In man and domestic animals, scarring in the skin after trauma, surgery, burn or sports injury is a major medical problem, often resulting in adverse aesthetics, loss of function, restriction of tissue movement and/or growth and adverse psychological effects. Current treatments are empirical, unreliable and unpredictable: there are no prescription drugs for the prevention or treatment of dermal scarring. Skin wounds on early mammalian embryos heal perfectly with no scars whereas wounds to adult mammals scar. We investigated the cellular and molecular differences between scar-free healing in em

Acoustic droplet ejection (ADE) enables nanoliter-scale liquid handling for complex microplate assays, yet translating experimental designs into validated, instrument-ready instructions remains a bottleneck. We present PickliPy, an open-source framework that converts spreadsheet-based assay designs into validated ADE picklists. PickliPy.Assay supports combinatorial, dose-response, and multi-addition time-course dispensing, while PickliPy.Screen extends to high-throughput workflows, including library reformatting and shortlisting. Across biological contexts, the framework generated reproducible

Ranibizumab is a key anti-VEGF therapeutic used to improve treatment efficacy and reduce injection frequency in neovascular retinal diseases. Because experimental pharmacokinetic data in humans are limited, computational modeling provides an effective means to predict ocular drug behavior. In this study, a three-dimensional computational model of the human eye was developed in COMSOL Multiphysics 6.3 to analyze the pharmacokinetics of ranibizumab following intravitreal injection. The model also quantified drug concentration near the macula, the primary target site for neovascular disease, and

Major depressive disorder (MDD) is a prevalent and disabling illness marked by high rates of recurrence, suicide, and self-harm, yet current first-line antidepressants remain inadequate for a substantial proportion of patients. An increasing number of preclinical studies now indicate that β-hydroxybutyrate (BHB) has significant antidepressant potential, and several candidate mechanisms have been proposed, including NLRP3 inflammasome inhibition, HDAC inhibition, GPR109A activation, and enhancement of BDNF-dependent neuroplasticity. However, translation to the clinic remains constrained: eviden

Mice have been used as a valuable model for understanding pathophysiological mechanisms of urinary tract infection for almost six decades. Mice offer many advantages including genetic manipulation to test the role of genes and mechanisms, the availability of germ-free mice, and similarities to humans in innate immune defenses and the strain-dependent presence of vesicoureteral reflux. However, like with humans, the mouse bladder urine above the urinary sphincter has generally been assumed to be sterile. Yet, given the presence of urobiomes in other mammals and the emerging role of the human ur

BACKGROUND AND OBJECTIVES: Riluzole is the only treatment known to improve survival in amyotrophic lateral sclerosis (ALS) patients. However, its efficacy and dosing are limited by hepatic toxicity and interindividual pharmacokinetic variability. Recent experimental studies in hounds have shown that continuous intrathecal (IT) administration of riluzole is well tolerated and achieves significantly higher spinal cord tissue levels. We report the first 2 human ALS cases treated with IT riluzole.
METHODS: A catheter was inserted into the lumbar cistern and advanced to the midcervical region unde

Pre-implantation development arrest poses a significant challenge in infertility treatment cycles. This study aims to evaluate the effect of Insulin, Transferrin, Selenium (ITS), and CHIR99021 on arrested human embryos. Arrested human embryos were obtained from the Embryology Department of the Royan Institute. After determining optimal concentrations, the embryos were assigned to control, CHIR99021, and ITS groups and cultured for 48-72 h. The arrest rate significantly decreased in the ITS and CHIR99021 groups compared to the control group (P < 0.05). The developmental rate up to the pre-morul

Human trophoblast stem (TS) cells are informative in vitro models for the generation and testing of biologically meaningful hypotheses. The goal of this project was to derive patient-specific TS cell lines from clinically available chorionic villus biopsies. Cell outgrowths were captured from human chorionic villus tissue specimens cultured in modified human TS cell medium. Cell colonies emerged early during the culture and cell lines were established and passaged for several generations. Karyotypes of the newly established chorionic villus-derived trophoblast stem (TSCV) cell lines were deter

Understanding human-virus protein-protein interactions is critical for studying molecular mechanisms driving viral infection, immune evasion, and propagation, thereby informing strategies for public health. Here, we introduce a novel multimodal deep learning framework that integrates high-confidence experimental datasets to systematically predict putative interactions between human and viral proteins. Our approach incorporates two complementary tasks: binary classification for interaction prediction and conditional sequence generation to identify interacting protein partners. By leveraging pro

INTRODUCTION: Asthma is a disease characterized by chronic inflammation of the airway mucosa that causes tissue remodeling and a reversible decrease in airflow. The causative agent of asthma is still unknown; however, several studies have shown that environmental factors such as allergens present in pollens are involved. This project's objective was to develop and evaluate a model of respiratory hypersensitivity in Vietnamese minipigs, which is closer in many aspects to humans than rodents, using Phleum pratense allergenic pollen extract.
METHODS: In this hypersensitivity model, human-like si

The usage of plastics in life and industrial applications has led to global environmental pollution by micro- and nanoplastics (MPs/NPs). Despite their widespread occurrence in the environment, little is known about their presence in humans and the potential implications for human health, particularly maternal and fetal health during the prenatal and neonatal periods. Studies on experimental animals indicate that exposure to MPs/NPs can lead to neurological abnormalities in offspring and hemodynamic alterations in the placenta and fetal cerebral arteries. These findings underscore the need for

The etiopathogenesis of type 1 diabetes mellitus (T1DM) is a complex multifactorial process that involves an intricate network of genetic, epigenetic, immunological, and environmental factors. Despite the advances in recent years, some aspects of the mechanisms involved in triggering the disease are still unclear. Infections with certain viruses have been suggested as possible environmental triggers for the autoimmune process that leads to selective and progressive destruction of pancreatic β-cells and insufficiency of insulin production, which is its hallmark. In this review, advances in know

A challenging task for the biological neural signal-based human-exoskeleton interface is to achieve accurate lower limb movement prediction of patients with hemiplegia in rehabilitation training scenarios. The human-exoskeleton interface based on single-modal biological signals such as electroencephalogram (EEG) is currently not mature in predicting movements, due to its unreliability. The multimodal human-exoskeleton interface is a very novel solution to this problem. This kind of interface normally combines the EEG signal with surface electromyography (sEMG) signal. However, their use for th

Many people worldwide suffer from hepatitis C virus (HCV) infection, which is frequently persistent. The lack of efficient vaccines against HCV and the unavailability of or limited compliance with existing antiviral therapies is problematic for health care systems worldwide. Improved small animal models would support further hepacivirus research, including development of vaccines and novel antivirals. The recent discovery of several mammalian hepaciviruses may facilitate such research. In this study, we demonstrated that bank voles (Clethrionomys glareolus) were susceptible to bank vole-associ

PURPOSE: Endocervical mucus changes play a key role in regulating fertility throughout the menstrual cycle and in response to hormonal contraceptives. Non-human primates (NHP) provide the most translational animal model for reproductive tract studies, as they have hormonally-regulated menstrual cycles and mucus changes, similar to women.
EXPERIMENTAL DESIGN: We used TMT labelling and LC-LC/MS to compare the proteins found in the mucus of the rhesus macaque to the mucus of the human endocervix. Data are available via ProteomeXchange with identifier PXD021710.
RESULTS: We found 3048 total prot

Introduction: Duchenne muscular dystrophy (DMD) is an X-linked handicapping disease due to the loss of an essential muscle protein dystrophin. Dystrophin-null animals have been extensively used to study disease mechanisms and to develop experimental therapeutics. Despite decades of research, however, treatment options for DMD remain very limited.Areas covered: High-throughput high-content screening and precision medicine offer exciting new opportunities. Here, the authors review animal models that are suitable for these studies.Expert opinion: Nonmammalian models (worm, fruit fly, and zebrafis

Fragmentation of the Golgi ribbon is a common feature of Parkinson´s disease and other neurodegenerative diseases. This alteration could be the consequence of the anterograde and retrograde transport imbalance, α-synuclein aggregates, and/or cytoskeleton alterations. Most information on this process has been obtained from cellular and animal experimental models, and as such, there is little information available on human tissue. If the information on human tissue was available, it may help to understand the cytopathological mechanisms of this disease. In the present study, we analyzed the morp

The industrialization of the agricultural sector has significantly increased the use of chemicals such as pesticides. Therefore, exposure to them is unavoidable, which makes it necessary to assess their safety for humans at actual exposure doses. This paper aims to determine toxicity of three types of pesticides toward human immune cells (HL-60 and U-937): glyphosate (GLY), deltamethrin (DEL), and chlorothalonil (CHL). Cell viability, membrane integrity, inflammation induction, and antioxidant activity were evaluated to determine differences in cellular response to the tested plant protection

Retinal injury is the most common ocular impairment associated with shaken baby syndrome (SBS), which could lead to vision loss and blindness. However, a woodpecker does not develop retinal hemorrhages or detachment even at a high acceleration of 1,000×g during pecking. To understand the mechanism of retinal injury and its resistance strategy, we put insight into the special ability of the woodpecker to protect the retina against damage under acceleration-deceleration impact. In this study, the structural and mechanical differences on the eyes of the woodpecker and human were analyzed quantita
