The well-known biomaterial Ca-hydroxyapatite (Hap) in its pristine form holds the top ranking position in the field of biomedical research and extensive investigation is continuing across the globe to enhance its competency. Hence, having the intention to introduce superior physiognomies (e.g. cytotoxicity, haemocompatibility, and bioactivity coupled with antimicrobial and antioxidant activity) in Hap, in this research work, we exposed Hap to 200 kGy γ-radiation. As a result, γ-radiated Hap exhibited extreme antimicrobial (more than 98%) and moderate (∼34%) antioxidant properties. On the other…

Patient-derived organoids have revolutionized biomedical research and therapies by "transferring the patient into the Petri dish". In vitro access to human lung organoids representing distal lung tissue, i.e. alveolar organoids, would facilitate research pertaining to a wide range of medical conditions and might open for a future approach to individualized treatment. We propose a protocol to derive a single human lung biopsy towards both alveolar and bronchiolar organoids. By modulating Wnt pathway, we obtained a differential gene expression of the main markers for both subtypes, such as a high…

More than one billion people are affected by neglected tropical diseases (NTDs) and many of these diseases are preventable. While the grouping of these conditions as NTDs has generated vast mapping, mass drug administration and surveillance programmes, there is growing evidence of gaps and weaknesses in purely biomedical approaches, and the need for responses that also recognise the social determinants of health. In order to unpack the social and political determinants of NTDs, it is important to view the problem from a social science perspective. Given this background, the Social Sciences for

Pigs are emerging as important large animal models for biomedical research, and they may represent a source of organs for xenotransplantation. The MHC is pivotal to the function of the immune system in health and disease, and it is particularly important in infection and transplant rejection. Pigs deficient in class I MHC could serve as important reagents to study viral immunity as well as allograft and xenograft rejection. In this study, we report the creation and characterization of class I MHC knockout pigs using the Cas9 nuclease and guide RNAs. Pig fetal fibroblasts were genetically engin…

Through extensive dietary and dental surveys among infants and children living in Hawai'i starting in the late 1920s, medical researchers transformed immigrant and indigenous children's mouths into objects of pathological comparison, establishing sites of alternative empirical and epistemological contact that are endemic to U.S. Pacific empire. These studies resulted in the extension of odontoclasia, a veterinary diagnosis, from dogs to humans. As a dietary antidote, researchers recommended the wider consumption of poi, a starchy Hawaiian staple. Although this appears to be a novel endorsement

Here, we report a toxicity study, conducted on zebrafish larvae, of a series of coumarin and sulfamide compounds that were previously reported as inhibitors of human (h) metalloenzymes, carbonic anhydrases (CAs, EC 4.2.1.1). Due to the high relevance of hCA inhibitors as theragnostic agents, it is of pivotal importance to address safety issues that may arise from the initial in vivo toxicological assessment using zebrafish, a relevant model for biomedical research. None of the reported compounds showed adverse phenotypic effects or tissue damage on developing zebrafish larvae after 5 days of e…

Historically biomedical research has examined genetic influences on mental health but these approaches have been limited, likely due to the broad heritability of brain-related disorders (e.g., 30-90%). Epigenetic modifications, such as DNA methylation, are environmentally sensitive mechanisms that may play a role in the origins and progression of mental illness. Recently, genome-wide disruptions of 5-hydroxymethylcytosine (5hmC) were associated with the development of early and late onset mental illnesses such as autism and Alzheimer's disease, bringing new hope to the field of psychiatry. Her…

Advances in genome-editing technologies and sequencing of animal genomes enable researchers to generate genome-edited (GE) livestock as valuable animal models that benefit biological researches and biomedical and agricultural industries. As birds are an important species in biology and agriculture, their genome editing has gained significant interest and is mainly performed by using a primordial germ cell (PGC)-mediated method because pronuclear injection is not practical in the avian species. In this method, PGCs can be isolated, cultured, genetically edited in vitro, and injected into a reci

Conditions of captivity of primates used in biomedical research may have deleterious effects on the welfare of the animals and consequently on the reliability of the research. We investigated the effects of cage size and cage complexity, two fundamental characteristics of captive conditions, on the behaviour of the common marmoset (Callithrix jacchus jacchus). We found an increase in the general level of activity and significant variation in the frequencies of specific behaviours with an increase in cage size and also with cage complexity. Stereotyped behaviours, which occurred in the small ca…

BACKGROUND: HIV prevention research has been fraught with ethical concerns since its inception. These concerns were highlighted during HIV vaccine research and have been elaborated in microbicide research. A host of unique ethical concerns pervade the microbicide research process from trial design to post-trial microbicide availability. Given the urgency of research and development in the face of the devastating HIV pandemic, these ethical concerns represent an enormous challenge for investigators, sponsors and Research Ethics Committees (RECs) both locally and internationally.
