Recombinant avian leukosis viruses of subgroup J isolated from field infected commercial layer chickens with hemangioma and myeloid leukosis possess an insertion in the E element.

BACKGROUND: Five isolates (JS09GY2, JS09GY3, JS09GY4, JS09GY5, and JS09GY6) of avian leukosis virus subgroup J (ALV-J) were isolated from six infected commercial layer flocks displaying both hemangioma and myeloid leukosis (ML), which shared the same parental line, in China in 2009. RESULTS: All six of the commercial layer chickens examined showed hemangiomas on their body surface or feet. Some developed hemangiomas in their internal organs, causing hepatorrhexis and blood loss. Histopathologically different stages of hemangiomas with ML in the liver, heart, and spleen, were observed. Five vi…

Lymphoid organ size varies among inbred lines 6(3) and 7(2) and their thirteen recombinant congenic strains of chickens with the same major histocompatibility complex.

The objective was to evaluate lymphoid organ size in chickens from a series of 13 recombinant congenic strains (RCS) and their highly inbred parental lines (6(3) and 7(2)). The parental line 6(3) was selected for resistance to tumors induced by Marek's disease virus and avian leukosis viruses, whereas line 7(2) was selected for susceptibility to these tumors. Each RCS on the average contains a random one-eighth of genome from the donor line 7(2). Previous studies have shown that lines 6(3) and 7(2) differ in the size of primary lymphoid organs; i.e., the bursa of Fabricius (BF) and the lobes o…

Mechanisms of T cell hyperreactivity in obese strain (OS) chickens with spontaneous autoimmune thyroiditis: lack in nonspecific suppression is due to a primary adherent cell defect.

The disturbed homeostasis of the immune system in Obese strain (OS) chickens with spontaneous autoimmune thyroiditis consists in a general T cell hyperreactivity (concanavalin A hyperresponsiveness, interleukin 2 (IL 2) hypersecretion), particularly expressed by those lymphocytes which infiltrate the diseased thyroid gland. This abnormality has been attributed to a defective regulation of both IL 2 production and IL 2 function by low m.w. factors, which are present in serum and splenocyte culture supernatants of normal chickens, but deficient in the OS. In the present study we identified the c…

Association of persistent synthesis of viral DNA with macrophage accessory cell dysfunction induced by avian retrovirus myeloblastosis-associated virus of subgroup B inducing osteopetrosis in chickens.

This investigation concentrates on a regenerative anemia and immunosuppression occurring in the absence of osteopetrosis. Polyclonal activation of T-cells was used as an in vitro test system to study immunosuppression induced by the avian myeloblastosis-associated virus of Subgroup B inducing osteopetrosis [MAV-2(O)]. T-cell unresponsiveness in vitro was attributed to a defect in an accessory cell function of the macrophage. Counterflow centrifugation fractionation followed by mixing experiments indicated that the T-cell population from immunosuppressed chickens responded to mitogen stimulatio…

Research note: Improved virus isolation strategies enhance the detection rate for avian leukosis virus eradication in a local chicken breed in China.

The detection rate of avian leukosis virus subgroup K (ALV-K) has been increasing annually in Chinese local breeds, its poor replicative capacity limits the success of virus isolation. To improve ALV-K eradication, this study assessed the detection limits of the magnetic-particle chemiluminescent assay and ELISA kits using a standardized virus stock. Additionally, we also determined the impact of serial passage during virus isolation on the positive detection rate. Using a standardized ALV-K stock, the magnetic-particle chemiluminescent kit detected positive signals at a 256-fold dilution, whe

Multidimensional GWAS analyses on longitudinal phenotypes reveal candidate genes regulating multi-stage egg production traits in Wannan yellow chicken.

Egg production performance directly determines the economic viability of indigenous chicken breeding. However, the genetic regulation of multi-stage egg production traits remains difficult to characterize due to their complex and dynamic nature. Here, we integrated a multidimensional GWAS framework, including single-trait GWAS, multi-trait GWAS (MTAG), and longitudinal trajectory-based GWAS (TrajGWAS), to identify stage-specific and shared genetic effects underlying egg production traits in Wannan yellow chickens (WNY). Whole-genome sequencing of 354 WNY hens (10× depth) and quality control yi…

Nanopore long-read transcriptome sequencing reveals miR-22-3p-mediated transcriptomic remodeling and alternative splicing in chicken pectoral muscle.

The full-length transcriptomic architecture mediated by gga-miR-22-3p in poultry muscle development remains largely unexplored. To address this, a lentiviral knockdown vector targeting miR-22-3p was constructed, and an in vivo miR-22-3p knockdown model was established in the pectoral muscle of 14-day-old Qingyuan partridge chickens through subalar injection. Oxford Nanopore Technologies (ONT) long-read RNA sequencing was employed to profile the full-length transcriptome and its remodeling under miR-22-3p knockdown. In total, 24,727 unannotated transcripts were identified. Differential expressi…

Inhibitory effects of the herbal quorum sensing quencher isoorientin on spoilage characteristics of Serratia liquefaciens S01: In vitro and in a smoked chicken leg model.

The study aimed to screen herbal-derived quorum sensing (QS) quenchers and investigate their inhibitory effects on the Serratia liquefaciens S01, a specific spoilage bacterium in smoked chicken legs. Targeting the QS proteins LuxI and LuxR of S. liquefaciens S01, ten compounds were screened as potential QS quenchers by molecular docking. Among these compounds, isoorientin exhibited a relatively low minimum inhibitory concentration (MIC, 250 μg/mL), potent quorum quenching activity, and the strongest binding affinity. In vitro experiments, sub-MIC concentrations of isoorientin significantly inh

Atomic Force Microscopy-Based Nanomechanical Profiling Reveals Oxidative Stress-Associated Structural Alterations and Curcumin-Associated Protection in Chicken Erythrocytes.

Chicken erythrocytes are nucleated blood cells that provide a useful model for assessing oxidative stress-related membrane injury, but the conventional hemolysis assays do not resolve single-cell nanoscale mechanical alterations. Here, we established an atomic force microscopy (AFM)-based workflow to characterize structural and nanomechanical alterations associated with 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH) exposure and curcumin-associated protection in chicken erythrocytes. Hemolysis assays were used to select a moderate injury condition and a protective curcumin dose. Confocal

The V577S mutation in the S2 subunit of QX-type IBV attenuates viral adaptation to chicken oviduct epithelial cells and pathogenicity in laying hens.

BACKGROUND: The cell and tissue tropism of infectious bronchitis virus (IBV) varies among strains and genotypes, with the S2 subunit of the spike protein serving as a key determinant. METHODS AND RESULTS: In this study, we compared S2 sequences of IBV strains with distinct adaptation to chicken oviduct epithelial cells (COEC) and identified seven potential sites associated with COEC adaptation. Using the highly COEC-adapted GI-19 genotype strain zy30 as a backbone, we generated a series of S2 single-point mutants via reverse genetics.
