[Value of the in situ hybridization technique in the diagnosis of human papilloma virus infections of the uterine cervix. Correlations between the human papilloma virus type and the morphological features].

Clinico-pathologic, epidemiologic and molecular analytic studies have shown that: i) the squamous cancer of the uterine cervix derives from intraepithelial lesions with different degrees of atypia; ii) human papillomavirus (HPV) can play a fundamental role in the progression of these lesions. We have examined 219 uterine cervix biopsies by means of the in situ hybridization technique (ISH) using biotinylated probes for 6/11, 16/18 and 31/35/51 HPV strains. The histologic lesions were as follows: 104 (49.7%) focal koilocytotic lesions 26 (11.8%) low grade intraepithelial lesions (SIL) 45 (20.5%

Genetic cause of a juvenile form of Tay-Sachs disease in a Lebanese child.

Abnormality in the beta-hexosaminidase alpha gene underlying the clinical phenotype of a Lebanese patient with a juvenile form of Tay-Sachs disease has been studied. Clinical features were progressive spasticity, ataxia, and cognitive decline. The protein coding sequence of several beta-hexosaminidase alpha-chain complementary DNAs isolated by polymerase chain reaction was completely normal except for a G-to-A transition at nucleotide position 1511 within exon 13, which resulted in substitution of the normal arginine 504 (CGC) with histidine (CAC). Although the patient was from a first-cousin

Tay-Sachs disease in Moroccan Jews: deletion of a phenylalanine in the alpha-subunit of beta-hexosaminidase.

Tay-Sachs disease is an inherited lysosomal storage disorder caused by defects in the beta-hexosaminidase alpha-subunit gene. The carrier frequency for Tay-Sachs disease is significantly elevated in both the Ashkenazi Jewish and Moroccan Jewish populations but not in other Jewish groups. We have found that the mutations underlying Tay-Sachs disease in Ashkenazi and Moroccan Jews are different. Analysis of a Moroccan Jewish Tay-Sachs patient had revealed an in-frame deletion (delta F) of one of the two adjacent phenylalanine codons that are present at positions 304 and 305 in the alpha-subunit

Sequence analysis of the S RNA of the African arenavirus Mopeia: an unusual secondary structure feature in the intergenic region.

Mopeia virus is an apparently nonpathogenic African arenavirus which can protect animals from subsequent challenge by the closely related Lassa virus. As a step toward understanding these differences in pathogenicity and the means by which Mopeia virus infection can protect against subsequent Lassa virus infection, cDNA clones corresponding to 3419 nucleotides of Mopeia virus S RNA were isolated and sequenced. Two open reading frames, encoding the glycoprotein precursor (GPC) and nucleocapsid (N) proteins, were located in the ambisense arrangement characteristic of the arenaviruses. Comparison

Sequence analysis of the promoter region of the glioblastoma derived T cell suppressor factor/transforming growth factor (TGF)-beta 2 gene reveals striking differences to the TGF-beta 1 and -beta 3 genes.

Human glioblastoma cells secrete a factor termed glioblastoma derived T cell suppressor factor (G-TsF) or transforming growth factor beta 2 (TGF-beta 2) which inhibits the response of T cells to mitogenic or antigenic stimulation. In the present study we isolated the promoter region of the G-TsF/TGF-beta 2 gene. The promoter region shares no homology to the promoter of the TGF-beta 1 or the 5' region of the TGF-beta 3 gene and harbours several familiar DNA motifs, including the cytokine-1 region, an octamer-like sequence, Sp1- and AP-2-like elements and a putative NF-kappa B site. In contrast

Similarities in transthyretin gene expression and differences in transcription factors: liver and yolk sac compared to choroid plexus.

The serum thyroxine-binding protein, transthyretin (TTR), is made by hepatocytes and by choroid plexus epithelium in adults and by yolk sac cells in embryogenesis. Four hepatocyte nuclear factors (HNF-1, -3, and -4 and C/EBP) that are present in liver but not in most other adult tissues bind DNA sites in the TTR gene that are sufficient to direct transgenic expression. Three of these proteins were also found in yolk sac cells, which also express the transgene. A limited transgenic construct is not active in the choroid plexus and a TTR-producing choroid plexus tumor lacks three of the liver-en

Philadelphia chromosome-negative chronic myelogenous leukemia without breakpoint cluster region rearrangement: a chronic myeloid leukemia with a distinct clinical course.

The hallmarks of chronic myelogenous leukemia (CML) include the Philadelphia chromosome (Ph) translocation [t (9;22)(q34;q11)] and consistent molecular genetic aberrations: a break within a restricted 5.8 kb DNA segment, bcr, on chromosome 22q11; transposition of the c-abl protooncogene from chromosome 9q34 to 22q11; and formation of a hybrid bar-abl gene encoding an abnormal 210 Kd bcr-abl protein with augmented tyrosine kinase enzymatic activity. These molecular phenomena may occur even in the absence of cytogenetic evidence of the Ph translocation. They are highly specific and sensitive mar

Two distinct cDNAs for human IMP dehydrogenase.

IMP dehydrogenase (EC 1.1.1.205), the rate-limiting enzyme of de novo GTP biosynthesis, is a promising target in antileukemic chemotherapy. We have isolated two distinct cDNA clones (types I and II) encoding IMP dehydrogenase from a human spleen cDNA library. Both clones encode closely related proteins of 514 residues showing 84% sequence identity. Northern hybridization analyses of poly(A)+ RNA from human normal leukocytes and human ovarian tumors demonstrated a striking contrast in mRNA expression in that type I mRNA is the main species in normal leukocytes and type II predominates over type

Marked decrease of mitochondrial DNA with multiple deletions in a patient with familial mitochondrial myopathy.

Muscle mitochondrial DNA (mtDNA) from a patient with mitochondrial myopathy was examined by Southern blotting. Her family history suggests autosomal dominant inheritance of this disorder. In contrast to other cases of this myopathy that are associated with a compensatory increase of mitochondria, in this patient, the content of mtDNA was markedly decreased (15% of controls), whereas protein of the ATP synthase beta-subunit was not decreased appreciably as judged by Western blotting. In addition, the mitochondrial DNA had multiple deletions, which were located between the replication origins of

Expression of two kallikrein gene family members in the rat prostate.

We have characterized two kallikrein gene family members expressed in the prostate and submaxillary glands of rats. One mRNA (S3) is identical with the previously characterized submaxillary gland S3 mRNA that encodes an enzyme closely related to tonin. The second mRNA (P1) encodes a novel kallikrein-like enzyme that retains key amino acid residues responsible for the characteristic enzymatic cleavage specificity of kallikrein. Two P1-specific oligonucleotide probes derived from the P1 mRNA sequence were used to demonstrate the presence of P1 mRNA in the prostate and submaxillary glands and its

Human oculocutaneous albinism caused by single base insertion in the tyrosinase gene.

Tyrosinase-negative oculocutaneous albinism (OCA) is an inborn error of metabolism, characterized by a complete lack of melanin pigments in the eyes and skin. We have isolated and characterized the tyrosinase gene of one affected child (S.S.) with tyrosinase-negative OCA. Sequence analysis reveals a single-base insertion in the exon 2 that shifts the reading frame and introduces a premature termination signal (TGA codon) after the amino acid residue 298. Functional analysis of the mutated gene indicates that such a truncated tyrosinase lacking one potential copper-binding region is catalytical

Comparison of the primary structure of the 25 kDa ookinete surface antigens of Plasmodium falciparum and Plasmodium gallinaceum reveal six conserved regions.

The gene encoding the 25 kDa ookinete surface antigen (Pgs25) of Plasmodium gallinaceum has been cloned using an oligonucleotide probe directed against one of the EGF-like domains of the P. falciparum 25 kDa ookinete surface antigen (Pfs25). The Pgs25 gene codes for a polypeptide of 215 amino acids, two amino residues less than Pfs25. The deduced amino acid sequence contains a putative signal sequence at the amino-terminus, four tandemly repeated EGF-like domains, and a hydrophobic region at the carboxyl-terminus. By comparing Pgs25 with Pfs25, six conserved regions, consisting of six or more

Selection against lethal alleles in females heterozygous for incontinentia pigmenti.

Studies of five heterozygous females from three kindreds segregating incontinentia pigmenti indicate that cells expressing the mutation have been eliminated from skin fibroblast cultures and in varying degrees from hematopoietic tissues. Clonal analysis was carried out using G6PD variants and methylation patterns at the HPRT locus. Our results confirm X linkage in these families and suggest that selection against cells expressing mutations that are lethal to males in utero may help ameliorate the deleterious phenotype in carrier females.

Development of a triplex FMCA assay for genotyping three genes, ADH1B, ADH1C, and ALDH2, involved in alcohol metabolism.

Three functional single nucleotide variants (SNVs)‒ALDH2 rs671 (p.E504K), ADH1C rs698 (p.I350V), and ADH1B rs1229984 (p.R48H)‒are key genetic determinants of human alcohol metabolism. These variants significantly affect drinking behavior and are associated with liver disease and increased risks of several malignancies, including esophageal and gastric cancers. We developed a triplex fluorescent probe-based melting curve analysis (FMCA) assay for the simultaneous detection of these three SNVs. The assay was validated by comparing FMCA results with Sanger sequencing using genomic DNA from 94 Jap

Challenging the specificity of chlorzoxazone and 4-nitrophenol as marker substrates for CYP2E1 activity in Supersomes™.

Cytochrome P450 2E1 (CYP2E1) plays a pivotal role in the metabolism and detoxification of low molecular-weight xenobiotics, as well as in their bioactivation into toxic and DNA-reactive intermediates. Prominent examples include drugs and chemicals like acetaminophen and N-nitrosamines. Chlorzoxazone and 4-nitrophenol are widely used as probe substrates to assess CYP2E1 activity, however, their specificity remains controversial. This study aimed to reevaluate the selectivity of chlorzoxazone and 4-nitrophenol toward CYP2E1 using recombinant Supersomes™ expressing individual human CYP isoforms.

Development of an Acetaldehyde-Specific Fluorescent Probe: Strategy and Applications.

Acetaldehyde (AA), the principal metabolite of ethanol, is a highly reactive species capable of forming covalent adducts with DNA and proteins, contributing to the pathogenesis and progression of multiple diseases. Conventional methods for detecting endogenous AA are commonly invasive and lack specificity against other aldehydes, hampering in-depth investigation of its in vivo mechanisms and biological functions. To overcome these limitations, we developed a fluorescent probe, TZ11008, for the selective and noninvasive detection of AA in living systems. The sensing strategy employs 2-aminobenz

Streamlined stable CHO cell line development using droplet microfluidics with image-based monoclonality assessment.

Rapid generation of monoclonal CHO cell lines remains a key bottleneck in mammalian cell line development (CLD), often requiring months of iterative screening and cloning. Here, we present an integrated droplet microfluidic workflow for high-throughput screening and isolation of antibody-producing ExpiCHO-S cells using a FRET-based secretion assay. The FRET assay was developed and optimized for picodroplet-based screening by evaluating probe configuration, donor-to-acceptor ratio, antibody concentration range, and signal saturation behavior. The platform enables picodroplet-based detection of

A DNAzyme-CRISPR cascade strategy for preamplification-free detection of Mycobacterium tuberculosis.

Tuberculosis (TB) remains one of the most fatal infectious diseases worldwide, successful treatment is often limited by insufficient diagnostic capabilities. It creates a pressing need for diagnostic methods that combine high sensitivity, specificity, and operational robustness. In this work, we developed a DNAzyme-CRISPR cascade strategy (Dz-CRISPR) for the specific identification of the IS6110 sequence of Mycobacterium tuberculosis (MTB). This system integrated a thermodynamically stabilized hairpin probe, an Arch-shaped signal transduction switch, and an allosterically activated CRISPR-Cas1

Molecular diagnostic tests for isoniazid-resistant tuberculosis: a scoping review.

The paucity of diagnostic tests for isoniazid-resistant tuberculosis is concerning, given its status as the most common form of drug-resistant tuberculosis and a gateway to multidrug-resistant diseases. Molecular drug-susceptibility testing has improved access to timely diagnosis of rifampicin-resistant tuberculosis, but testing for isoniazid-resistant tuberculosis still remains rare. In this Review, we assessed the characteristics of molecular drug-susceptibility testing for detection of isoniazid-resistant tuberculosis, referencing the WHO target product profiles. 9243 citations were screene
