Microbiome Shifts in Peri-Implantitis: Longitudinal Characterization of Dysbiosis and Resolution.

OBJECTIVE: To characterize the longitudinal dynamics of the peri-implant microbiome in patients with peri-implantitis and healthy implants. METHODS: The peri-implant microbiome was characterized longitudinally in patients with peri-implantitis and healthy implants via 16S rRNA gene sequencing. Samples were collected at baseline, 3-, and 6-months post-treatment. Bioinformatic analysis was performed to identify significant microbial shifts over time RESULTS: At baseline, peri-implantitis sites exhibited significantly higher relative abundances of Prevotella (8.44%) and Fusobacterium (16.91%), c

How immunity shapes the long-term dynamics of influenza H3N2.

Since its emergence in 1968, influenza A H3N2 has caused yearly epidemics in temperate regions. While infection confers immunity against antigenically similar strains, new antigenically distinct strains that evade existing immunity regularly emerge ('antigenic drift'). Immunity at the individual level is complex, depending on an individual's lifetime infection history. An individual's first infection with influenza typically elicits the greatest response with subsequent infections eliciting progressively reduced responses ('antigenic seniority'). The combined effect of individual-level immune

Cocaine-induced gene regulation in D1 and D2 neuronal ensembles of the nucleus accumbens.

Cocaine use disorder is characterized by persistent drug-seeking behavior and a high risk of relapse, driven in part by lasting molecular and circuit adaptations in the nucleus accumbens. To explore the transcriptomic changes underlying these alterations, we employed fluorescence-activated nucleus sorting coupled with single-nucleus RNA sequencing to analyze D1 and D2 medium spiny neurons in this brain region of male mice subjected to acute cocaine exposure or to prolonged withdrawal from repeated cocaine exposure without or with an acute cocaine rechallenge. This approach allowed us to precis

Mechanistic Insights into SAM-Dependent Methyltransferases: A Review of Computational Approaches.

Methylation reactions catalyzed by S-adenosylmethionine (SAM)-dependent methyltransferases are essential to numerous biological functions, including gene expression regulation, epigenetic modifications, and biosynthesis of natural products. Dysregulation of these enzymes is associated with diseases, including cancer and neurodevelopmental disorders, making them attractive drug targets. This review explores the contribution of computational methods, particularly quantum chemical calculations and molecular dynamics (MD) simulations, in elucidating the mechanisms of SAM-dependent methyltransferas

Chronic Spinal Cord Injury Increases Spontaneous Intraspinal Neural Transmission and Spike Train Variability.

Spinal cord injury (SCI) often leads to increased spontaneous activity (SpAP) and responsiveness to sensory feedback. This hyperexcitable state contributes to the development and maintenance of SCI-related neuropathic pain (SCI-NP). In general, characterizations of SpAP in the context of SCI-NP focus on the dorsal horns and sensory-processing neurons. As a result, changes in SpAP in the integrative intermediate gray and the motor-dominant ventral horn after SCI are considerably less understood. Thus, it is unclear whether altered firing dynamics in SpAP throughout the dorso-ventral extent of t

Trajectory of intrinsic capacity among community-dwelling older adults in China: The China health and retirement longitudinal study.

BACKGROUNDS: Intrinsic capacity (IC), the sum of individual mental and physical capabilities, as well as living environment and behavior, jointly determine the functional ability of older adults, shifting the focus from disease to function. At the population level, IC in older adults is associated with adverse health outcomes, such as disability, falls, and death. At the individual level, IC changes dynamically. However, studies on the longitudinal IC trajectory and the factors influencing IC deterioration are limited. We aimed to analyze the IC trajectory and explore the risk factors for IC d

Understanding the transmission of bacterial agents of sapronotic diseases using an ecosystem-based approach: A first spatially realistic metacommunity model.

Pathogens such as bacteria, fungi and viruses are important components of soil and aquatic communities, where they can benefit from decaying and living organic matter, and may opportunistically infect human and animal hosts. One-third of human infectious diseases is constituted by sapronotic disease agents that are natural inhabitants of soil or aquatic ecosystems. They are capable of existing and reproducing in the environment outside of the host for extended periods of time. However, as ecological research on sapronosis is infrequent and epidemiological models are even rarer, very little inf

TMS disruption of the lateral prefrontal cortex increases neural activity in the default mode network when naming facial expressions.

Recognizing facial expressions is dependent on multiple brain networks specialized for different cognitive functions. In the current study, participants (N = 20) were scanned using functional magnetic resonance imaging (fMRI), while they performed a covert facial expression naming task. Immediately prior to scanning thetaburst transcranial magnetic stimulation (TMS) was delivered over the right lateral prefrontal cortex (PFC), or the vertex control site. A group whole-brain analysis revealed that TMS induced opposite effects in the neural responses across different brain networks. Stimulation

Intrinsic Dynamic and Static Nature of π···π Interactions in Fused Benzene-Type Helicenes and Dimers, Elucidated with QTAIM Dual Functional Analysis.

The intrinsic dynamic and static nature of the π···π interactions between the phenyl groups in proximity of helicenes 3-12 are elucidated with the quantum theory of atoms-in-molecules dual functional analysis (QTAIM-DFA). The π···π interactions appear in C-∗-C, H-∗-H, and C-∗-H, with the asterisks indicating the existence of bond critical points (BCPs) on the interactions. The interactions of 3-12 are all predicted to have a p-CS/vdW nature (vdW nature of the pure closed-shell interaction), except for 2Cbay-∗-7Cbay of 10, which has a p-CS/t-HBnc nature (typical-HBs with no covalency). Plasma Metabolomic Changes in Children with Cerebral Palsy Exposed to Botulinum Neurotoxin.

The long-term effect of botulinum neurotoxin A (BoNT-A) on children with cerebral palsy (CP) is unclear, and how the dynamic changes of metabolites impact the duration of effect remains unknown. To tackle this, we collected 120 plasma samples from 91 children with spastic CP for analysis, with 30 samples in each time point: prior to injection and 1, 3, and 6 months after injection. A total of 354 metabolites were identified across all the time points, 39 of which exhibited significant changes (with tentative IDs) (p values <0.05, VIP > 1). Principal component analysis and partial least-squares

Dynamic Biomolecular "Mask" Stabilizes Zn Anode.

Zn anode is confronted with serious Zn dendrite growth and water-induced parasitic reactions, which severely hinders the rapid development and practical application of aqueous zinc metal batteries (AZMBs). Herein, inspired by sodium hyaluronate (SH) biomolecules in living organisms featured with the functions of water retention, ion-transport regulation, and film-formation, the SH working as a dynamic and self-adaptive "mask" is proposed to stabilize Zn anode. Benefiting from the abundant functional groups with high hydrophilicity and zincophilicity, SH molecule can constrain active water mole

Repetitive transcranial magnetic stimulation of the cerebellum improves ataxia and cerebello-fronto plasticity in multiple system atrophy: a randomized, double-blind, sham-controlled and TMS-EEG study.

Cerebellar ataxia is the predominant motor feature of multiple system atrophy cerebellar subtype (MSA-C). Although repetitive transcranial magnetic stimulation (TMS) of the cerebellum is growingly applied in MSA, the mechanism is unknown. We examined dynamic connectivity changes of 20 patients with MSA and 25 healthy controls using TMS combined with electroencephalography. Observations that significantly decreased dynamic cerebello-frontal connectivity in patients have inspired attempts to modulate cerebellar connectivity in order to benefit MSA. We further explore the therapeutic potential of

Frontoparietal and salience network alterations in obsessive–compulsive disorder: insights from independent component and sliding time window analyses.

BACKGROUND: Resting-state functional MRI (fMRI) studies commonly report alterations in 3 core networks in obsessive–compulsive disorder (OCD) — the frontoparietal network, the default mode network and the salience network — defined by functionally connected infraslow oscillations in ongoing brain activity. However, most of these studies observed static functional connectivity in the brains of patients with OCD. METHODS: To investigate dynamic functional connectivity alterations and widen the evidence base toward the triple network model in OCD, we performed group-based independent component a

Combination of inductive effect of lipopolysaccharide and in situ mechanical conditioning for forming an autologous vascular graft in vivo.

Autologous vascular grafts have the advantages of better biocompatibility and prognosis. However, previous studies that implanted bare polymer tubes in animals to grow autologous tubular tissues were limited by their poor yield rates and stability. To enhance the yield rate of the tubular tissue, we employed a design with the addition of overlaid autologous whole blood scaffold containing lipopolysaccharides (LPS). Furthermore, we applied in vivo dynamic mechanical stimuli through cyclically inflatable silicone tube to improve the mechanical properties of the harvested tissues. The effectivene

There are two sides to every story: implications of asymmetry on breast support requirements for sports bra manufacturers.

This study aimed to investigate: (1) the prevalence and magnitude of breast movement asymmetry, (2) the interaction between static and dynamic breast asymmetry and (3) the influence of sports bras on breast asymmetry during running. Position data were collected from 167 females whilst treadmill running and then a sub-group of 12 participants in different bra conditions. Breast movement asymmetry existed in 89% of participants, with resultant static breast position asymmetry larger in participants displaying dynamic asymmetry. Asymmetry was most commonly caused (60% to 75%) by greater movement

Does Tenofovir-containing First-line Antiretroviral Therapy Mitigate the Impact of Pretreatment Non-nucleoside Reverse Transcriptase Inhibitor Drug Resistance?

Redox homeostasis is a critical part of physiological processes. Disruption of such homeostasis is associated with various pathological conditions. Tools for studying cellular redox states are important for understanding various biological mechanisms and for the targeted delivery of drugs and/or imaging agents. Along this line, there have been reaction-based fluorescent probes capable of detecting either oxidative stress or hypoxia. However, tools are not available to detect dynamic redox changes in either direction. Toward that end, we describe chemistry that allows for detecting both reducti

Interventional Treatment of Pulmonary Lymphatic Anomalies.

Pulmonary lymphatic diseases have been recognized for many years and have been referred as pulmonary lymphangiectasia, pulmonary lymphangiomatosis, plastic bronchitis, and idiopathic chylothorax or chylopericardium. The lymphatic etiology of these conditions has been determined by detection of cystic lymphatic structures on biopsy or postmortem examination. Development of new imaging techniques such as dynamic contrast-enhanced magnetic resonance lymphangiography has allowed better understanding of pathophysiology of these conditions. Dynamic contrast-enhanced magnetic resonance lymphangiograp

Dynamic modeling of the outlet of a pulsatile pump incorporating a flow-dependent resistance.

Outlet tube models incorporating a linearly flow-dependent resistance are widely used in pulsatile and rotary pump studies. The resistance is made up of a flow-proportional term and a constant term. Previous studies often focused on the steady state properties of the model. In this paper, a dynamic modeling procedure was presented. Model parameters were estimated by an unscented Kalman filter (UKF). The subspace model identification (SMI) algorithm was proposed to initialize the UKF. Model order and structure were also validated by SMI. A mock circulatory loop driven by a pneumatic pulsatile p

Variable ligand- and receptor-binding hot spots in key strains of influenza neuraminidase.

Influenza A continues to be a major public health concern due to its ability to cause epidemic and pandemic disease outbreaks in humans. Computational investigations of structural dynamics of the major influenza glycoproteins, especially the neuraminidase (NA) enzyme, are able to provide key insights beyond what is currently accessible with standard experimental techniques. In particular, all-atom molecular dynamics simulations reveal the varying degrees of flexibility for such enzymes. Here we present an analysis of the relative flexibility of the ligand- and receptor-binding area of three ke
