Bioadhesive Nanoparticles in Topical Drug Delivery: Advances, Applications, and Potential for Skin Disorder Treatments.

Skin disorders are the fourth most common cause of all diseases, which affect nearly one-third of the world's population. Topical drug delivery can be effective in treating a range of skin disorders, including microbial infections, skin cancer, dermatitis, burn injury, wounds, and psoriasis. Bioadhesive nanoparticles (BNPs) can serve as an efficient topical drug delivery system as they can serve dual purposes as bioadhesives and nanocarriers, which can mediate targeted drug delivery, prolong retention time, and deepen drug penetration through skin layers. There is an increasing demand for BNP-

Microbiome: Role in Inflammatory Skin Diseases.

As the body's largest organ, the skin harbors a highly diverse microbiota, playing a crucial role in resisting foreign pathogens, nurturing the immune system, and metabolizing natural products. The dysregulation of human skin microbiota is implicated in immune dysregulation and inflammatory responses. This review delineates the microbial alterations and immune dysregulation features in common Inflammatory Skin Diseases (ISDs) such as psoriasis, rosacea, atopic dermatitis(AD), seborrheic dermatitis(SD), diaper dermatitis(DD), and Malassezia folliculitis(MF).The skin microbiota, a complex and ev

HTLV-1-associated infective dermatitis demonstrates low frequency of FOXP3-positive T-regulatory lymphocytes.

BACKGROUND: Human T-lymphotropic virus (HTLV)-1-associated infective dermatitis (ID) is a rare severe chronic eczema, considered as a harbinger for the development of cutaneous adult T-cell leukemia/lymphoma (ATLL) and/or HTLV-1-associated myelopathy (HAM)/tropical spastic paraparesis (TSP). The pathogenesis of ID remains unclear. High numbers of peripheral blood CD4+ CD25+ FoxP3+ regulatory T cells (Tregs) have been reported in ATLL and HAM/TSP. OBJECTIVE: To investigate the status of Tregs, unknown to date, and the histopathological features of ID. METHODS: We studied 16 skin biopsies from

Recurrences of anaphylaxis: trends and risk factors.

PURPOSE OF REVIEW: Recurrences of anaphylaxis is a concern due to its unpredictability and long-term burden to patients and healthcare systems. This review examines recurrence rates, associated risk factors, and gaps in current knowledge to guide improved clinical management. RECENT FINDINGS: From 1240 initial records, 11 studies fulfilled the inclusion criteria of our systematic review. Recurrences of anaphylaxis rates range from 2.6 to 17.6% of patients after a first episode of anaphylaxis, with a rising trend over time. Personal history of food allergy is the most consistent associated ris

Ozone in the Development of Pediatric Asthma and Atopic Disease.

Ozone (O3) is a ubiquitous outdoor air pollutant, which may be derived from various primary pollutants such as nitrates, hydrocarbons, and volatile organ compounds through ultraviolet radiation exposure, and has been shown to negatively impact respiratory health. O3 is the most common noninfectious environmental cause of asthma exacerbations among children and adults. Its effects on pediatric respiratory health could be due to multiple physiologic factors that may contribute to enhanced O3 exposure seen in children compared with adults, including differences in lung surface area per unit of bo

Biologics for allergy: therapeutic potential for ocular allergic diseases and adverse effects on the eye.

Biologics applying antibodies against IgE, IL-5, IL-5 receptor α, IL-4 receptor α, and IL-13 have dramatically improved recent treatment outcomes in allergic diseases including asthma, rhinitis, and atopic dermatitis. However, these drugs have not been approved for ocular allergic diseases such as allergic conjunctivitis, vernal keratoconjunctivitis, and atopic keratoconjunctivitis. Although the putative mechanisms suggest that these drugs should have beneficial effects in patients with ocular allergies and some studies have reported such beneficial effects, various adverse ocular symptoms hav

The Role of Staphylococcus aureus in Patients with Chronic Sinusitis and Nasal Polyposis.

PURPOSE OF REVIEW: Staphylococcus aureus (S. aureus) is correlated with the development of persistent severe inflammatory disease of the upper airway including chronic rhinosinusitis with nasal polyps (CRSwNP). The presence of S. aureus is associated with atopic disease including allergic rhinitis and atopic dermatitis and is associated with poor outcomes. RECENT FINDINGS: Several different strains of S. aureus generate different toxins and gene products that can account for organism pathogenicity. S. aureus bacteria and its antigens shape the bacterial and fungal microbiome and the mucosal n

Shotgun Metagenomics Reveals Skin Microbiome Composition and Function in Infant Atopic Disease.

BACKGROUND: Atopic dermatitis (AD), food sensitization (FS), and food allergy (FA) frequently co-occur in infancy, but the factors underlying distinct atopic phenotypes remain unclear. Although FLG null mutations are major genetic risk factors for AD, they explain only part of disease heritability, suggesting a potential role for the skin microbiome. This study examined how early-life skin microbiome composition and its interaction with host genetics contribute to distinct atopic phenotypes in infancy. METHODS: We analyzed > 1000 skin swabs from 429 infants in the VITALITY cohort using deep s

The role of interleukin-1β in the immune response to contact allergens.

Interleukin-1β (IL-1β) is an important pro-inflammatory cytokine that has an effect on almost every cell lineage in the body. By blocking IL-1β and investigating the IL-1β signaling pathway, several studies have demonstrated a central role of IL-1β in the response to contact allergens. This review summarizes the current literature regarding the basic immunological mechanisms mediated by IL-1β in the different phases of allergic contact dermatitis (ACD) and highlights potential IL-1β-targeted treatment options, which in the future may be relevant in the treatment of patients with ACD. This revi

Successful management of DPP-4 inhibitor-induced bullous pemphigoid with Stapokibart injection in an elderly diabetic patient: a case report.

Dipeptidyl peptidase-4 (DPP-4) inhibitors, such as linagliptin, have been implicated in inducing bullous pemphigoid (BP) in elderly diabetic patients. Stapokibart is a humanized monoclonal antibody targeting interleukin-4 receptor subunit alpha (IL-4Rα), primarily used for atopic dermatitis, with no prior reports of its application in BP. This article describes a case of DPP-4 inhibitor-induced bullous pemphigoid treated with Stapokibart. Following Stapokibart therapy, the patient exhibited complete resolution of skin lesions, significant improvement in quality of life, and no adverse reaction
