Extracellular vesicles-derived microRNAs expression as biomarkers for neurological radiation injury: Risk assessment for space exploration.

Space missions pose threats to the health of the astronauts due to long-term exposure to galactic cosmic rays and solar particle events comprised predominantly of medium to high energy protons, energetic helium ions, and energetic high atomic number particles (HZEs). While the tissue-specific effects of radiation have been studied extensively, the changes in exosomal miRNA expression levels in response to acute radiation exposure have not been assessed. Extracellular vesicles (EVs) originate from the host cells and contain nucleic acid and proteins that can modify the physiology of the receivi

Mechanical Power and Ventilator Variables in Patients With Acute Respiratory Distress Syndrome: A Pilot Prospective Cohort.

BACKGROUND: Mechanical power (MP) integrates multiple ventilatory variables and reflects the total energy transferred to the lungs during mechanical ventilation, potentially contributing to ventilator-induced lung injury (VILI) in acute respiratory distress syndrome (ARDS). This study evaluated the association between MP, ventilatory variables, and mortality in patients with ARDS. METHODS: In this prospective cohort study conducted between May 2025 and May 2026, 50 mechanically ventilated patients with ARDS were enrolled and categorized into survivors (n=25) and non-survivors (n=25). Ventilat…

A Hybrid OPES-eABF Framework for Efficient Exploration and Data-Driven Collective Variable Discovery in Complex Free-Energy Landscapes.

Molecular dynamics (MD) simulations are powerful tools for studying biomolecular systems, but they are fundamentally limited by accessible time scales, making the study of rare events such as protein folding or ligand unbinding computationally challenging. While several enhanced-sampling methods exist to accelerate such processes, their efficacy depends critically on the choice of collective variables (CVs), which are typically intuition-driven and must be defined a priori. To address this, we introduced a unified framework that couples the recently developed OPES-eABF hybrid scheme with a dat

Technical note: Errors introduced when using Dose Voxel Kernels for estimating absorbed dose from radiopharmaceutical therapies involving alpha emitters.

BACKGROUND: In radiopharmaceutical therapies (RPT) involving beta emitters, absorbed dose (Dabs) calculations often employ the use of dose voxel kernels (DVK). Such methods are faster and easier to implement than Monte Carlo (MC) simulations. Using DVK methods implies a non-stochastic distribution of particles. This is a valid assumption for betas where thousands to tens of thousands of particles traversing the cell nucleus are required to achieve cell kill. However, alpha particles have linear energy transfers (LET) that are ∼500 times higher than LETs of betas. This results in a significant …

Novel Multi-Segment Foot Model Incorporating Plantar Aponeurosis for Detailed Kinematic and Kinetic Analyses of the Foot With Application to Gait Studies.

Kinetic multi-segment foot models have been proposed to evaluate the forces and moments generated in the foot during walking based on inverse dynamics calculations. However, these models did not consider the plantar aponeurosis (PA) despite its potential importance in generation of the ground reaction forces and storage and release of mechanical energy. This study aimed to develop a novel multi-segment foot model incorporating the PA to better elucidate foot kinetics. The foot model comprised three segments: the phalanx, forefoot, and hindfoot. The PA was modeled using five linear springs conn

Secondary abnormalities of mitochondrial DNA associated with neurodegeneration.

The central nervous system has a particularly high energy requirement, thus making it very susceptible to defects in mitochondrial function. A number of neurodegenerative diseases, in particular Parkinson's disease (PD), Huntington's disease (HD) and Friedreich's ataxia (FRDA), are associated with mitochondrial dysfunction. The identification of a mitochondrial complex-I defect in PD provides a link between toxin models of the disease, and clues to the pathogenesis of idiopathic PD. We have undertaken genomic transplantation studies involving the transfer of mitochondrial DNA (mtDNA) from PD p

Analyses of Mössbauer mechanical measurements indicate that the cochlea is mechanically active.

Using the results of Mössbauer measurements, mechanical activity in the cochlea was tested for by comparing the measured basilar membrane (BM) transverse velocity amplitude with that calculated for a lossless mechanically passive system, derived from the measured BM velocity phase. If the cochlea is considered to be a lossless mechanically passive system, then the transverse velocity amplitude can be calculated from the group velocity and the relative variation of stiffness along the BM. The group velocity can be derived from the Mössbauer phase measurements, and the relative variation of stif

Optimization of spectral photon-counting CT reconstruction parameters for improved detection of peritoneal metastases compared with energy-integrating detector dual-energy CT: a phantom study.

OBJECTIVE: To determine the best combination of reconstruction parameters on a spectral photon-counting CT (SPCCT) system to improve detection of peritoneal metastases (PMs) on virtual monoenergetic images (VMIs) at 40 keV. MATERIALS AND METHODS: A spectral phantom was scanned using SPCCT and dual-energy CT (EID-DECT) at 8.5 mGy (CTDIvol). 40 keV VMIs were reconstructed on EID-DECT using a level 4 Spectral algorithm and kernel B. For SPCCT, two reconstruction kernels (STD-B and HRB) and four iDose4 levels (i5/i7/i9/i11) were used. Noise power spectrum (NPS) and task-based transfer function (T

Quantum mechanics/molecular mechanics studies on mechanistic photophysics of epigenetic C5-halogenated DNA nucleosides: 2'-deoxy-5-chlorocytidine and 2'-deoxy-5-bromocytidine in aqueous solution.

In this work, we have employed the high-level QM(CASPT2//CASSCF)/MM method to study the photophysical mechanisms of two important metabolized DNA/RNA nucleoside byproducts, i.e., 2'-deoxy-5-chlorocytidine (5CldCyd) and 2'-deoxy-5-bromocytidine (5BrdCyd), in aqueous solution. On the basis of our optimized minimum-energy structures, conical intersections, and crossing points, as well as the computed associated excited-state relaxation pathways involving the different internal conversion (IC) and intersystem crossing (ISC) processes in and between the S1, T1, T2, and S0 states, we have suggested

31P-MRS saturation transfer for assessing human hepatic ATP synthesis at clinical field strength.

BACKGROUND: 31P-magnetic resonance spectroscopy (MRS) saturation transfer (ST) allows for noninvasive investigation of liver energy metabolism by assessing flux rates of adenosine triphosphate (ATP) synthesis. However, this technique has rarely been applied at clinical field strengths because of long examination times and contamination from muscle tissue. Our aim was to establish a new method to robustly assess ATP synthesis using a clinical scanner. METHODS: A prospective single-center study was performed (January 2023-August 2024) within the German Diabetes Study. We established a suitable

Effects of sodium chloride intake on urea-N recycling and renal urea-N kinetics in lactating Holstein cows.

The effects of high (2.5% of DM) versus normal dietary sodium chloride (NaCl) intake on renal urea-N kinetics and urea-N metabolism were investigated in 9 rumen-cannulated and multicatheterized lactating dairy cows in a crossover design with 21-d periods. It was hypothesized that urinary urea-N excretion would be greater, and BUN concentration lower in response to greater diuresis induced by high NaCl intake. Also, urea-N transport across ruminal and portal-drained viscera (PDV) tissues was hypothesized to be affected by dietary sodium intake. A second experiment was conducted using 8 lactatin

Investigation of Interface Characteristics and Physisorption Mechanism in Quantum Dots/TiO2 Composite for Efficient and Sustainable Photoinduced Interfacial Electron Transfer.

Owing to their superior stability compared to those of conventional molecular dyes, as well as their high UV-visible absorption capacity, which can be tuned to cover the majority of the solar spectrum through size adjustment, quantum dot (QD)/TiO2 composites are being actively investigated as photosensitizing components for diverse solar energy conversion systems. However, the conversion efficiencies and durabilities of QD/TiO2-based solar cells and photocatalytic systems are still inferior to those of conventional systems that employ organic/inorganic components as photosensitizers. This is b
