this facet of their function is critical to understand because these ligands are often co-expressed and, hence, they may compete with or compensate for one another, depending on the specific cellular context. This review describes particular germ cell maturation s

Surgical management of pineal region tumors.

The authors advocate an aggressive surgical approach to pineal region tumors to provide a definitive histological diagnosis and to facilitate extensive tumor removal. This strategy is based on their surgical experience in 160 operations for pineal region tumors in which operative mortality was 4% with 3% permanent major morbidity. One-third of pineal tumors were benign for which surgery alone was usually curative. A gross total removal was possible in 31 of 107 malignant tumors. The supracerebellar infratentorial approach was preferred in 86% of patients. The tumors displayed considerable hist

Generation of a rhesus macaque harboring a multivalent reporter for assessing gene editing outcomes.

Somatic gene editing therapies offer the potential for lasting treatments for a range of genetic disorders, with a host of therapies at various stages of clinical testing. The development of safe, effective therapies required relevant pre-clinical animal models capable of measuring on-target delivery and detecting off-target editing. Here we present the creation of a transgenic rhesus macaque encoding the Bivalent Enhanced Traffic Light Editing (BETLE) system, a multi-color fluorescent reporter that provides a simple readout for multiple modes of gene editing, including off-target effects. We

The dystonia-associated Torsin1A sustains CLCC1 function in membrane fusion of the nuclear envelope for NPC biogenesis.

DYT1 dystonia is an incurable movement disorder caused by a loss-of-function mutation in Torsin1A, an endoplasmic reticulum (ER)-resident AAA+ ATPase. Here, we use Drosophila and human cells to shed light on Torsins' mode of action. Fly germ cells lacking dTorsin arrest in development with defects in nuclear pore complex (NPC) biogenesis due to impaired nuclear envelope membrane fusion. We identify the conserved membrane protein Chloride Channel CLIC-like protein 1 (CLCC1) as a Torsin1A interaction partner whose absence phenocopies membrane fusion defects caused by Torsin deletion. CLCC1 is en…

Spermatogonial stem cell-based cellular therapy for non-obstructive azoospermia: a narrative review.

Spermatogonial stem cells (SSCs) constitute the foundation of male sperm production and fertility. SSCs are characterised by their capacity for self-renewal and differentiation into spermatozoa throughout a male's reproductive life, thereby transmitting genetic information to subsequent generations. During embryonic development, SSCs are derived from postnatal testicular gonocytes, which originate from long-lived primordial germ cells. A large proportion of men's infertility is detected by sperm analysis. Unusual sperm parameters include sperm concentration, morphology and motility. Male infer…

Cyst-independent oocyte phagocytosis builds the female reproductive reserve in mice.

During ovariogenesis, more than two-thirds of germ cells are sacrificed to improve the quality of the remaining oocytes. However, the detailed mechanisms behind this selection process are not fully understood in mammals. Here, we developed a high-resolution, four-dimensional ovariogenesis imaging system to track the progression of oocyte fate determination in live mouse ovaries. Through this, we identified a cyst-independent oocyte phagocytosis mechanism that plays a key role in determining oocyte survival. We found that oocytes act as individual cells, rather than connected cyst structures, d…
