The aged tumor microenvironment can impair antitumor T cell function, but the molecular regulators that become limiting in this setting remain poorly defined. In a recent issue of Cell, Chen and colleagues harness in vivo CRISPR screening to compare the effects of genetic perturbations in tumor-reactive CD8+ T cells across young and aged hosts. The screen reveals that increased T cell persistence or an effector-like transcriptional state does not necessarily translate into improved tumor control and identifies Dusp5 and Zfp219 as distinct functional regulators. DUSP5 broadly restrains ERK-depe

Despite the known threat of Aeromonas caviae to aquaculture, its effects on mandarin fish (Siniperca chuatsi) were unknown. This study decisively addresses this critical knowledge gap by successfully isolating and identifying a highly pathogenic strain of A. caviae from diseased mandarin fish during a natural outbreak. We systematically profiled its virulence, establishing a lethal dose 50 % (LD50) of 1.25 × 106 CFU/mL, which confirms its high pathogenicity. Further histopathological examinations revealed extensive tissue damage, including severe necrotic lesions in vital organs such as the li

Porcine epidemic diarrhea virus (PEDV) is an enteric alphacoronavirus that causes severe diarrhea and high mortality in neonatal pigs, leading to substantial economic loss in the porcine industry. Previous studies have primarily focused on the spike protein because of its role in viral entry and induction of neutralizing antibody responses. However, accumulating evidence indicates that other viral components also contribute to host immune modulation and pathogenesis. This review summarizes the current knowledge on PEDV structural proteins, with an emphasis on membrane proteins as regulators of

The efficient transmission of SARS-CoV-2 caused the COVID-19 pandemic, which affected millions of people around the globe. Despite extensive efforts, specific therapeutic interventions and preventive measures against COVID-19 and its consequences, such as long COVID, have not yet been identified due to the lack of a comprehensive knowledge of the SARS-CoV-2 biology. Therefore, a deeper understanding of the sophisticated strategies employed by SARS-CoV-2 to bypass the host antiviral defense systems is needed. One of these strategies is the inhibition of the Ras GTPase-activating protein-binding

Enteroviruses (EVs) are a group of highly contagious RNA viruses that can cause a wide range of diseases, from mild infections to severe complications like neurological disorders and myocarditis. This review focuses on the innate immune evasion strategies employed by EVs, highlighting their mechanisms and consequences. EVs evade host immune responses through various tactics, including inhibiting pathogen recognition receptors (PRRs) such as toll-like receptors (TLRs) and RIG-I-like receptors (RLRs), disrupting key signaling pathways like nuclear factor kappa-B (NF-κB) and (JAK)-signal transduc

Host factor-mediated post-translational modification of coronavirus proteins has been demonstrated as an important strategy for regulating viral proliferation. Identification of key host genes involved in this process may provide potential therapeutic targets. In this study, we used the complementary reverse genetic system to determine that UBXN7 promotes SARS-CoV-2 viral double-stranded RNA (dsRNA) production and also promotes the replication of other human coronaviruses. However, UBXN7 does not affect the replication of VSV and RSV, suggesting that it may be a potential pan human coronaviral

HIV-1 is a well-studied example of a pathogen that has evolved an intermediate level of virulence that maximises transmission. For a trait to evolve it must be heritable, and although viral load-a proxy for disease severity-has been shown to be a heritable trait, it is surprising that specific heritable viral factors remain mostly elusive. Rapid within-host evolution is also expected to diminish heritability. We hypothesised that rather than a small number of mutations of large effect determining viral load, the number of slightly deleterious mutations could be key. As a proof of principle, we

PURPOSE: Using molecular techniques, we have previously shown that carnivorous mammals of the family Mustelidae might be common definitive hosts for various protozoan Sarcocystis species. In the present study we aimed to unravel whether Sarcocystis species using ungulates as intermediate hosts and canids or felids as definitive hosts can be found in intestine of mustelids.
METHODS: Small intestine samples of 93 individual mustelids of five different species from Lithuania were examined. Sarcocystis species were identified based on species-specific PCR and subsequent cox1 sequencing.
RESULTS:

BACKGROUND AND AIM: The dysbiosis of gut microbiota has been reported in acute pancreatitis. However, the direction and magnitude between host microbiota and pancreas remains to be established. This study investigated the association between gut microbiota and acute pancreatitis using Mendelian randomization (MR) methods.
METHODS: Summary statistics of gut microbiota abundance and acute pancreatitis were extracted from genome-wide association studies (GWAS). The two-sample bidirectional MR design was employed to assess genetic association between the microbiota and pancreatitis, followed by a

Background: The intestinal microbiota can regulate numerous host functions, including the immune response. Through fermentation, the microbiota produces and releases microbial metabolites such as short-chain fatty acids (SCFAs), which can affect host homeostasis. There is growing evidence that the gut microbiome can have a major impact on cancer. Specific gut microbial composition and metabolites are associated with tumor status in the host. However, their effects on the antitumor response have scarcely been investigated. Natural killer (NK) cells play an important role in antitumor immunity d

Prior studies have defined multiple, but inconsistent, roles for the enigmatic pattern recognition receptor NLRX1 in regulating several cancer-associated biological functions. In this study, we explore the role of NLRX1 in the highly metastatic murine 4T1 mammary tumor model. We describe a functional dichotomy of NLRX1 between two different cellular contexts: expression in healthy host cells versus expression in the 4T1 tumor cells. Using Nlrx1-/- mice engrafted with 4T1 tumors, we demonstrate that NLRX1 functions as a tumor suppressor when expressed in the host cells. Specifically, NLRX1 in h

Sialic acids consist of nine-carbon keto sugars that are commonly found at the terminal end of mucins. This positional feature of sialic acids contributes to host cell interactions but is also exploited by some pathogenic bacteria in evasion of host immune system. Moreover, many commensals and pathogens use sialic acids as an alternative energy source to survive within the mucus-covered host environments, such as the intestine, vagina, and oral cavity. Among the various biological events mediated by sialic acids, this review will focus on the processes necessary for the catabolic utilization o

Parasites negatively affect biological processes within their hosts, which may alter for example health, growth, and reproductive ability. Non-native invasive parasites, in particular, may have large effects on the endemic hosts, given that the hosts lack evolved specific defences against such parasites. The swim bladder nematode Anguillicola crassus, an invasive parasite originating from Asia, is found in the European eel (Anguilla anguilla, L. 1758), since the 1980s. We investigated whether A. crassus affected several indicators related to health of the European eel (spleen- and liver size,

Communicable respiratory viral infections pose both epidemic and pandemic threats and broad-spectrum antiviral strategies could improve preparedness for these events. To discover host antiviral restriction factors that may act as suitable targets for the development of host-directed antiviral therapies, we here conduct a whole-genome CRISPR activation screen with influenza B virus (IBV). A top hit from our screen, beta-1,3-glucuronyltransferase 1 (B3GAT1), effectively blocks IBV infection. Subsequent studies reveal that B3GAT1 activity prevents cell surface sialic acid expression. Due to this

Soil-transmitted helminths cause widespread disease, infecting ~1.5 billion people living within poverty-stricken regions of tropical and subtropical countries. As adult worms inhabit the intestine alongside bacterial communities, we determined whether the bacterial microbiota impacted on host resistance against intestinal helminth infection. We infected germ-free, antibiotic-treated and specific pathogen-free mice, with the intestinal helminth Heligmosomoides polygyrus bakeri. Mice harboured increased parasite numbers in the absence of a bacterial microbiota, despite mounting a robust helmint

Influenza is a serious global health threat that shows varying pathogenicity among different virus strains. Understanding similarities and differences among activated functional pathways in the host responses can help elucidate therapeutic targets responsible for pathogenesis. To compare the types and timing of functional modules activated in host cells by four influenza viruses of varying pathogenicity, we developed a new DYNAmic MOdule (DYNAMO) method that addresses the need to compare functional module utilization over time. This integrative approach overlays whole genome time series expres

Ebola virus (EBOV) is a virulent pathogen, notorious for inducing life-threatening hemorrhagic fever, that has been responsible for several outbreaks in Africa and remains a public health threat. Yet, its pathogenesis is still not completely understood. Although there have been numerous studies on host transcriptional response to EBOV, with an emphasis on the clinical features, the impact of EBOV infection on post-transcriptional regulatory elements, such as microRNAs (miRNAs), remains largely unexplored. MiRNAs are involved in inflammation and immunity and are believed to be important modulat

Influenza is a worldwide plague caused by the influenza virus (IAV) infection, which is initiated by specific recognition with sialic acids on host cell surface. Bovine lactoferrin (bLf) is a sialoglycoprotein belonging to the transferrin family, and it plays an important role in immune regulation. It also shows toxicity against cancer cells and pathogenic microorganisms including bacteria, fungi, and virus. The purpose of this study is to assess the roles of the sialylated glycans on bLf against IAV. To this end, bLf were first treated with sodium periodate to destroy its sialylated glycans.

Fluctuations of the cytosolic calcium ion (Ca2+) concentration regulate a variety of cellular functions in all eukaryotes. Cells express a sophisticated set of mechanisms to balance the cytosolic Ca2+ levels and the signals that elevate Ca2+ in the cytosol are compensated by mechanisms that reduce it. Alterations in Ca2+-dependent homeostatic mechanisms are the cause of many prominent diseases in humans, such as heart failure or neuronal death. The genetic tractability of Toxoplasma gondii and the availability of genetic tools enabled the use of Genetically Encoded Calcium Indicators (GECIs) ex
