Parasite-altered host behavior in the face of a predator: manipulation or not?

Parasitologists have generally accepted the idea that parasite-induced alterations in host behavior increase the chance for parasite survival and transmission or ensure the completion of its life cycle. The aim of the present study was to investigate modifications in the behavior of Taenia crassiceps-infected BALB/c mice in the face of a predator. The experiments showed modifications in the response of infected mice in comparison with uninfected controls on exposure to a predator final host. However, different studies lead us to suggest that the observed modifications are likely to be a second

Comparative infectivity of Fasciola hepatica metacercariae from isolates of the main and secondary reservoir animal host species in the Bolivian Altiplano high human endemic region.

Fascioliasis due to Fasciola hepatica (Linnaeus, 1758) is an endemic disease on the Northern Bolivian Altiplano, where human prevalences and intensities are the highest known, sheep and cattle are the main reservoir hosts, and pigs and donkeys the secondary ones. Investigations were carried out to study the viability of metacercariae experimentally obtained from eggs shed by naturally infected Altiplanic sheep, cattle, pigs and donkeys. A total of 157 Wistar rats were infected with doses of 5, 10, 20 and 150 metacercariae. Metacercariae aged for different number of weeks were used to analyse t

Effect of acute phase proteins, especially alpha 2-macroglobulin, on granuloma formation around Schistosoma mansoni eggs in the rat.

A new model for the study of granuloma formation in the liver is described. Rats received an injection of 20,000 Schistosoma mansoni eggs in the portal vein and granuloma formation was evaluated at 3, 5 and 7 weeks post-injection. Liver collagen was estimated at the same time and serum procollagen III peptide, a marker of collagenesis, weekly. With this model, wherein the number of S. mansoni eggs and the time of injury are standardized, the effect of high levels of acute phase proteins especially alpha 2-macroglobulin on granuloma formation was studied. It appeared that in rats with high leve

Penicillium-mediated toxicity and microbial dysbiosis are associated with fruiting body abortion in artificially cultivated Chinese cordyceps.

Chinese cordyceps consists of fruiting body and sclerotia (larvae part) formed through the parasitism of the insect Hepialidae by the fungus Ophiocordyceps sinensis, but artificial cultivation is hindered by fruiting body abortion, severely impacting yield and quality. The interactions between the fungus, its host insects, and soil are critical for its development. Multi-omics analyses were conducted to compare normally and abnormally developing samples, examining microbial communities and metabolites in fungus-colonized host larvae and the mycosphere soil. Abnormal fruiting body development w

Brain proteomic differences between wild-type and CD44- mice induced by chronic Toxoplasma gondii infection.

Chronic clinical Toxoplasma gondii (T. gondii) infection is the primary disease state that causes severe encephalitis. CD44 is a member of the cell adhesion molecule family and plays an important role in T. gondii infection. However, proteomic changes in CD44 during chronic T. gondii infection have rarely been reported. Thus, an iTRAQ-based proteomic study coupled with 2D-LC-MS/MS analysis was performed to screen CD44-related proteins during chronic T. gondii infection. As a result, a total of 2612 proteins were reliably identified and quantified. Subsequently, 259, 106, and 249 differentially

Localisation to lipid rafts correlates with increased function of the Gal/GalNAc lectin in the human protozoan parasite, Entamoeba histolytica.

Entamoeba histolytica is the causative agent of dysentery and liver abscess and is prevalent in developing countries. Adhesion to the host is critical to infection and is mediated by amoebic surface receptors. One such receptor, the Gal/GalNAc lectin, binds to galactose or N-acetylgalactosamine residues on host components and consists of heavy (Hgl), light (Lgl) and intermediate (Igl) subunits. The mechanism by which the lectin assembles into a functional complex is not known. The parasite also relies on cholesterol-rich domains (lipid rafts) for adhesion. Therefore, it is conceivable that raf

Nanoarchaeum equitans and Ignicoccus hospitalis: new insights into a unique, intimate association of two archaea.

Nanoarchaeum equitans and Ignicoccus hospitalis represent a unique, intimate association of two archaea. Both form a stable coculture which is mandatory for N. equitans but not for the host I. hospitalis. Here, we investigated interactions and mutual influence between these microorganisms. Fermentation studies revealed that during exponential growth only about 25% of I. hospitalis cells are occupied by N. equitans cells (one to three cells). The latter strongly proliferate in the stationary phase of I. hospitalis, until 80 to 90% of the I. hospitalis cells carry around 10 N. equitans cells. Fu

Heterogeneous distribution of avian malaria parasites across mosquito species: disproportionate involvement of Culex restuans in the Midwestern United States.

BACKGROUND: Avian malaria parasites and related haemosporidians are diverse and widespread vector-borne parasites that circulate within complex host-vector networks. Despite their ecological importance, their diversity and circulation within mosquito communities remain poorly understood in temperate North America. The Midwestern United States, located along major North American migratory flyways, supports diverse avian habitats and mosquito assemblages, providing an ideal setting to investigate these interactions. Here, we characterized the occurrence, phylogenetic diversity, and mosquito-line

Organoid-based in vitro system and reporter for the study of Cryptosporidium parvum sexual reproduction.

Many advances have been made recently in our understanding of Cryptosporidium's asexual cycle and sexual differentiation. However, the process of fertilization, which is required for transmission of infectious oocysts, is not well understood. Typical cancer cell-based culture only allows robust exploration of asexual cycle and sexual differentiation of Cryptosporidium. To facilitate exploration of sexual reproduction in Cryptosporidium parvum, we developed an organoid-based culture system that supports Cryptosporidium's full life cycle and a novel fertilization reporter. Organoid-derived monol

Helminth Infections and Diabetes: Mechanisms Accounting for Risk Amelioration.

The global prevalence of type 2 diabetes mellitus (T2D) is increasing rapidly, with an anticipated 600 million cases by 2035. While infectious diseases such as helminth infections have decreased due to improved sanitation and health care, recent research suggests a link between helminth infections and T2D, with helminths such as Schistosoma, Nippostrongylus, Strongyloides, and Heligmosomoides potentially mitigating or slowing down T2D progression in human and animal models. Helminth infections enhance host immunity by promoting interactions between innate and adaptive immune systems. In T2D, t

Comparing infrared spectroscopic methods for the characterization of Plasmodium falciparum-infected human erythrocytes.

Malaria, caused by parasites of the species Plasmodium, is among the major life-threatening diseases to afflict humanity. The infectious cycle of Plasmodium is very complex involving distinct life stages and transitions characterized by cellular and molecular alterations. Therefore, novel single-cell technologies are warranted to extract details pertinent to Plasmodium-host cell interactions and underpinning biological transformations. Herein, we tested two emerging spectroscopic approaches: (a) Optical Photothermal Infrared spectroscopy and (b) Atomic Force Microscopy combined with infrared s

Effects of schistosomes on host anti-viral immune response and the acquisition, virulence, and prevention of viral infections: A systematic review.

Although a growing number of studies suggest interactions between Schistosoma parasites and viral infections, the effects of schistosome infections on the host response to viruses have not been evaluated comprehensively. In this systematic review, we investigated how schistosomes impact incidence, virulence, and prevention of viral infections in humans and animals. We also evaluated immune effects of schistosomes in those coinfected with viruses. We screened 4,730 studies and included 103. Schistosomes may increase susceptibility to some viruses, including HIV and Kaposi's sarcoma-associated h

Flow cytometric enumeration of parasitaemia and haematologic changes in trypanosoma-infected mice.

African trypanosomiasis is a severe parasitic disease affecting both man and livestock. It is crucial to expand our fundamental knowledge of the intimate interactions between trypanosomes and their vertebrate hosts in order to develop new and efficient control strategies. The mouse model of trypanosomiasis is the most popular for research purposes because of all the logistic advantages of using this species. Studies of any aspect of trypanosomiases in the mouse systematically require the quantification of some phenotypic traits which translate its degree of resistance/susceptibility to the dis

Sm22.6 antigen is an inhibitor to human thrombin.

The physiological role of Schistosomiasis mansoni 22.6 antigen (sm22.6 Ag) and its pathogenic effect on the human host has never been reported. Recombinant sm22.6 Ag is a homogenous polymer under non-denaturing/non-reducing conditions, and an inhibitor to human thrombin. Kinetic and Western blot assays show that the recombinant protein interacts with human thrombin and inhibits proteolytic activity of the protease. Tests of whole blood revealed that coagulation time was significantly delayed (3-5 times longer) in the presence of the recombinant protein at a concentration similar to thrombin in

The trypanosomiases.

The trypanosomiases consist of a group of important animal and human diseases caused by parasitic protozoa of the genus Trypanosoma. In sub-Saharan Africa, the final decade of the 20th century witnessed an alarming resurgence in sleeping sickness (human African trypanosomiasis). In South and Central America, Chagas' disease (American trypanosomiasis) remains one of the most prevalent infectious diseases. Arthropod vectors transmit African and American trypanosomiases, and disease containment through insect control programmes is an achievable goal. Chemotherapy is available for both diseases, b

Short report: limited and short-lasting humoral response in Taenia solium: seropositive households compared with patients with neurocysticercosis.

Epidemiologic data suggest that 30-40% of Taenia solium-seropositive people become spontaneously negative without acquiring cysticercosis. To compare the responses of these individuals with those of patients with neurocysticercosis, we screened seropositive persons among family members of 16 patients. We searched for specific antibodies in patients and their 118 households by an enzyme-linked immunoelectrotransfer blot assay using specific glycoproteins of T. solium metacestodes. We found six seropositive individuals without neurocysticercosis among members of four families. The matching patie

Not all infected fish become mushy: phenotypic expression underestimates Unicapsula seriolae infection in yellowtail kingfish (Seriola lalandi).

Yellowtail Kingfish (YTK) (Seriola lalandi) is widely distributed throughout the Indo-Pacific and is a significant recreational and commercial species in Australia. In New South Wales (NSW), a post-cooking mushy flesh condition has been reported, though its cause remains unconfirmed. This study combined morpho-molecular diagnostics with a citizen-science mushy fish reporting platform to identify associated parasites and to map the spatio-temporal distribution of mushy flesh and infections in NSW. Unicapsula seriolae was detected in 38.9% (95% exact CI: 23.1-56.5%) of YTK donated by recreationa

Multi-Epitope Vaccine Design Against Leishmania donovani: An Immunoinformatic Based In Silico Approach.

Leishmaniasis, caused by Leishmania donovani, remains a major neglected tropical disease (NTD) with limited therapeutic options and the absence of a universally effective vaccine. Multi-epitope vaccines offer a promising strategy for combating this intracellular parasite by stimulating a robust and specific immune response. In this study, an immunoinformatics-driven, in silico reverse vaccinology approach was utilized to design a multi-epitope vaccine targeting key surface-exposed proteins of L. donovani , namely C-type lectin, Proteophosphoglycan (PPG4), Hydrophilic Acylated Surface Protein (

Transcriptional Profiling of the Landes Goose Cecum Following Infection with Eimeria stigmosa Isolated from Shanxi Province, North China.

Though a number of Eimeria species have been described in geese, the host responses to these parasites at the molecular level have yet to be explored. In the present study, fresh fecal samples were collected for single-oocyst isolation. The recovered oocysts were identified based on molecular analysis using PCR and sequencing. Subsequently, we examined the transcriptional response of the Landes goose cecum following infection with the isolated Eimeria strain. Molecular analysis showed that the isolated Eimeria strain was Eimeria stigmosa, which was named the E. stigmosa SX-01 strain. Transcrip
