Aspirin eugenol ester attenuates alcoholic liver injury via the NRF2/SLC7A11/GPX4 pathway-mediated ferroptosis suppression.

BACKGROUND: Alcoholic liver injury (ALI) is a prevalent chronic liver disease worldwide and represents a major public health burden. Previous pharmacological and pharmacodynamic studies have indicated that aspirin eugenol ester (AEE) exerts multiple pharmacological activities, including protective effects against non-alcoholic fatty liver disease. However, whether AEE can ameliorate ALI and the underlying mechanisms remain unclear. PURPOSE: This study sought to explore the protective effects of AEE on ALI and to delineate the associated molecular mechanisms. METHODS: This study investigated

Dioscin attenuates D‑galactose‑induced retinal pigment epithelium senescence and epithelial-mesenchymal transition via the TGF‑β2/PI3K/Akt/β‑catenin pathway.

PURPOSE: This study aimed to evaluate the protective effects of Dioscin in an age-related macular degeneration (AMD) -like model and to explore its potential therapeutic mechanisms. METHODS: The therapeutic potential of Dioscin for AMD was investigated using D-galactose-induced senescence models in ARPE-19 cells (an in vitro preliminary screening model) and C57BL/6J mice using RT-qPCR, western blotting, immunofluorescence, and SA-β-gal staining. Bioinformatic analysis and molecular docking identified TGF-β2 as a computationally predicted potential target of Dioscin in AMD. Key mechanisms invo

Social disconnection integrates genetic and proteomic risks in suicidal ideation and depression.

Suicidal ideation (SI) and major depressive disorder (MDD) are complex psychiatric conditions arising from the interplay of genetic liability, molecular processes, and psychosocial factors. While these dimensions have been extensively studied in isolation, their joint contribution to SI and MDD remains unclear. This study integrates multi-modal data to elucidate these synergistic effects and develop robust models for individual-level risk stratification. Leveraging longitudinal multi-modal data from 13,085 UK Biobank participants, we integrated genomic, proteomic, and social connection profile

3D-QSAR and free energy landscape analysis for predicting novel JAK3 inhibitors in rheumatoid arthritis therapy.

UNLABELLED: JAK3 is a main tyrosine kinase within the JAK kinase family, whose inhibition plays a crucial role in the treatment of rheumatoid arthritis (RA). Multiple selective JAK3 inhibitors with different scaffolds exist. Decernotinib, a pyrrolopyridine-based RA candidate, showed early potential but was discontinued during Phase II. Currently, no selective JAK3 inhibitor is available for rheumatoid arthritis. In this research, an integrated ligand- and structure-based approach is employed to develop novel JAK3 inhibitors. Two 3D-QSAR models were developed based on CoMFA and CoMSIA methods,

Ultrasound targeted fatty acid nitroalkene-containing lipid nanoparticles attenuate inflammation and reverse myocardial fibrosis.

RATIONALE: Myocardial ischemia-reperfusion injury (MIRI) induces oxidative stress and inflammatory signaling that drive fibroblast activation, myocardial fibrosis, and progressive cardiac dysfunction, for which effective targeted therapies remain limited. The electrophilic fatty acid nitroalkene 10-nitro-octadec-9-enoic acid (NO₂-FA) exhibits anti-inflammatory and antifibrotic properties but pharmacological actions have been limited by extents of myocardial delivery. Ultrasound-targeted cavitation (UTC) using lipid-shelled gas-filled nanoparticles (LNPs) enables spatially controlled drug relea

A [68Ga]-labeled PDGFRβ-targeting peptide PET probe for assessing MASH-related fibrosis in diet-induced preclinical models and human liver specimens.

Metabolic dysfunction-associated steatohepatitis (MASH) is characterized by hepatocellular injury, inflammation, and progressive fibrosis, yet current non-invasive tests do not directly assess the cellular drivers of fibrogenesis. Platelet-derived growth factor receptor beta (PDGFRβ) is highly expressed on activated hepatic stellate cells (aHSC), a major effector cell population in MASH. A PDGFRβ-targeted peptide was synthesized and radiolabeled with gallium-68. We investigated whether PDGFRβ-targeted PET imaging could enable non-invasive molecular assessment of MASH severity. Dynamic PET/CT,

Machine Learning for Toxicity Prediction Using Chemical Structures: Pillars for Success in the Real World.

Machine learning (ML) is increasingly valuable for predicting molecular properties and toxicity in drug discovery. However, toxicity-related end points have always been challenging to evaluate experimentally with respect to in vivo translation due to the required resources for human and animal studies; this has impacted data availability in the field. ML can augment or even potentially replace traditional experimental processes depending on the project phase and specific goals of the prediction. For instance, models can be used to select promising compounds for on-target effects or to deselect

The correlation of liquid biopsy genomic data to radiomics in colon, pancreatic, lung and prostatic cancer patients.

INTRODUCTION: With the advances in artificial intelligence (AI) and precision medicine, radiomics has emerged as a promising tool in the field of oncology. Radiogenomics integrates radiomics with genomic data, potentially offering a non-invasive method for identifying biomarkers relevant to cancer therapy. Liquid biopsy (LB) has further revolutionized cancer diagnostics by detecting circulating tumor DNA (ctDNA), enabling real-time molecular profiling. This study explores the integration of radiomics and LB to predict genomic alterations in solid tumors, including lung, colon, pancreatic, and

Baicalin restores innate lymphoid immune imbalance during exacerbation of COPD.

Chronic obstructive pulmonary disease (COPD) is characterized by immune dysregulation, including altered innate lymphoid cell (ILC) immune responses, particularly during exacerbations (ECOPD). Baicalin, a natural compound prevalent in various herbal medicines, has shown promise as a therapeutic candidate in ECOPD. However, its potential and molecular mechanism for addressing ILC immune imbalance during ECOPD remain poorly understood. First, this study conducted a cross-sectional analysis of ILC immune responses in stable COPD patients and those experiencing exacerbations. Then, clinical findin

Tissue Factor Driven Coagulation and Inflammation in Lung Injury: Insights From In Vitro and In Vivo Models.

Lung injury and fibrosis involve a complex interplay between inflammation and coagulation, with tissue factor (TF/CF-III/Thromboplastin/CD142) acting as a crucial mediator. Despite its known role in initiating the extrinsic coagulation cascade, TF's contribution to fibrin deposition in lung injury remains underexplored. This study examines the dysregulation of the extrinsic coagulation cascade and its pathological implications in lung injury, contributing to the progression of pulmonary fibrosis (PF). Utilizing bleomycin (BLM), transforming growth factor-β (TGF-β), and TF models, we systematic

Identification of NET formation and the renoprotective effect of degraded NETs in lupus nephritis.

To explore molecular biomarkers associated with the pathophysiology and therapy of lupus nephritis (LN), we conducted a joint analysis of transcriptomic data from 40 peripheral blood mononuclear cells (PBMCs) (GSE81622) and 21 kidney samples (GSE112943) from the Gene Expression Omnibus database using bioinformatics. A total of 976 and 2,427 differentially expressed genes (DEGs) were identified in PBMCs and renal tissues. Seven and two functional modules closely related to LN were identified. Further enrichment analysis revealed that the neutrophil activation pathway was highly active in both P

DNA methylation profiling to predict recurrence risk in meningioma: development and validation of a nomogram to optimize clinical management.

BACKGROUND: Variability in standard-of-care classifications precludes accurate predictions of early tumor recurrence for individual patients with meningioma, limiting the appropriate selection of patients who would benefit from adjuvant radiotherapy to delay recurrence. We aimed to develop an individualized prediction model of early recurrence risk combining clinical and molecular factors in meningioma. METHODS: DNA methylation profiles of clinically annotated tumor samples across multiple institutions were used to develop a methylome model of 5-year recurrence-free survival (RFS). Subsequent

Acute manganese treatment restores defective autophagic cargo loading in Huntington's disease cell lines.

The molecular etiology linking the pathogenic mutations in the Huntingtin (Htt) gene with Huntington's disease (HD) is unknown. Prior work suggests a role for Htt in neuronal autophagic function and mutant HTT protein disrupts autophagic cargo loading. Reductions in the bioavailability of the essential metal manganese (Mn) are seen in models of HD. Excess cellular Mn impacts autophagic function, but the target and molecular basis of these changes are unknown. Thus, we sought to determine if changes in cellular Mn status impact autophagic processes in a wild-type or mutant Htt-dependent manner.

Key genes and histopathological alterations in hepatic fibrogenesis after segmental cholestasis in weaned rats.

BACKGROUND: The selective bile duct ligation (sBDL) model has been proposed to study fibrosis progression in segmental cholestasis, replicating histopathological features of biliary obstruction in both liver parenchyma with biliary obstruction (BO) and without biliary obstruction (WBO). However, the molecular mechanisms driving fibrogenesis in WBO parenchyma remain unclear. This study aimed to characterize gene expression and histological alterations in fibrogenesis using the sBDL model. METHODS: Liver samples were collected at 1, 4, and 8 weeks post-surgery from both BO and WBO parenchyma, u

Increased confidence of radiomics facilitating pretherapeutic differentiation of BRAF-altered pediatric low-grade glioma.

OBJECTIVES: Currently, the BRAF status of pediatric low-grade glioma (pLGG) patients is determined through a biopsy. We established a nomogram to predict BRAF status non-invasively using clinical and radiomic factors. Additionally, we assessed an advanced thresholding method to provide only high-confidence predictions for the molecular subtype. Finally, we tested whether radiomic features provide additional predictive information for this classification task, beyond that which is embedded in the location of the tumor. METHODS: Random forest (RF) models were trained on radiomic and clinical fe

Distinctive Molecular Risk Factors Between MDS and MDS/AML Defined by ICC.

Myelodysplastic syndromes/neoplasms (MDS) represent a heterogeneous group of clonal hematopoietic stem cell diseases with high risks of acute myeloid leukemia (AML) transformation. To emphasize the characteristics of AML transformation, the International Consensus Classification (ICC) has classified MDS with excess blasts (10%-19%) as MDS/AML. Recently, a clinical-molecular prognostic model International Prognostic Scoring System Molecular (IPSS-M) is developed, which improves the risk stratification of MDS. However, these molecular risk factors were analyzed in a cohort of highly heterogeneou

Molecular Mechanisms and Biomarkers Driving the Transition From Chronic Atrophic Gastritis to Gastric Cancer.

Gastric cancer continues to be a major contributor to global cancer morbidity and mortality. Although the Correa cascade is widely used as a model for gastric carcinogenesis, the molecular mechanisms driving the transition from chronic atrophic gastritis to invasive cancer have not been comprehensively integrated into a unified molecular framework. This review aims to synthesize current knowledge of the molecular events associated with each stage of the Correa cascade and to evaluate the clinical relevance of established and emerging biomarkers for risk stratification, early diagnosis, and per
