A Retrospective Study Evaluating the Efficacy of Fludarabine, Cytarabine, and Granulocyte Colony Stimulating Factor with/without Idarubicin Followed by Second Allogeneic Stem Cell Transplant in the Treatment of Relapsed/Refractory Acute Myeloid Leukemia/Myelodysplastic Syndrome Following First Allogeneic Stem Cell Transplant.

Allogeneic stem cell transplantation (allo-SCT) remains the only curative treatment for most patients with acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS). Unfortunately, a significant number of patients with AML/MDS will remain refractory to or subsequently relapse following allo-SCT. There is no consensus currently regarding the standard of care for relapsed AML/MDS after allo-SCT. Thirty-two patients with relapsed AML/MDS following first allo-SCT were treated with fludarabine, cytosine arabinoside, and granulocyte colony-stimulating factor, with or without idarubicin (FLAG ±

Short-chain polyphosphates induce tau fibrillation and neurotoxicity in human iPSC-derived retinal neurons.

The onset of Alzheimer's Disease and Frontotemporal Dementia is closely associated with the aggregation of tau, a multifunctional protein essential for neuronal stability and function. Given the role of tau aggregation in neurodegeneration, understanding the mechanisms behind its fibril formation is crucial for developing therapeutic interventions to halt or reverse disease progression. However, the structural complexity and diverse aggregation pathways of tau present significant challenges, requiring comprehensive experimental studies. In this research, we demonstrate that short-chain polypho

The transforming role of wharton's jelly mesenchymal stem cell-derived exosomes for diabetic foot ulcer healing: a randomized controlled clinical trial.

BACKGROUND: Diabetic foot ulcers (DFUs), which have high rates of recurrence, amputation, and death, are a significant complication in the therapy of diabetes. Chronic inflammation, vascular dysfunction, and peripheral neuropathy are the results of their etiology, which includes dysregulated glucose homeostasis. These elements contribute to the poor clinical outcomes of DFUs and their complexity. Exosomes, which are natural nanovesicles that promote intercellular communication by transporting functional molecular cargos such as proteins, lipids, and nucleic acids, are being investigated as nov

Macrophage iron overload promotes osteoporosis via Cxcl3- and Cxcl7-mediated paracrine signaling in ovariectomized mice.

BACKGROUND: Iron is an essential element for fundamental biological processes, but excess iron contributes to toxicity and is associated with osteoporosis in iron-overload disorders. Although iron chelators are routinely used to treat hematologic iron-overload conditions, emerging evidence suggests their potential applicability in osteoporosis treatment. METHOD: Iron chelators were administered to ovariectomized (OVX) mice to investigate the relationship between iron overload and osteoporosis. Iron accumulation in macrophages was demonstrated through iron and F4/80 co-staining. We analyzed si

Recent Advances in Enterovirus A71 Infection and Antiviral Agents.

Enterovirus A71 (EV-A71) is one of the major causative agents of hand, foot, and mouth disease (HFMD) that majorly affects children. Most of the time, HFMD is a mild disease but can progress to severe complications, such as meningitis, brain stem encephalitis, acute flaccid paralysis, and even death. HFMD caused by EV-A71 has emerged as an acutely infectious disease of highly pathogenic potential in the Asia-Pacific region. In this review, we introduced the properties and life cycle of EV-A71, and the pathogenesis and the pathophysiology of EV-A71 infection, including tissue tropism and host r

The roles and regulation of TBX3 in development and disease.

TBX3, a member of the ancient and evolutionary conserved T-box transcription factor family, is a critical developmental regulator of several structures including the heart, mammary glands, limbs and lungs. Indeed, mutations in the human TBX3 lead to ulnar mammary syndrome which is characterized by several clinical malformations including hypoplasia of the mammary and apocrine glands, defects of the upper limb, areola, dental structures, heart and genitalia. In contrast, TBX3 has no known function in adult tissues but is frequently overexpressed in a wide range of epithelial and mesenchymal der

Non-polio enterovirus infection and electrophysiological changes in human iPSC-derived neural networks.

BACKGROUND: The non-polio enteroviruses (NPEV) enterovirus D68 (EV-D68) and enterovirus A71 (EV-A71) are highly prevalent and considered pathogens of increasing health concern due to their neurotropic potential. Severe neurological complications of usually mild and self-limiting NPEV infections include meningitis, encephalitis, and acute flaccid paralysis, especially in children and immunocompromised patients. Despite clinical burden, the underlying neuropathogenesis of EV-D68 and EV-A71 remains poorly understood. In particular, the impact of the infection on neural function has not been clear

Managing Erectile Dysfunction After Pelvic Radiotherapy.

INTRODUCTION: Radiotherapy (RT) is an integral component of various multidisciplinary treatment approaches for pelvic malignancies, used both in the radical and adjuvant setting. Despite the improvement of radiotherapy modalities, pelvic radiotherapy often induces permanent erectile dysfunction (ED). However, post-treatment ED receives little medical attention, since patient follow-up focuses mainly on cancer recurrence surveillance. The present review aims to summarize the current treatment modalities against radiation-induced erectile dysfunction (RIED) for patients who have received pelvic

Molecular mechanisms and multi-target therapeutic strategies of diabetic nephropathy: from pathogenesis to precision interventions.

Diabetic nephropathy (DN) has become the primary cause of end-stage renal disease globally, and its epidemiological burden intensifies alongside the surging prevalence of diabetes. The pathogenesis involves complex interactions among metabolic dysregulation, oxidative stress, inflammatory responses, and fibrotic signaling pathways. Hyperglycemia drives renal injury through activation of the RAAS and accumulation of advanced glycation end products (AGEs), while aberrant activation of key signaling pathways such as TGF-β/Smad3 and NF-κB further promotes renal fibrosis. Current clinical diagnosis

Risk factors for bleeding in haemato-oncology patients-a nested case-control study: The BITE study protocol (Bleeding In Thrombocytopenia Explained).

INTRODUCTION: Haemato-oncological patients often receive platelet count driven prophylactic platelet transfusions to prevent bleeding. However, many prophylactically transfused patients still bleed. More knowledge on risk factors for bleeding is therefore needed. This will enable identification of bleeding risk profiles on which future transfusion policy can be optimised. The present BITE study (Bleeding In Thrombocytopenia Explained) aims to identify clinical conditions and biomarkers that are associated with clinically relevant bleeding events. METHODS AND ANALYSIS: A matched case-control s

Evaluating the Role of CXCR3 in Pain Modulation: A Literature Review.

CXCR3 is a well-known receptor involved in immune cell recruitment and inflammation. Pathological inflammation leads to pain stimulation and hence nociception. Therefore, we decided to review the recent research on CXCR3 to identify its precise role in the modulation of pain in a variety of clinical conditions targeting various regions of the body. Studies were selected from PubMed Medline, which relate CXCR3 to the progression of diseases with either bone cancer pain, neuropathic pain, cystitis pain, osteoarthritis and rheumatoid arthritis pain, dental pain, in particular, periodontitis and p

Targeting amyloid clearance in Alzheimer's disease as a therapeutic strategy.

Targeting the amyloid-β (Aβ) peptide cascade has been at the heart of therapeutic developments in Alzheimer's disease (AD) research for more than 25 years, yet no successful drugs have reached the marketplace based on this hypothesis. Nevertheless, the genetic and other evidence remains strong, if not overwhelming, that Aβ is central to the disease process. Most attention has focused on the biosynthesis of Aβ from its precursor protein through the successive actions of the β- and γ-secretases leading to the development of inhibitors of these membrane proteases. However, the levels of Aβ are ma

Ruxolitinib: a new treatment option for myelofibrosis.

Myelofibrosis is a myeloproliferative neoplasm characterized by bone marrow fibrosis and extramedullary hematopoiesis. Evolution of myelofibrosis can lead to life-threatening complications, including transformation to leukemia, thrombotic events, and hemorrhagic episodes. The only curative therapy for myelofibrosis is allogeneic hematopoietic stem cell transplantation. Because this disease manifests primarily in the older population, many patients diagnosed with myelofibrosis are not considered medically fit for such aggressive therapy. Other available medical therapies do not halt disease pro

NCI First International Workshop on the Biology, Prevention, and Treatment of Relapse after Allogeneic Hematopoietic Stem Cell Transplantation: report from the Committee on the Epidemiology and Natural History of Relapse following Allogeneic Cell Transplantation.

Allogeneic hematopoietic stem cell transplantation (alloHSCT) is increasingly being used for treatment of hematologic malignancies, and the immunologic graft-versus-tumor effect (GVT) provides its therapeutic effectiveness. Disease relapse remains a cause of treatment failure in a significant proportion of patients undergoing alloHSCT without improvements over the last 2-3 decades. We summarize here current data and outline future research regarding the epidemiology, risk factors, and outcomes of relapse after alloHSCT. Although some factors (eg, disease status at alloHSCT or graft-versus-host
