Cubosomes as Versatile Multifunctional Lipid-Based Nanocarriers for Drug Delivery: A Comprehensive Review.

Cubosomes (QBS) are self-assembled, nanostructured lipid carriers characterized by a bicontinuous cubic liquid crystalline architecture with a three-dimensional honeycomb-like morphology. Their unique internal structure allows simultaneous encapsulation of hydrophilic, lipophilic, and amphiphilic compounds, enabling sustained, site-specific, and targeted drug delivery through various routes, including oral, ocular, nasal, transdermal, and vaginal administration. Composed primarily of amphiphilic lipids such as Glyceryl Monooleate (GMO) and stabilized with surfactants like Pluronic F-127, QBS e…

Unlocking the potential of lipid-based nanoparticles for intranasal drug delivery in Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by dopaminergic neuronal loss, α-synuclein aggregation, mitochondrial dysfunction, and persistent neuroinflammation. Despite symptomatic advances, the blood-brain barrier (BBB) continues to restrict the delivery of many potentially disease-modifying agents. Intranasal (IN) administration, by exploiting direct olfactory and trigeminal pathways, offers a non-invasive means to bypass the BBB. When combined with lipid-based nanoparticles (LNPs), this route has shown promise in enhancing central nervous system target

VPS13C/PARK23 initiates lipid transfer and membrane remodeling for efficient lysosomal repair.

Perturbations in lysosome integrity are tightly linked to neurological disorders and ageing, but the underlying pathogenic mechanisms are incompletely understood. Using an unbiased proteomic approach, we here identified the bridge-like lipid transport protein VPS13C/PARK23 as a key component of a global early response pathway to lysosome damage. VPS13C readily binds lysosomes under mechanical or osmotic tension in anticipation of membrane lesions. The latter trigger a conformational change in the protein's C-terminus, involving its ATG2C domain acting as sensor of damage-induced lipid packing

Systemic lipid metabolic alterations and the tumor microenvironment: A hidden driver of immune dysfunction.

Emerging evidence suggests that systemic dyslipidemias, traditionally recognized as major risk factors for cardiovascular disease, are critical modulators of immune responses. Meanwhile, cancer immunology research focuses on exploiting the immune system to more effectively target tumor cells by enhancing antitumor immunity and overcoming immune suppression within the tumor microenvironment (TME). The growing evidence connecting dyslipidemia-related diseases to tumor progression highlights lipid dysregulation as a potential driver of immune dysfunction in cancer. Elevated circulating lipids, be…

METTL16 promotes MASLD progression through the regulation of lipid synthesis and immune response.

BACKGROUND: Methyltransferase-like 16 (METTL16) is a recently identified m6A RNA methyltransferase. Although its role in hepatocellular carcinoma has been explored, its function in metabolic dysfunction-associated steatotic liver disease (MASLD) remains elusive. METHODS: Expression profiles of methylation-related genes were analyzed in a cohort comprising 206 MASLD patients and 10 healthy controls. Functional assays were conducted using MASLD cell models and diet-induced mouse models. Bioinformatic analyses were performed to evaluate immune cell infiltration and signaling pathway activation. P

Comprehensive Engineering of Ionizable Lipid Nanoparticles and mRNA Elements for Next-Generation Vaccines.

Messenger RNA (mRNA) vaccines in lipid nanoparticles (LNPs) are effective, yet immunity can wane, and reactogenicity increases with dose. We integrate ionizable lipid design, LNP formulation, and untranslated region (UTR) tuning to improve potency at low doses. From a 96-member biodegradable ionizable lipid library, stereodefined H9T6 emerged as the lead ionizable lipid. Design of experiments (DoE) identified an H9T6:DOPG:cholesterol:PEG composition (40:16:43.5:0.5 mol %) that achieved more than 3.5-fold higher dendritic-cell transfection than SM-102, increased endosomal escape, and localized

The association between percentage body fat and lipid profiles in different phenotypes of PCOS.

OBJECTIVES: Polycystic ovary syndrome (PCOS) is a heterogeneous endocrine disorder linked to metabolic disturbances and increased cardiovascular risk. While the Rotterdam criteria classify PCOS into distinct phenotypes, the impact of percentage body fat (PBF) on lipid profiles across these subtypes remains unclear. This study aims to assess whether the association between PBF and lipid parameters differs among PCOS phenotypes. The research question is: "Does the relationship between PBF and lipid profile vary across PCOS phenotypes defined by the Rotterdam criteria?". MATERIAL AND METHODS: A

Porphyrin-lipid nanotheranostics for multimodal imaging of nodal disease in preclinical oral cancers.

RATIONALE: Cervical lymph node metastases in oral cancer patients are a frequent occurrence and important prognostic factor. Anatomical and molecular imaging modalities can identify neck metastases with varying sensitivity and specificity but perform poorly in clinically negative neck nodes with microscopic disease. Herein we investigate the use of porphyrin-lipid nanotheranostics (PS) for multimodal detection of neck disease in preclinical models of oral cancer. METHODS: Xenograft models of tongue cancer were established in nude rats using MOC2 mouse oral squamous cells. PS nanoparticles wer

Reversible lipid-mediated pH-gating of connexin-46/50 by cryo-EM.

Gap junctions, formed by connexin proteins, establish direct electrical and metabolic coupling between cells, enabling coordinated tissue responses. These channels universally respond to intracellular pH changes, closing under acidic conditions to limit the spread of cytotoxic signals during cellular stress, such as ischemia. Using cryo-electron microscopy (cryo-EM), we uncover insights into the structural mechanism of pH-gating in native lens connexin-46/50 (Cx46/50) gap junctions. Mild acidification drives lipid infiltration into the channel pore, displacing the N-terminal (NT) domain and st

Molecular mechanisms of exercise-induced improvements in Alzheimer's disease: a focus on lipid homeostasis.

Alzheimer's disease (AD) is the most prevalent type of dementia, and its pathophysiological mechanisms involve multiple factors, including genomic factors, metabolomic factors, and environmental factors. Lipid dysregulation occurs both centrally and peripherally in patients with AD, and the severity is closely associated with disease progression. Applied studies based on genome-wide association studies, genomic analyses, lipidomic analyses, mass spectrometry, and machine learning, have identified lipids as a key potential target for early diagnosis and intervention in AD. However, due to the c…

NSDHL as a regulator of lipid peroxidation and ER stress-mediated apoptosis in ovarian cancer.

Elevated expression of NAD(P)-dependent steroid dehydrogenase-like (NSDHL), a key enzyme in cholesterol biosynthesis, is associated with ovarian cancer progression and poor prognosis. However, the mechanisms underlying its tumor-promoting effects remain unclear. Here, we show that high NSDHL expression correlates with adverse clinical outcomes, particularly in aggressive subtypes such as high-grade serous ovarian cancer. NSDHL knockdown in cultured ovarian cancer cells reduced viability, triggered lipid peroxidation, and disrupted cholesterol and redox homeostasis, accompanied by elevated reac

The evolution of lipid nanoparticles: Paving the way for next-generation nucleic acid medicines.

Nucleic acid-based therapies have emerged as promising strategies for the regulation of gene expression and the production of therapeutic antigens or proteins for a series of diseases, including cancers, rare diseases, and infectious diseases. However, their clinical application faces challenges. These include high molecular weight, limited cellular uptake, and susceptibility to enzymatic degradation by nucleases in vivo. Both viral and non-viral delivery vectors have been developed as a means of addressing these limitations, including lipid nanoparticles (LNPs), exosomes, polymers, and inorga

Apolipoprotein E4 in Alzheimer's Disease: Role in Pathology, Lipid Metabolism, and Drug Treatment.

Alzheimer's Disease (AD) is a neurodegenerative disorder characterized by cognitive decline and memory loss. Among the genetic risk factors linked to AD, the Apolipoprotein E4 (ApoE4) remains the strongest. It is well known that carrying the ApoE4 isoform is associated with advanced AD pathology, blood-brain barrier (BBB) disruption, and changes in lipid metabolism. In this review, we provide an overview of the role of centrally and peripherally produced ApoE in AD. After this introduction, we focus on new findings regarding ApoE4's effects on AD pathology and BBB function. We then discuss Apo

Programming Nonlinear Interfacial Mechanics of Synthetic Cells: Lipid Geometry and DNA Nanostructures.

Soft interfaces formed by lipid membranes are fundamental to living cells, synthetic cells, and membrane-based soft materials. However, a quantitative framework linking molecular organization with nonlinear interfacial mechanics remains elusive. Here, we establish an analytical framework that captures the nonlinear elastic response of lipid-membrane-coated synthetic cells under micropipette aspiration. Incorporating both area stretching and curvature bending enables the model to quantitatively reproduce the complete pressure-displacement response within the small-deformation regime. This appro

Pro-resolving lipid mediators in diseases: exploring the molecular basis and clinical implication.

Inflammation resolution is now understood as an active and highly coordinated biological process rather than a passive decline in inflammatory signals. When this program fails, inflammation may become persistent and gradually shift to a chronic pathological state. Such unresolved inflammation is increasingly recognized as a core driver of cardiovascular disease, metabolic disorders, autoimmune pathologies, neurodegeneration, and tumor progression. Although conventional anti-inflammatory drugs can suppress inflammatory mediators, they do not restore immune balance or actively promote resolution
